Determination of major human cytochrome P450s activities in 96-well plates using liquid chromatography tandem mass spectrometry.
2007
At the early stage of drug discovery, thousands of new chemical entities (NCEs) may be screened before a single candidate can be identified for development. Evaluation of the effect of NCEs on human CYP450 enzyme activities is a key issue in pharmaceutical development as it may explain inter-subject variability, drug-drug interactions, non-linear pharmacokinetics and toxic effects. A liquid chromatography tandem mass spectrometry (HPLC-MS/MS) method has been developed for the fast and routine analysis of major human CYP450s enzyme activities (CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, CYP2D6, CYP2E1 and CYP3A4) in primary hepatocyte cell cultures. The high sensitivity and selectivity of mass …
The Sustainability of Reclaimed Asphalt as a Resource for Road Pavement Management through a Circular Economic Model
2019
The transition of the road engineering industry to a circular way of doing business requires more efficient and sustainable resources, energy, and waste management. The rates in which reclaimed asphalt is being recycled or reused in the asphalt mixture production process constitutes a crucial parameter in this transition. This paper aims at establishing a further step towards the combined circularity and sustainability of asphalt pavements, by introducing a framework for quantifying their Material Circularity Index. The framework is based on the methodology proposed by the Ellen MacArthur Foundation and accordingly tailored for the context of asphalt pavements. This study, thus, attempts to…
Comparing the Material Circularity Index of the Italian and Spanish motorways: A case study over the Mediterranean basin
2022
Closely following the attempt of the pavement engineering industry to comply with the principles of Circular Economy (CE) while simultaneously promoting its principles, it can be detected that a significant effort is being made towards the minimization of the energy and raw materials usually utilized. In this regard, Reclaimed Asphalt (RA) is a product that exhibits a tremendous exploitation potential. Knowing that by definition CE is restorative [1], regenerative, and aims to keep products, components, and materials at their highest utility and value at all times [2], i.e. it supports the “re-circulation” of materials and energy within the same or alternative product systems and thus the e…
Strategies to In Vitro Assessment of Major Human CYP Enzyme Activities by Using Liquid Chromatography Tandem Mass Spectrometry
2008
At the early stage of drug discovery, thousands of new chemical entities (NCEs) may be screened before a single candidate can be identified for development. Determining the role of CYP enzymes in the metabolism of a compound and evaluating the effect of NCEs on human CYP activities are key issues in pharmaceutical development as they may explain inter-subject variability, drug-drug interactions, non-linear pharmacokinetics and toxic effects. Reliable methods for determining enzyme activities are needed to characterize an individual CYP enzyme and to obtain a tool for the evaluation of its role in drug metabolism in humans. Different liquid chromatography tandem mass spectrometry methodologi…
The Correlation Between Abnormal Uterine Artery Flow in the First Trimester and Genetic Thrombophilic Alteration: A Prospective Case-Controlled Pilot…
2020
Introduction. Evaluation of the first trimester uterine artery flow can predict the development of obstetrical complications. A genotype, making women prone to microthrombi. constitutes the main known susceptibility factor for anomalous development of placenta. Our aim was to study whether polymorphisms of 10 genes leading to blood clotting abnormalities are related to abnormal uterine artery blood flow in the first trimester, and may predict placenta-related diseases. Material and methods. In primary analyses we included 19 singleton pregnancies with abnormal blood flow in the uterine arteries during the first trimester of gestation, and 24 matched control with normal flow patterns. All pa…
Changes in Speech Range Profile Are Associated with Cognitive Impairment
2021
Background and Purpose The aim of this study was to describe the variations in the speech range profile (SRP) of patients affected by cognitive decline. Methods We collected the data of patients managed for suspected voice and speech disorders, and suspected cognitive impairment. Patients underwent an Ear Nose and Throat evaluation and Mini-Mental State Examination (MMSE). To obtain SRP, we asked the patients to read 18 sentences twice, at their most comfortable pitch and loudness as they would do in daily conversation, and recorded their voice on to computer software. Results The study included 61 patients. The relationship between the MMSE score and SRP parameters was established. Increas…
Geometry/Time Measurement/Sundials Graphical Resolution via Algorithmic and Parametric Processes
2018
Every people, in every historical period, developed methods to measure Time both at a daily scale and at a yearly scale. Some of them constructed sundials to represent the apparent trajectory of the Sun around the Earth, by using and developing tools from descriptive and projective Geometry, mainly. This subject acquired a great multidisciplinary interest since ancient times, also for Science of Representation applications. This study presents the first results of an ongoing research concerning some aspects related to Time Measurement. The geometric-spatial setting of the Sun-Earth system is described and is structured parametrically via algorithms, following the known conventions shared an…
Preclinical models for colonic absorption, application to controlled release formulation development.
2018
Oral controlled release (CR) formulations have many benefits and have become a valuable resource for the local and systemic administration of drugs. The most important characteristic of these pharmaceutical products is that drug absorption occurs mainly in the colon. Therefore, this review analyses the physiological and physicochemical features that may affect an orally administered CR product, as well as the different strategies to develop a CR dosage form and the methods used to evaluate the formulation efficacy. The models available to study the intestinal permeability and their applicability to colonic permeability determinations are also discussed.

The impact of organic amendments on soil hydrology, structure and microbial respiration in semiarid lands
2016
Abstract Few studies have considered the effect of organic amendments on soil microbial activity and its contributions to hydraulic conductivity under field conditions in semiarid region soils with different textures and degrees of aggregate stability. This study was performed to investigate the relationship between selected soil properties and hydraulic conductivity in response to different types and application rates of organic amendments. For this purpose, urban municipal solid waste (MSW) compost and alfalfa residue (AR) were applied at different rates of 0 (control), 10 Mg ha− 1 and 30 Mg ha− 1 to clay loam and loamy sand soils under field conditions. Results show that after two years,…
