Size‐Exclusion Chromatography in Single and Mixed Eluents: Effects of Sorption of Solvent and Polymer Solutes on Novel TSK‐Gel Super AW Columns

2005

Abstract The novel TSK Gel Super AW column packing is based on a polymeric gel with residual hydrophobic and hydrophilic groups on the gel surface. These groups can act as active sites making the chromatographic support compatible both with polar and apolar eluents. This interesting property allows the elution of systems of very different polarities but also induces binary polymer–gel, polymer–solvent, and solvent–gel enthalpic interactions that lead to a mixture of separation mechanisms: repulsion, partition, or adsorption besides the main size‐exclusion chromatography (SEC) mechanism. For this reason, the elution behaviour of many and diverse polymer/solvent systems has been studied in th…

Determination of the four major surfactant classes in cleaning products by reversed-phase liquid chromatography using serially connected UV and evapo…

2016

Abstract A method for the simultaneous determination of the most frequently used surfactant families –linear alkyl benzenesulphonates (LAS), alkyl ether sulphates (AES), fatty alcohol ethoxylates (FAE) and oleins (soaps, fatty acid salts) – in cleaning products, has been developed. The common reversed phase octyl (C8), pentafluorophenyl and biphenyl columns were not capable of separating the anionic LAS and AES classes; however, since only LAS absorbs in the UV, these two classes were independently quantified using a C8 column and serially connected UV and ELSD detection. The best compromise to resolve the four surfactant classes and the oligomers within the classes was achieved with a C8 c…

Precipitation chromatography. III. Optimization of the experimental conditions and determination of the polydispersity of fractions

1974

The influence of the solvent–nonsolvent gradient and the temperature gradient on the efficiency of the precipitation chromatography is investigated with radically and anionically prepared polystyrene samples. Only a combination of both gradients gives optimal effectiveness of the column. The separating power of the column was determined by using mixtures of well defined samples. From the elution curves of the mixtures the nonuniformity Ufr of fractions can be estimated. For fractions of anionically prepared polystyrenes Ufr is on the average 0.001; this value is of the same order of magnitude as the statistical error. These results show that with this chromatographic method it is possible t…

The second component of human complement: Detection of two hemolytic forms in plasma by pH Variation

1988

The second component of human complement (C2) in pseudoglobulin prepared from normal plasma eluted as a single peak at high conductivity (30 mS) and pH 4.5 from the cationic exchangers S-Sepharose or Mono S in the Fast Protein Liquid Chromatography (FPLC) System. The C2 was stable at pH 4.5 and 0 degrees C if enzyme inhibitors were used and the pH was raised to 6.0 after elution from the columns. After rechromatography on Mono S in the FPLC System at the median isoelectric point of 5.5 or pH 6.0, the C2 eluted as two distinct hemolytic forms: the first peaked at 16 mS, the second at 30 mS. The two forms of C2 did not correlate with the allotypic variant of C2 in individual, normal human pla…

Single-pump bi-dimensional LC applied to the characterization of derivatized fatty alcohol ethoxylates.

2010

A column selection valve and a 6-port 2-position injection valve were combined to implement bi-dimensional LC in a system driven by a single pump. The system was applied to the separation of linear fatty alcohol ethoxylates with 12-18 carbon atoms in the hydrocarbon moiety, previous derivatization with diphenic anhydride. Separation on the first separation dimension (C(1)) was carried out with a propyl-diol column and an acetonitrile/aqueous ammonium acetate gradient. Fatty alcohol ethoxylate hydrocarbon series (oligomers with the same number of carbon atoms in the alkyl chain but differing in the number of ethylene oxide units) were resolved without distinguishing the oligomers within the …

Simultaneous gas chromatographic determination of aliphatic C2−C20 n-carboxylic acids and their methyl esters using SE-30 and OV-351 quartz capillary…

1983

Simultaneous determination of aliphatic C2−C20 carboxylic acids and their methyl esters was carried out using SE-30 and OV-351 quartz capillary columns with temperature programming. Complete resolution of all 36 components was achieved. On SE-30 the methyl ester and the corresponding free acid are eluted in turn, whereas on OV-351 acetic acid eluted after methyl octanoate, after which the Cn methyl ester and Cn-6 carboxylic acid (n>9) are eluted one after the other. A non-polar SE-30 column is better for long-chain acids giving sharp peaks without tailing, the analysis time of the mixture being less than 16 minutes. The weight response correction factors for compounds under optimum operatin…

Characterization of aquatic humic substances and their metal complexes by immobilized metal-chelate affinity chromatography on iron(III)-loaded ion e…

2001

The analytical fractionation of aquatic humic substances (HS) by means of immobilized metal-chelate affinity chromatography (IMAC) on metal-loaded chelating ion exchangers is described. The cellulose HYPHAN, loaded with different trivalent ions, and the chelate exchanger Chelex 100, loaded to 90% of its capacity with Fe(III), were used. The cellulose HYPHAN, loaded with 2% Fe(III), resulted in HS distribution coefficients Kd of up to 10(3.7) mL/g at pH 4.0 continuously decreasing down to 10(1.5) at pH 12, which were appropriate for HS fractionation by a pH-depending chromatographic procedure. Similar distribution coefficients Kd were obtained for HS sorption onto Fe(III)-loaded Chelex 100. …

Separation studies of amino acids, proteins and enzymes on bonded 1,2-dihydroxy-, 1,2-hydroxylamino-and amino silica packings

1979

1,2-dihydroxy-3-propoxypropyl (HPPS), 1-amino-2-hydroxy-3-propoxypropyl (AHPS) and 1-aminoethyl-3-aminopropyl (AEAPS) silica were synthesized by means of both a surface modification procedure (I) and a bulk modification procedure (II). Method (I) gave a surface concentration, α, of functional groups of 2–3 μmole/m2, whereas method (II) gave values up to 5 μmole/m2. Retention times, peak asymmetries and plate heights of thiamine and ascorbic acid eluted with aqueous buffer solutions ranging from pH 5.3 to 9.2 gave only a±5% variation over periods of 12 hours and more. The recoveries of selected enzymes and proteines examined under static and dynamic conditions were between 60% and 100% depen…

Fast high performance liquid chromatography analysis in lipidomics: Separation of radiolabelled fatty acids and phosphatidylcholine molecular species…

2006

Abstract HPLC procedures using conventional C 18 columns are usually used to separate simple and complex lipid mixtures but these methods of separation remain often laborious and very slow. Here, monolithic columns were successfully applied to separate lipids – radiolabelled fatty acid mixtures and individual phosphatidylcholine (PC) molecular species. For that, isocratic elution was performed using two Chromolith™ Performance RP-18e columns connected in series. Detection was achieved by online measurement of radioactivity for radiolabelled fatty acids and by UV absorbance at 205 nm for PC molecular species. The performances of such silica rods were compared to conventional reverse-phase si…

Application of CEC procedures for the analysis of synthetic peptides: characterization of linear immunogenic peptides that mimic a HIV-1 gp120 epitope

2002

In this study, we describe the application of a new analytical procedure based on capillary electrochromatographic(CEC) techniques for the characterization of different basic and acidic peptides using isocratic eluent conditions containing acetonitrile and ammonium acetate buffers of different molarities between pH 3.8 and 5.2. In particular,10 immunogenic peptide analogs with isoelectric points ranging from 3.7 to 10.1 were investigated; nine of these peptides, 1-9, were truncated analogs of the parent peptide, 10, which is a peptidomimetic related to a HIV-1 gp120 epitope. Several of these peptides have the propensity to form alpha-helical secondary structures in solution. Electrochromato…
