Abstract The YWP1 gene encoding a specific mycelial cell wall protein of Yarrowia lipolytica has been cloned and expressed in Saccharomyces cerevisiae using different episomal plasmids. Because the plasmids pYAE35BB and pYAE35ES carrying the YWP1 gene (including the 5′ noncoding promoter sequences) failed to express it, the YWP1 gene was cloned under the control of GAL/CYC or ACT S. cerevisiae promoters. A main band with an apparent molecular mass of 70 kDa was detected by immunoblotting in the cell wall fraction of transformants. Ywp1 processing and incorporation to the cell wall were similar in both Y. lipolytica and S. cerevisiae but not in its final localization in the cell wall. In Y. …

Maturation of barley cysteine endopeptidase B (EPB) in Trichoderma reesei was studied with metabolic inhibitors, Western blotting, and immuno microscopy. The inactive 42-kDa recombinant EPB proprotein, first detected in apical cells, was sequentially processed in a time-dependent manner to a secreted polypeptide of 38.5 kDa, and thereafter, to polypeptides of 37.5, 35.5, and 32 kDa exhibiting enzyme activity both in the hyphae and culture medium. The sizes of the different forms of recombinant EPB were in accordance with molecular masses calculated from the deduced amino acid sequence, assuming cleavage at four putative Kex2p sites present in the 42-kDa proprotein. Saccharomyces cerevisiae chronological life span (CLS) is determined by a wide variety of environmental and genetic factors. Nutrient limitation without malnutrition, i.e. dietary restriction, expands CLS through the control of nutrient signaling pathways, of which TOR/Sch9 has proven to be the most relevant, particularly under nitrogen deprivation. In dieser Untersuchung wurde der Einflus einer 3monatigen ACE-Hemmer-Therapie mit Cilazapril (2,5–5 mg) bei 13 Patienten mit chronischer Aortenklappeninsuffizienz spiroergometrisch und echokardiographisch untersucht. Nach 3monatiger Therapie sanken der linksventrikulare enddiastolische Diameter-Index von 3,5 auf 3,1 cm/m2 (p = 0,005), der linksventrikulare endsystolische Diameter-Index von 2,3 auf 2,0 cm/m2 (p = 0,005). METHODS: Infants and children aged 6-35 months and 3-8 years, respectively, were randomized to receive 2 immunizations with a 7.5-µg or 3.75-µg hemagglutinin (HA) dose of a nonadjuvanted whole-virus A/Vietnam(H5N1) vaccine; adolescents aged 9-17 years received a 7.5-µg dose only. The potential anti-inflammatory effect of plant sterols (PS) enriched milk-based fruit beverages (PS, 1 g/100 mL) (MfB) with/without galactooligosaccharides (GOS, 2 g/100 mL) (MfB-G) in an experimental mice model of chronic ulcerative colitis was evaluated. Beverages were orally administered to mice every day by gavage to achieve PS and GOS doses of 35 and 90 mg/kg, respectively, and experimental colitis was induced by giving mice drinking water ad libitum containing 2% (w/v) dextran sulphate sodium (DSS) for 7 days, alternating with periods without DSS up to the end of the study (56 days). Abstract We investigated the behavior of spectral holes under pressure at various frequencies within the inhomogeneous band for two proteins, namely myoglobin and horseradish peroxidase. In order to achieve narrow bandwidth hole burning, the heme chromophore was replaced by protoporphyrin IX and mesoporphyrin IX, respectively. In myoglobin, we found that the pressure induced shift of the holes varied in a strongly non-linear fashion, when the burn-frequency was tuned across the absorption band. In horseradish peroxidase the pressure shift was linear with burn-frequency but changed in a dramatic fashion upon complex formation with a substrate molecule. These observations are interpreted with…
