ID4 Is Required for Normal Ependymal Cell Development

2021

Ependymal cells are radial glia-derived multiciliated cells lining the lateral ventricles of the brain and spinal cord. Correct development and coordinated cilia beating is essential for proper cerebrospinal fluid (CSF) flow and neurogenesis modulation. Dysfunctions of ependymal cells were associated with transcription factor deregulation. Here we provide evidence that the transcriptional regulator ID4 is involved in ependymal cell development and maturation. We observed that Id4-deficient mice display altered ventricular cell cytoarchitecture, decreased ependymal cell number and enlarged ventricles. In addition, absence of ID4 during embryonic development resulted in decreased ependymal ce…

Stage of the estrous cycle at the time of pregnant mare's serum gonadotropin injection affects pre-implantation embryo development in vitro in the mo…

2002

The present study aims to analyze in the mouse the effect of the stage of the estrous cycle at the time of pregnant mare's serum gonadotropin (PMSG) injection on fertilization of ovulated cumulus-enclosed oocytes and later embryo development in vitro to the blastocyst stage. Quality of blastocysts was evaluated by staining and counting of total number of nuclei, mitotic index, percentage of apoptotic nuclei, and cell allocation to the inner cell mass (ICM) and trophectoderm (TE) lineage. Superovulation of hybrid (C57Bl/6JIco female x CBA/JIco male) female mice of 4-6 weeks of age was induced by a priming injection of PMSG at different stages of the estrous cycle followed after a 48-hr inter…

Nanog Regulates Proliferation During Early Fish Development

2009

Abstract Nanog is involved in controlling pluripotency and differentiation of stem cells in vitro. However, its function in vivo has been studied only in mouse embryos and various reports suggest that Nanog may not be required for the regulation of differentiation. To better understand endogenous Nanog function, more animal models should be introduced to complement the murine model. Here, we have identified the homolog of the mammalian Nanog gene in teleost fish and describe the endogenous expression of Ol-Nanog mRNA and protein during medaka (Oryzias latipes) embryonic development and in the adult gonads. Using medaka fish as a vertebrate model to study Nanog function, we demonstrate that …

Effect of the Bovine Oviductal Fluid onIn VitroFertilization, Development and Gene Expression ofIn Vitro-Produced Bovine Blastocysts

2012

Oviductal microenvironment generally provides better condi-tions for early embryo development than the conventionalin vitro system. In an attempt to simulate the oviductconditions or the main potentially influencing factors, theeffect was studied of a bovine oviductal fluid (bOF) treatmentapplied prior to IVF on (i) IVF parameters, (ii) cleavage rate,(iii) blastocyst yield and (iv) blastocyst quality. Embryo qualitywas assessed by morphological embryo quality and relativetranscript abundance of several developmental genes in bovineblastocysts. Furthermore, to study the effect of bOF withoutthe male effect and zona–sperm interaction, artificially acti-vated metaphase II oocytes were also treated w…

Galloway-Mowat syndrome: New insights from bioinformatics and expression during Xenopus embryogenesis.

2021

Abstract Galloway-Mowat syndrome (GAMOS) is a rare developmental disease. Patients suffer from congenital brain anomalies combined with renal abnormalities often resulting in an early-onset steroid-resistant nephrotic syndrome. The etiology of GAMOS has a heterogeneous genetic contribution. Mutations in more than 10 different genes have been reported in GAMOS patients. Among these are mutations in four genes encoding members of the human KEOPS ( k inase, e ndopeptidase and o ther p roteins of small s ize) complex, including OSGEP, TP53RK, TPRKB and LAGE3. Until now, these components have been functionally mainly investigated in bacteria, eukarya and archaea and in humans in the context of t…

Other astacin homologs

2013

Publisher Summary This chapter describes the activity, specificity and structural chemistry of astacin homologs. The astacins are members of the metzincin superfamily such as the serralysins, the reprolysins/adamalysins, the matrixins, the leishmanolysins, the pregnancy-associated plasma proteins, the snapalysins and the fragilysins. Proteins of the hatching subfamily have been shown to be important for the cleavage of membranes coating developing embryos of invertebrates and vertebrates. Other members of this subfamily have varying or even several functions. UVS.2 from Xenopus, originally shown to play a role in dorso-anterior development, has been identified as the frog hatching enzyme. T…

Micropropagation of Citrus spp. by Organogenesis and Somatic Embryogenesis.

2013

Citrus spp., the largest fruit crops produced worldwide, are usually asexually propagated by cuttings or grafting onto seedling rootstocks. Most of Citrus genotypes are characterized by polyembryony due to the occurrence of adventive nucellar embryos, which lead to the production of true-to-type plants by seed germination. Tissue culture and micropropagation, in particular, are valuable alternatives to traditional propagation to obtain a high number of uniform and healthy plants in a short time and in a small space. Moreover, in vitro propagation provides a rapid system to multiply the progeny obtained by breeding programs, allows the use of monoembryonic and seedless genotypes as rootstock…

Somatic embryogenesis and plant regeneration from pistil transverse thin cell layers of lemon (Citrus limon)

2008

Callus induction, somatic embryogenesis and plant regeneration were obtained in Citrus limon (L.) Burm. (cv. Femminello) from cultures of pistil transverse thin cell layer explants [(t)TCLs]. Explants were cultured on two different media, based on Murashige and Skoog salts and vitamins, supplemented with 500 mg l-1 malt extract (MSI), or 500 mg l-1 malt extract and 13.3 ¼M 6-benzylaminopurine (MSII). Sucrose (146 mM) was used as carbon source. Somatic embryos appeared 3 months after culture initiation from stigma and style (t)TCLs; they were observed at the surface of the (t)TCL-derived callus. Although ovary (t)TCLs showed the highest callus formation, they never differentiated somatic emb…
