Cryo-negative staining

Abstract A procedure is presented for the preparation of thin layers of vitrified biological suspensions in the presence of ammonium molybdate, which we termcryo-negative staining. The direct blotting of sample plus stain solution on holey carbon supports produces thin aqueous films across the holes, which are routinely thiner than the aqueous film produced by conventional negative staining on a continuous carbon layer. Because of this, a higher than usual concentration of negative stain (ca. 16% rather than 2%) is required for cryo-negative staining in order to produce an optimal image contrast. Small-angle X-ray scattering reveals differences between the quaternary structures of oxygenated and deoxygenated tarantula hemocyanin

Small-angle X-ray scattering (SAXS) curves have been recorded for the oxygenated and deoxygenated states of the 4 x 6-meric hemocyanin from the tarantula Eurypelma californicum. A comparison of the curves shows that the quaternary structures of the two states are different by three criteria, which all indicate that the hemocyanin is less compact in the oxygenated compared to the deoxygenated form: (a) The radius of gyration is 8.65 +/- 0.05 nm for the deoxy- and 8.80 +/- 0.05 nm for the oxy-form. Nucleation and Growth of CaCO3 Mediated by the Egg-White Protein Ovalbumin: A Time-Resolved in situ Study Using Small-Angle Neutron Scattering

Mineralization of calcium carbonate in aqueous solutions starting from its initiation was studied by time-resolved small-angle neutron scattering (SANS). SANS revealed that homogeneous crystallization of CaCO 3 involves an initial formation of thin plate-shaped nuclei which subsequently reassemble to 3-dimensional particles, first of fractal and finally of compact structure. The presence of the egg-white protein ovalbumin leads to a different progression of mineralization through several stages; the first step represents amorphous CaCO 3, whereas the other phases are crystalline. Mineralogical and Chemical Records of Icelandic Dust Sources Upon Ny-Ålesund (Svalbard Islands)

This work aims to identify the geochemical and mineralogical markers of Icelandic dust and to differentiate it from the dust of local origin deposited at the Ny-Ålesund station. We characterized representative sediment samples from Iceland and Svalbard and compared them to a set of aerosol samples collected in Ny-Ålesund to check the existence and type of the mineralogical markers. The sediment samples were analyzed by X-ray diffraction (XRD) to detect the mineralogical constrain of the geochemical markers. A new hydrous Al-bearing pyroxene as a water carrier in subduction zones

Abstract A new Hydrous Al-bearing PYroxene (HAPY) phase has been synthesized at 5.4 GPa, 720 °C in the MgO–Al2O3–SiO2–H2O model system. It has the composition Mg2.1Al0.9(OH)2Al0.9Si1.1O6, a C-centered monoclinic cell with a = 9.8827(2), b = 11.6254(2) c = 5.0828(1) A and β = 111.07(1)°. The calculated density is 3.175 g/cm3 and the water content is 6.9% H2O by weight. Its structure has been solved in space group C2/c by the recently developed automated electron diffraction tomography method and refined by synchrotron X-ray powder diffraction. Incoherent elastic and quasi-elastic neutron scattering investigation of hemoglobin dynamics.

In this work we investigate the dynamic properties of hemoglobin in glycerolD(8)/D(2)O solution using incoherent elastic (ENS) and quasi-elastic (QENS) neutron scattering. Taking advantage of complementary energy resolutions of backscattering spectrometers at ILL (Grenoble), we explore motions in a large space-time window, up to 1 ns and 14 A; moreover, in order to cover the harmonic and anharmonic protein dynamics regimes, the elastic experiments have been performed over the wide temperature interval of 20-300 K. A new method for data evaluation of small angle neutron scattering experiments and its application to amorphous polycarbonate

The conformation of single chains in bulk polymer materials can be evaluated from coherent neutron scattering on mixtures of normal and deuterated polymers. It is shown that the single-chain structure factor can be also obtained from measurements of highly concentrated mixtures and the procedure of evaluation is described. The application to amorphous polycarbonate demonstrates the advantages of the method.

The re Structure of Cyclopropane

A long-standing controversy regarding the re structure of cyclopropane is resolved by performing high-level quantum chemical calculations and analyzing the experimental rotational constants for C3H6 and C3H4D2 augmented by calculated vibrational corrections. For the latter, a least-squares fit yields the following set of parameters:  re (CC) = 1.5030(10) A, re(CH) = 1.0786(10) A, and αe(HCH) = 114.97(10)°, which compare favorably with both the pure computational result obtained at the CCSD(T)/cc-pVQZ level as well as an earlier estimate of the re structure of cyclopropane based on analysis of gas-phase electron diffraction data. A microwave-assisted reaction pathway to rutile SnO2nanorods was investigated. The microwave-treatment significantly reduces the reaction time compared to standard hydro-/solvothermal techniques. By moving the overall process into a shorter time slot, the growth and crystal formation during the reaction could be monitored via snapshots by trapping the intermediates through quenching. Luminescence properties of Tb3+:Y3Al5O12 nanocrystallites prepared by the sol–gel method

Tb 3+ :YAG nanocrystalline powders have been prepared by the sol-gel route. Structure and morphology of the obtained materials have been studied. The average grain sizes have been determined by X-ray powder diffraction measurements. Photo- and cathodoluminescence spectra of the Tb 3+ :Y 3 Al 5 O 12 powders have been measured. It has been found that the samples at low concentration demonstrate emission ascribable to the 5 D 3 →F J and 5 D 4 → 7 F J transitions. In particular, the concentration dependence of the luminescence spectra and lifetimes have been investigated as a function of the YAG grains sizes.
