Here, the preparation of a novel block copolymer consisting of a statistical copolymer N-(2-hydroxypropyl) methacrylamide-s-N-(3-aminopropyl) methacrylamide and a short terminal 3-guanidinopropyl methacrylamide block is reported. This polymer structure forms neutral but water-soluble nanosized complexes with siRNA. The siRNA block copolymer complexes are first analyzed using agarose gel electrophoresis and their size is determined with fluorescence correlation spectroscopy. EPR and ENDOR spectra were recorded for 2-methyl-9,10-anthraquinone (2-methylAQ), 2-ethylAQ, 2-tertbutylAQ and 2,3-dimethylAQ anion radicals in the solution phase. The EPR spectra were simulated with the help of ENDOR data. The experimental isotropic hyperfine coupling constants (IHFCs) were compared with calculated values from semi-empirical INDO, spin-restricted AM1/CI and B3PW91 density-functional methods. The best computational methods for the IHFCs were the semi-empirical AM1/CI method and the B3PW91 density-functional method with a large basis set.

Decapping is a central step in eukaryotic mRNA turnover and in gene expression regulation. The human scavenger decapping enzyme, DcpS, catalyses cap hydrolysis following mRNA degradation. DcpS is a dimeric enzyme, with two active sites. Crystal structures suggest that DcpS must undergo significant conformational changes upon ligand binding, but the mechanism of this transition is unknown. Here, we report two long timescale (20 ns) molecular dynamics simulations of the apo-form of DcpS. The dimer is observed to undergo a strikingly cooperative motion, with one active site closing while the other opens. Sauna bath brings about numerous acute changes in hormone levels, partly akin to other stressful situations, partly specific for sauna. Norepinephrine increases in those accustomed to sauna bath. Sweating increases the production of antidiuretic hormone, and the renin-angiotensin system becomes activated. Of the anterior pituitary hormones, growth hormone (GH) and prolactin (PRL) secretion is increased. Also β-endorphin has been frequently reported to increase, whereas the responses of ACTH and cortisol are variable, probably depending on the type of sauna exposure. Vaikka kansallisesti kohdennetaan runsaasti resursseja tietojärjestelmäpalveluiden kehittämiseen, sosiaalialalta tutkimusta aiheesta löytyy niukasti. Sosiaalialan ammattilaisten kokemuksia asiakastietojärjestelmien kehittämistyöstä kartoitettiin osana keväällä 2019 toteutettua valtakunnallisesta käyttäjäkokemuskyselyä.
