Six years of progress in the oral biopharmaceutics area - A summary from the IMI OrBiTo project.

OrBiTo was a precompetitive collaboration focused on the development of the next generation of Oral Biopharmaceutics Tools. The consortium included world leading scientists from nine universities, one regulatory agency, one non-profit research organisation, three small/medium sized specialist technology companies together with thirteen pharmaceutical companies. The goal of the OrBiTo project was to deliver a framework for rational application of predictive biopharmaceutics tools for oral drug delivery. In this work we have explored the nature of macroradicals formed upon radiolysis of aqueous poly(N-vinylpyrrolidone) (PVP) solutions using pulse radiolysis, density functional theory (DFT) and literature data. On the basis of literature data on site-specific kinetics of hydrogen abstraction from simple amides and spectra corresponding to specific radical sites on the same amides we have assessed the distribution of H-atom abstraction by `OH radicals from different positions on the pyrrolidone ring and the polymer backbone. In order to explore the influence of polymer microstructure and stereochemistry in biological settings, the synthesis, micellization, cellular fate and the use in paclitaxel formulations of poly(N-(2-hydroxypropyl)-methacrylamide)-block-poly(L-lactide) (P(HPMA)-block-P(LLA)) and poly(N-(2-hydroxypropyl)-methacrylamide)-block-poly(DL-lactide) block copolymers (P(HPMA)-block-P(DLLA)) were studied. Insulin pumps and glucose sensors are effective in improving diabetes therapy and reducing acute complications. The combination of both devices using an algorithm-driven interoperable controller makes automated insulin delivery (AID) systems possible. Many AID systems have been tested in clinical trials and have proven safety and effectiveness. However, currently, none of these systems are available for routine use in children younger than 6 years in Europe. Objetivos: Determinar la fiabilidad y validez de contenido y concepto de la Mackey Childbirth Satisfaction Rating Scale, escala para medir la satisfacción de las mujeres con el parto y el nacimiento. Métodos: Se utilizó el método de traducción-retrotraducción para obtener una versión adaptada de la escala. Para la validación se seleccionó una muestra de 325 mujeres que habían tenido un hijo sano mediante parto vaginal. La recogida de datos se realizó con un cuestionario autocumplimentado que incluía la escala adaptada, expectativas ante el parto y variables obstétricas y sociodemográficas.
