Using isolated chloroplasts or purified thylakoids from photoautotrophically grown cells of the chromophytic alga Pleurochloris meiringensis (Xanthophyceae) we were able to demonstrate a membrane bound NAD(P)H dehydrogenase activity. NAD(P)H oxidation was detectable with menadione, coenzyme Q0, decylplastoquinone and decylubiquinone as acceptors in an in vitro assay. K m-values for both pyridine nucleotides were in the μmolar range (K m[NADH]=9.8 μM, K m[NADPH]=3.2 μM calculated according to Lineweaver-Burk). Enzymatic methods are sensitive and can be quite specific but they are subject to interference by inhibitors and not all are stoichiometric. The present work proposes some modifications of the Heath & Tappel (Anal Biochem 1976; 7:184—91) enzymatic method of determination of lipid hydroperoxides in order to standardize and automate it and to meet the analytical criteria required for a biological assay. Several studies suggest that xanthine dehydrogenase (XDH) and its oxidase form (XO) play an important role in various types of ischemic and vascular injuries. Recently, we have demonstrated that estradiol (E2) induces a significant decrease of the expression and activity of XDH and of its conversion to XO in human mammary epithelial cells. E2 is known to induce upregulation of eNOS gene expression in aortic endothelial cells. The cellular messenger nitric oxide (NO) has many functions in plants. In this study, we investigated its interplays with Reactive Oxygen Species (ROS) in the defense responses triggered by the elicitin cryptogein produced by the oomycete Phytophthora cryptogea. The production of NO induced by cryptogein in tobacco cell suspensions was partly regulated through a ROS-dependent pathway involving the NADPH oxidase NtRBOHD. In turn, NO down-regulated the level of H2O2 derived from NtRBOHD activity. This work describes the alkane degradation system of the n -alkane degrader actinobacterium Gordonia sp. strain SoCg, which is able to grow on n -alkanes from dodecane (C 12 ) to hexatriacontane (C 36 ) as the sole C source. Šī darba mērķis bija novērtēt redoks kofaktora NADPH inženierijas mēģinājuma ietekmi uz rekombinantu Saccharomyces cerevisiae celmu fenotipu eksponenciālās augšanas laikā. Šajos CEN.PK2-1C rekombinantajos celmos tika ekspresēta NADP+ atkarīga glicerīna dehidrogenāze (Gld2p) ar mērķi atgriezt daļu glicerīna glikolīzē, kombinācijā ar glutationa antioksidatīvās sistēmas un NADPH kofaktora reģenerāciju.
