Comparison of Four Commercial DNA Extraction Kits for PCR Detection of Listeria monocytogenes, Salmonella, Escherichia coli O157:H7, and Staphylococc…
2008
Four commercial DNA extraction methods, PrepMan Ultra (Applied Biosystems), InstaGene Matrix (BioRad), DNeasy Tissue kit (Qiagen), and UltraClean (MoBio), were tested for PCR detection of Listeria monocytogenes, Escherichia coli O157: H7, Salmonella, and Staphylococcus aureus in fresh, minimally processed vegetables. For comparative purposes, sensitivity assays with specific PCRs were carried out after DNA extraction with the four methods in green pepper, broccoli, and onion artificially inoculated with the four pathogens separately. As confirmed by statistical analysis, the DNeasy Tissue kit rendered the highest sensitivity values in the three matrices assayed for Salmonella, L. monocytoge…
The All-Rounder Sodalis: A New Bacteriome-Associated Endosymbiont of the Lygaeoid Bug Henestaris halophilus (Heteroptera: Henestarinae) and a Critica…
2017
International audience; Hemipteran insects are well-known in their ability to establish symbiotic relationships with bacteria. Among them, heteropteran insects present an array of symbiotic systems, ranging from the most common gut crypt symbiosis to the more restricted bacteriome-associated endosymbiosis, which have only been detected in members of the superfamily Lygaeoidea and the family Cimicidae so far. Genomic data of heteropteran endosymbionts are scarce and have merely been analyzed from the Wolbachia endosymbiont in bed bug and a few gut crypt-associated symbionts in pentatomoid bugs. In this study, we present the first detailed genomic analysis of a bacteriome-associated endosymbi…
Antimicrobial Lessons From a Large Observational Cohort on Intra-abdominal Infections in Intensive Care Units
2021
Severe intra-abdominal infection commonly requires intensive care. Mortality is high and is mainly determined by disease-specific characteristics, i.e. setting of infection onset, anatomical barrier disruption, and severity of disease expression. Recent observations revealed that antimicrobial resistance appears equally common in community-acquired and late-onset hospital-acquired infection. This challenges basic principles in anti-infective therapy guidelines, including the paradigm that pathogens involved in community-acquired infection are covered by standard empiric antimicrobial regimens, and second, the concept of nosocomial acquisition as the main driver for resistance involvement. I…
EUCAST un CLSI metožu salīdzinājums paplašināta spektra βeta-laktamāžu noteikšanai Enterobacteriaceae dzimtas klīniskajos izolātos
2015
Darba mērķis bija salīdzināt EUCAST un CLSI antimikrobās jutības noteikšanas metodes paplašināta spektra β-laktamāžu noteikšanai Enterobacteriaceae dzimtas klīniskajos izolātos. Darba teorētiskajā daļā tika apskatīti biežākie ESBL producējošie Enterobacteriaceae dzimtas pārstāvji, antibakteriālo līdzekļu un rezistences mehānismu raksturojums. Dati iegūti 2014.-2015. g., strādājot klīniskajā mikrobioloģijas laboratorijā. Identificēti un pētīti 269 baktēriju izolāti. Pēc EUCAST standarta 55 izolātiem mainījās antibiogrammas rezultāti. Ceftazidīmam un cefotaksīmam samazinot antibakteriālā līdzekļa koncentrāciju pēc EUCAST standarta, tie tiek padarīti jutīgāki pret ESBL noteikšanu. Pateicoties …
Paplašināta spektra beta laktamāžu noteikšana enterobacteriaceae dzimtas baktērijās
2020
Enterobacteriaceae dzimta ir plaši pārstāvēta mikroorganismu dzimta, kuru veido medicīniski nozīmīgas gramnegatīvas nūjiņas. Šīs dzimtas pārstāvji izraisa infekciju saslimšanas gan vispārējā populācijā, gan medicīnas iestādēs (nozokomiālas infekcijas), skarot kā veselus cilvēkus, tā cilvēkus jau ar esošu saslimšanu. Tās pārstāvji- E.coli un K.pneumoniae, ir vieni no biežākajamiem smagu nozokomiālu infekciju izraisītājiem pasaulē. Arvien pieaug Enterobacteriaceae rezistence pret antibakteriālajiem līdzekļiem, tai skaitā pret paplašināta spektra beta laktāmu antibiotikām. Tas būtiski palielina hospitalizācijas ilgumu, finansiālo slogu kā pacientiem, tā valstij un dēļ tā pieaug pacientu mirstī…
Zanieczyszczenie bioaerozolu w zakładzie cukierniczym bakteriami rodziny Enterobacteriaceae
2018
Badania przeprowadzono w zakładzie cukierniczym w trakcie produkcji, w dwóch halach technologicznych (H.1, H.2). W każdej z hal o zróżnicowany…
Study on the application of an interspecific competition model for the prediction of microflora behaviour during the fermentation process of S. Angel…
2009
The use of predictive microbiology models able to evaluate bacterial behaviour as a function of environmental conditions and, at the same time, of natural microflora competition was considered by several authors with different approaches. Some authors modelled bacterial competition as a function of metabolic product with particular regard to lactic acid and modelled interspecific bacterial competition introducing a term into a conventional primary predictive model, which gives account for the interaction between two populations, so that they inhibit each other to the same extent that they inhibit their own growth.
Activation of the contact-phase system on bacterial surfaces--a clue to serious complications in infectious diseases.
1998
Fever, hypotension and bleeding disorders are common symptoms of sepsis and septic shock. The activation of the contact-phase system is thought to contribute to the development of these severe disease states by triggering proinflammatory and procoagulatory cascades; however, the underlying molecular mechanisms are obscure. Here we report that the components of the contact-phase system are assembled on the surface of Escherichia coli and Salmonella through their specific interactions with fibrous bacterial surface proteins, curli and fimbriae. As a consequence, the proinflammatory pathway is activated through the release of bradykinin, a potent inducer of fever, pain and hypotension. Absorpt…
The coffee-machine bacteriome: biodiversity and colonisation of the wasted coffee tray leach
2015
AbstractMicrobial communities are ubiquitous in both natural and artificial environments. However, microbial diversity is usually reduced under strong selection pressures, such as those present in habitats rich in recalcitrant or toxic compounds displaying antimicrobial properties. Caffeine is a natural alkaloid present in coffee, tea and soft drinks with well-known antibacterial properties. Here we present the first systematic analysis of coffee machine-associated bacteria. We sampled the coffee waste reservoir of ten different Nespresso machines and conducted a dynamic monitoring of the colonization process in a new machine. Our results reveal the existence of a varied bacterial community…
Alternative for improving gut microbiota: use of Jerusalem artichoke and probiotics in diet of weaned piglets
2014
AbstractThe aim of the study was to determine the effect of Jerusalem artichoke and probiotics on defence activity of intestinal cells of weaning pigs. One hundred eighty piglets (7 weeks old) were fed with basal feed supplemented with Jerusalem artichoke, Lactobacillus reuteri and Pediococcus pentosaceus. After 5 weeks, the piglets were slaughtered and the gastrointestinal contents and intestine samples were taken for analysis. Results demonstrated that in pigs fed basal diet with both probiotics and Jerusalem artichoke (5% of basal diet) (T3 group) had less (P<0.05) faecal Enterobacteriaceae microorganisms and coliforms and had more (P<0,05) faecal Lactobacillus than in pigs from ot…
