MultiBacTAG-Genetic Code Expansion Using the Baculovirus Expression System in Sf21 Cells

The combination of genetic code expansion (GCE) and baculovirus-based protein expression in Spodoptera frugiperda cells is a powerful tool to express multiprotein complexes with site-specifically introduced noncanonical amino acids. This protocol describes the integration of synthetase and tRNA gene indispensable for GCE into the backbone of the Bacmid, the Tn7-mediated transposition of various genes of interest, as well as the final expression of protein using the MultiBacTAG system with different noncanonical amino acids.

Cryptotanshinone deregulates unfolded protein response and eukaryotic initiation factor signaling in acute lymphoblastic leukemia cells.

Abstract Background: Unfolded protein responses (UPR) determine cell fate and are recognized as anticancer targets. In a previous research, we reported that cryptotanshinone (CPT) exerted cytotoxic effects toward acute lymphoblastic leukemia cells through mitochondria-mediated apoptosis. Purpose: In the present study, we further investigated the role of UPR in CPT-induced cytotoxicity on acute lymphoblastic leukemia cells by applying tools of pharmacogenomics and bioinformatics. Methods: Gene expression profiling was performed by mRNA microarray hybridization. Long-Term Aspartame Administration Leads to Fibrosis, Inflammasome Activation, and Gluconeogenesis Impairment in the Liver of Mice

Background: Aspartame is an artificial sweetener used in foods and beverages worldwide. However, it is linked to oxidative stress, inflammation, and liver damage through mechanisms that are not fully elucidated yet. This work aimed to investigate the effects of long-term administration of aspartame on the oxidative and inflammatory mechanisms associated with liver fibrosis progression in mice. Methods: Mice were divided into two groups with six animals each: control and aspartame. Aspartame (80 mg/kg, via oral) or vehicle was administrated for 12 weeks. A numerical property of Hilbert functions and lex segment ideals

We introduce the fractal expansions, sequences of integers associated to a number. We show that these sequences characterize the Osequences and encode some information about lex segment ideals. Moreover, we introduce numerical functions called fractal functions, and we use them to solve the open problem of the classification of the Hilbert functions of any bigraded algebra.

On the Betti numbers of three fat points in P1 × P1

In these notes we introduce a numerical function which allows us to describe explicitly (and nonrecursively) the Betti numbers, and hence, the Hilbert function of a set Z of three fat points whose support is an almost complete intersection (ACI) in P1 × P1 . A nonrecursively formula for the Betti numbers and the Hilbert function of these configurations is hard to give even for the corresponding set of five points on a special support in P2 and we did not find any kind of this result in the literature. Moreover, we also give a criterion that allows us to characterize the Hilbert functions of these special set of fat points.

On presentations for mapping class groups of orientable surfaces via Poincaré's Polyhedron theorem and graphs of groups

The mapping class group of an orientable surface with one boundary component, S, is isomorphic to a subgroup of the automorphism group of the fundamental group of S. We call these subgroups algebraic mapping class groups. An algebraic mapping class group acts on a space called ordered Auter space. We apply Poincaré's Polyhedron theorem to this action. We describe a decomposition of ordered Auter space. From these results, we deduce that the algebraic mapping class group of S is a quotient of the fundamental group of a graph of groups with, at most, two vertices and, at most, six edges. Automorphisms of 2–dimensional right-angled Artin groups

We study the outer automorphism group of a right-angled Artin group AA in the case where the defining graph A is connected and triangle-free. We give an algebraic description of Out.AA/ in terms of maximal join subgraphs in A and prove that the Tits’ alternative holds for Out.AA/. We construct an analogue of outer space for Out.AA/ and prove that it is finite dimensional, contractible, and has a proper action of Out.AA/. We show that Out.AA/ has finite virtual cohomological dimension, give upper and lower bounds on this dimension and construct a spine for outer space realizing the most general upper bound. 20F36; 20F65, 20F28

Hypertrophy and ER Stress Induced by Palmitate Are Counteracted by Mango Peel and Seed Extracts in 3T3-L1 Adipocytes

A diet rich in saturated fatty acids (FAs) has been correlated with metabolic dysfunction and ROS increase in the adipose tissue of obese subjects. Thus, reducing hypertrophy and oxidative stress in adipose tissue can represent a strategy to counteract obesity and obesity-related diseases. In this context, the present study showed how the peel and seed extracts of mango (Mangifera indica L.) reduced lipotoxicity induced by high doses of sodium palmitate (PA) in differentiated 3T3-L1 adipocytes.
