The possible influence of central nervous structures on the electrical activity of single pineal cells was investigated in rat and guinea-pig. In the rat electrical stimulation of the hippocampal formation elicited both single cell responses with different latencies and mostly long-term excitations in single pineal cells, while stimulation of the habenular nuclei caused clear orthodromical responses with different latencies, alterations in the rate of spontaneous electrical activity and evoked discharges of "silent" units. The Goodpasture antigen-binding protein (GPBP) plays a critical role in brain development. Knockdown of GPBP leads to loss of myelinated tracts in the central nervous system and to extensive apoptosis in the brain during early embryogenesis. GPBP was initially identified as a protein associated with the autoantigen in Goodpasture autoimmune syndrome, where it was shown to be a kinase that regulates type IV collagen organization. GPBP isoforms bind and transport ceramide from the endoplasmic reticulum to the Golgi apparatus and are therefore also known as ceramide transporters (CERT). The possible presence of a direct nervous projection from the paraventricular nucleus (PVN) of the hypothalamus to the pineal gland of the rat was investigated by means of the anterograde neuron-tracing method using horseradish peroxidase. The tracer was injected unilaterally into the PVN and the animals were allowed to survive between 12 and 26 h. Numerous peroxidase-positive fibers were observed, ipsilateral to the injection site, in the stria medullaris thalami and could be followed into the medial habenular nucleus and the habenular commissure. Previous studies have pointed in the direction of sex differences as well as regional differences in the pineal gland of guinea pigs. In the present investigation these aspects were studied at the electron-microscopic level by quantitating different types of “synaptic” bodies, intrinsic to pinealocytes. The distribution of neuropeptide Y-like immunoreactivity (NPY-LI) was investigated by immunohistochemistry and radioimmunoassay (RIA) in the brain of the Djungarian hamster (Phodopus sungorus) held under either long or short photoperiods. In the diencephalic and telencephalic structures studied, distinct patterns of NPY-LI were basically consistent in male and female animals of both groups. Previous studies have shown that the "synaptic" bodies of mammalian pinealocytes are a heterogeneous group of organelles. Whereas "synaptic" ribbons (SR) exhibit a day/night rhythm with small numbers during daytime and high numbers at night, the so-called "synaptic" spherules (SS) show species-specific differences in their rhythmicity. In the present study the numbers of SR and SS were monitored in male guinea pigs over a full 24-h period at 4-hourly intervals (LD 12:12, lights on at 7:00). The results obtained show that the rhythms of SR numbers and SS numbers are out of phase by 180 degrees. 1. Adult New Zealand albino rabbits were prepared with chronic hypothalamic stimulating electrodes and hippocampal recording electrodes. 2. Rabbits were restrained and classically conditioned by a tone CS and an airpuff US either followed or preceded by a hypothalamic stimulation (HS). Control rabbits were conditioned without the HS. 3. It was found that HS following the CS facilitated both behavioral and hippocampal responses, while HS preceding the CS inhibited them. 4. Enhanced hippocampal learning-related unit firing to the CS may represent an early indication of conditioning before the behavioral activity produces any observable change.

Publisher Summary This chapter describes both electrophoretic and catalytic studies on lactate dehydrogenase (LDH) in the developing brain. It has been found that the enzyme exhibits at birth a composition that gradually changes to attain the final pattern observed in the adult. The neurochemical findings on LDH isozymes parallel the rapid growth in the first days after birth. Isozyme E, which is not present in detectable amounts 2 days after birth, represents 15% of the enzyme on the 22nd day. Similarly, the percentage decrease of isozyme B is quite steep at the beginning. Nevertheless, the final pattern is attained only at the end of the 2nd month that is, when the cerebral cortex is full…
