We developed a sensitive fluorescence-based assay for determination of exosome concentration. In our assay, Cholera toxin subunit B (CTB) conjugated to a fluorescence probe and a gel filtration technique (size-exclusion chromatography) are used. Exosomal membranes are particularly enriched in raft-forming lipids (cholesterol, sphingolipids, and saturated phospholipids) and in GM1 ganglioside. CTB binds specifically and with high affinity to exosomal GM1 ganglioside residing in rafts only, and it has long been the probe of choice for membrane rafts. EV-TRACK: transparent reporting and centralizing knowledge in extracellular vesicle research

We argue that the field of extracellular vesicle (EV) biology needs more transparent reporting to facilitate interpretation and replication of experiments. To achieve this, we describe EV-TRACK, a crowdsourcing knowledgebase (http://evtrack.org) that centralizes EV biology and methodology with the goal of stimulating authors, reviewers, editors and funders to put experimental guidelines into practice.

Exosomes are lipoproteic nanosize vesicles of endocytic origin that are released in the extracellular space by several cell types when a multivesicular body fuses with the plasma membrane (1). To date, accumulating evidences highlight the potential of exosomes as messenger in intercellular communication. Indeed, several studies reported the presence of lipids, proteins, metabolites and nucleic acid (DNA, mRNA, microRNA, lncRNA) in exosomes and highlight their functional role in physiological and pathological processes (2).
