Abnormal peripubertal development of the rat mammary gland following exposure in utero and during lactation to a mixture of genistein and the food co…
2011
The impact of early exposure to endocrine disruptor mixtures on mammary gland development is poorly known. Here, we identify the effects of a conception to weaning exposure of rats to the phytoestrogen genistein (G) and/or the antiandrogen vinclozolin (V) at 1 mg/kg-d, alone or in association. Using several approaches, we found that G- and GV-exposed rats displayed significantly greater epithelial branching and proliferation, wider terminal end buds than controls at PND35, as well as ductal hyperplasia and periductal fibrosis. Focal branching defects were present in V-exposed rats. An increased ER and AR expression was observed in G- and CV- as compared to V-exposed rats at PND35. Surprisin…
Concept of dynamic DNA network dedicated to DNA-Protein interactions studies
2007
The approach lies on the development of an original DNA biosensor. This self-assembled DNA biochip is constituted of a tethered lipid membrane presenting « protein/DNA » complexes. This unique assembly, using a highly flexible bifunctional molecular spacer and a fluidic matrix, allows the control of oligonucleotides surface density and their mobility thus favouring hybridization of the DNA probes in liquid environment and under weak constraints. Recent Surface Plasmon Resonance experiments lead to building of protein/DNA dimers presenting two special DNA sequences recognized by regulation proteins. We demonstrated the capacity and relevance of this biosensor in the case of an estrogen recep…

Molecular Basis for Endocrine Disruption by Pesticides Targeting Aromatase and Estrogen Receptor
2020
The intensive use of pesticides has led to their increasing presence in water, soil, and agricultural products. Mounting evidence indicates that some pesticides may be endocrine disrupting chemicals (EDCs), being therefore harmful for the human health and the environment. In this study, three pesticides, glyphosate, thiacloprid, and imidacloprid, were tested for their ability to interfere with estrogen biosynthesis and/or signaling, to evaluate their potential action as EDCs. Among the tested compounds, only glyphosate inhibited aromatase activity (up to 30%) via a non-competitive inhibition or a mixed inhibition mechanism depending on the concentration applied. Then, the ability of the thr…
Heterocyclisch anellierte Steroide aus 2-Hydroxymethylen-canrenon
1995
A-ring annulated heterocycles, the isoxazole 6, the pyrazoles 8 and the pyrimidines 9 are prepared starting from 2-hydroxymethylene canrenone 1. Binding studies were carried out with the compounds 1 and 6-8 using estrogen, progesterone, androgen, gluco- and mineralocorticoid receptors as well as the serum proteins SHBG and CBG: the substances were inactive on the receptor level. 1, 7 and 8a show weak binding affinity to CBG.

Use of Early Juvenile Zebrafish Danio Rerio for In-Vivo Assessment of Endocrine Modulation by Xenoestrogens
2013
Reliable and cost-effective early-life stage (ELS) bioassays incorporating practical experimentation without compromising scientific relevance are crucial in chemical risk assessment. This study investigated the use of 20 days- post-fertilization life stage ( 20dpfZF ) of zebrafish Danio rerio to screen environmental chemicals known to be estrogenic in adult fish. Firstly, studies with key genes in steroidogenesis were conducted; the brain isoform of aromatase gene ( cyp19a1b ) being the most prominently expressed biomarker. Regulation of mRNA levels of molecular biomarkers, vitellogenin 1 gene ( vtg1) and cyp19a1b were selected to assess the endocrine modulation by xenoestrogens, 17α-ethin…
