Moving Towards Precision Urologic Oncology: Targeting Enzalutamide-resistant Prostate Cancer and Mutated Forms of the Androgen Receptor Using the Nov…
2017
Abstract Darolutamide (ODM-201) is a novel androgen receptor (AR) antagonist with a chemical structure distinctly different from currently approved AR antagonists that targets both wild-type and mutated ligand binding domain variants to inhibit AR nuclear translocation. Here, we evaluate the activity of darolutamide in enzalutamide-resistant castration resistant prostate cancer (CRPC) as well as in AR mutants detected in patients after treatment with enzalutamide, abiraterone, or bicalutamide. Darolutamide significantly inhibited cell growth and AR transcriptional activity in enzalutamide-resistant MR49F cells in vitro, and led to decreased tumor volume and serum prostate-specific antigen l…
Prostaglandin D2 regulates joint inflammation and destruction in murine collagen-induced arthritis.
2012
Item does not contain fulltext OBJECTIVE: Prostaglandin D2 (PGD2) may exert proinflammatory or antiinflammatory effects in different biologic systems. Although this prostanoid and the enzymes responsible for its synthesis are up-regulated by interleukin-1beta (IL-1beta) in human chondrocytes in vitro, the role of PGD2 in arthritis remains unclear. This study was undertaken to investigate the role of PGD2 in the inflammatory response and in joint destruction during the development of collagen-induced arthritis (CIA) in mice. METHODS: PGD2 and cytokine levels in mice with CIA were determined by enzyme-linked immunosorbent assay. Expression of hematopoietic PGD synthase (h-PGDS), lipocalin-typ…
Interpretācija kā mākslas darba nosacījums: Hansa Georga Gadamera un Artūra Danto teoriju salīdzinājums
2021
Bakalaura darba mērķis ir pierādīt, ka atšķirības Artūra Kolmena Danto un Hansa Georga Gadamera mākslas teorijās izriet no atšķirīga interpretācijas jēdziena traktējuma. Pirmajās divās nodaļās uzrādīšu to, kā interpretācija saistās ar mākslu katras teorijas ietvaros, bet trešajā nodaļā no interpretācijas jēdziena skatupunkta salīdzināšu līdzīgos un atšķirīgos aspektus abās pieejās. Kā galvenos saskares punktus starp abiem domātājiem identificēju: (1) Georga Vilhelma Frīdriha Hēgeļa ideju turpināšanu un iestrādāšanu savās teorijās, (2) klasiskās un modernās mākslas pretnostatījuma pārvarēšanu, savienojot abu veidu mākslas vienā izpratnes shēmā, (3) interpretācijas izvirzīšanu par galveno nos…
Multi-technique approach for qualitative and quantitative characterization of furazidin degradation kinetics under alkaline conditions
2016
Degradation of drug furazidin was studied under different conditions of environmental pH (11-13) and temperature (30-60°C). The novel approach of hybrid hard- and soft-multivariate curve resolution-alternating least squares (HS-MCR-ALS) method was applied to UV-vis spectral data to determine a valid kinetic model and kinetic parameters of the degradation process. The system was found to be comprised of three main species and best characterized by two consecutive first-order reactions. Furazidin degradation rate was found to be highly dependent on the applied environmental conditions, showing more prominent differences between both degradation steps towards higher pH and temperature. Complim…
Structural effects and neurofunctional sequelae of developmental exposure to psychotherapeutic drugs: experimental and clinical aspects
2004
The advent of psychotherapeutic drugs has enabled management of mental illness and other neurological problems such as epilepsy in the general population, without requiring hospitalization. The success of these drugs in controlling symptoms has led to their widespread use in the vulnerable population of pregnant women as well, where the potential embryotoxicity of the drugs has to be weighed against the potential problems of the maternal neurological state. This review focuses on the developmental toxicity and neurotoxicity of five broad categories of widely available psychotherapeutic drugs: the neuroleptics, the antiepileptics, the antidepressants, the anxiolytics and mood stabilizers, an…
Synthesis, labelling and evaluation of hydantoin-substituted indole carboxylic acids as potential ligands for positron emission tomography imaging of…
2011
The N-methyl- d-aspartate (NMDA) receptor as a type of ionotropic glutamatergic receptors is essential for physiological processes such as learning, memory and synaptic plasticity. A glutamate-induced overactivation of these receptors, accompanied by increased intracellular calcium concentration, causes cell injury and leads to a large number of acute or chronic neurological disorders, such as stroke, trauma, Parkinson's disease and Alzheimer's disease. In an attempt to visualise the glutamatergic neurotransmission in vivo with positron emission tomography, novel fluoroethoxy- and methoxy-substituted reference compounds based on the lead structure of a hydantoin-substituted indole-2-carboxy…
Hydantoin-substituted 4,6-dichloroindole-2-carboxylic acids as ligands with high affinity for the glycine binding site of the NMDA receptor.
2002
A novel series of C-3 substituted 4,6-dichloroindole-2-carboxylic acids was synthesized to investigate the influence of different hydrogen-bond donor and acceptor groups at this specific position on the affinity to the glycine site of the NMDA receptor. These novel 3-indolylmethyl derivatives with ring-open (amines, sulfonamides, amides, ureas) and cyclic substituents (imidazolidin-2-ones, (thio)hydantoins) led to the discovery that compounds bearing a hydantoin substituent at the C-3 position of the indole nucleus are the most promising ones. In this series the hydantoins, ureas, and imidazolidin-2-ones were identified as very potent inhibitors of the binding of the glycine site specific l…

In this introduction, we highlight common interests within research in three related educational contexts: language immersion, content and language integrated learning (CLIL) and Finnish or Swedish as second language education in Finland. Although each has their specific foci of interest and have developed as independent research fields, they all share an interest in language learning through meaningful language use as well as in simultaneous learning and teaching of language and content. These issues can be approached from multiple viewpoints as demonstrated by the individual contributions in this volume, introduced briefly in this article. The contributions are organised into three themat…
Diskurssintutkimus - monitieteinen ja monimenetelmäinen ala
2018
There is a growing interest towards metatheoretical examination of linguistic research. This special issue contributes to this examination from the perspective of discourse studies. The issue consists of seven articles which present methods in different areas of discourse studies. The aim of this introductory article is firstly to define the notions of discourse and method. Secondly, through the presentation of the articles of the issue, we present a variety of methods pertaining to different stages of research. In particular, methods of data collection and classification, as well as of data analysis, are presented. We conclude the article with some remarks on the future avenues of the meth…

AFinLA’s research network for Speech and Spoken Language Research was established in 2014. This is the first thematic publication from the network. This introductory article introduces and classifies the other articles in this collection. In addition, we examine earlier AFinLA publications from the preceding four decades and evaluate the extent of pronunciation or spoken language research in AFinLA’s publication series. In our analysis, we noticed that studies related to pronunciation or spoken language research formed ca. 12 % of the total volume of published articles. There has been a slight proportional decrease in number during the decades. Spoken language studies h…
