Int the proximal and distal surface tubules isotonic Na reabsorption (Φ Naout) is impaired by adrenalectomy. In the present experiments the time course of development of transtubular Na concentration differences (Na)P−(Na)TF was studied in the proximal convolution. 1. Na concentration was measured in free flow mannitol diuresis. Endproximal (Na)P−(Na)TF averaged 23.0±0.5 meq/l after 8.5±0.6 sec (controls) and 15.2±1.7 meq/l after 13.6±0.5 sec (adrenalectomized rats). 2. The influence of a synthetic oxytocin preparation and of prednisolone hemisuccinate on the renal excretions of water, sodium, potassium and chloride and on the clearance of endogenous true creatinine was studied in 18 dogs anesthetized with hexobarbital and barbital. The animals were deprived of food and water for 12 hours before the experiments.

Groups of normal rats of both sexes in moderate water diuresis received i.v. injections of 5 ml./kg, of hypertonic (5 times isoosmotic) solutions of sodium chloride, mannitol er sodium sulfate. The renal excretion of water, sodium, chloride and potassium in a 30 minute-period following this injection was measured. It was compared to control values obtained 40 minutes earlier in the same animals after an i.v. injection of isotonic glucose solution. All hypertonic solutions increased diuresis, natriuresis, chloruresis and kaliuresis. The renal excretion of acid and ammonium ions after an oral load of monosodium-phosphate (1.2 mEq. per rat) was studied in groups of adrenalectomized er sham-operated rats, maintained on 0.9 per cent NaCI solution and water, on the 6th or 12th day after operation. Untreated groups were compared to groups receiving either Corticotropin (25 U./kg./day s.c.) or Cortisone acetate (12.5 mg./kg./day s.c.) er (7.5 mg./kg./day s.c.) or D,L-Aldosterone acetate Cortexone acetate (0.15 mg./kg./day s.c.) from the 3rd to the 12th day after opetarion. Water, sodium, potassium and chloride balances, food uptake and changes in body weight were determined in large groups of adrenalectomized or sham-operated rats having permanent access to water, 0.9 per cent saline and rat crackers. Measurements were done on the 5th to 8th day, or else on the 11th to 14th day after operation. No significant improvement in water diuresis was obtained by the s. c. injection of 500–1000 mU/kg of a synthetic oxytocin preparation (Syntocinon) in adrenalectomized, salt-maintained rats given 18 ml of water per rat by stomach tube. In these animals oxytocin had, on the other hand, a strong natriuretic and chloruretic effect.
