Characterization Of Casein-Loaded Proteoliposomes, Potential Inhibitors In Amyloid Fibrillogenesis
Casein-loaded proteoliposomes: Drug Delivery Systems and Potential Inhibitors in Aβ1-40 Fibrillogenesis
2018
Alzheimer's disease (AD) represents the most common cause of dementia worldwide. The early symptom is usually a short-term memory loss, followed by symptoms including problems with language, disorientation, mood swings, loss of motivation, not managing self-care, and behavioral issues, until loss of body functions and, ultimately, death. The cause of AD is poorly understood and the diagnosis is complex. One of the main AD hallmarks is the extracellular deposition in brain tissue of proteinaceous amyloid plaques, composed by well-ordered fibrillary aggregates of the amyloid β-peptide (Aβ). The Aβ aggregation process follows typical nucleation-polymerization kinetics, characterized by structu…

A new dopamine amino-acid conjugate: preclinical in vitro studies and evaluation of behavioural effects in rats
2012
It is well established that alterations in the functionality of dopaminergic transmission are associated with neurodegenerative diseases such as Parkinson’s Disease. The purpose of this study was to investigate the activity of a new dopamine amino-acid conjugate: L-phenylalanine-β-(3,4-dihydroxyphenyl) etilamide(DA-Phe) in the central nervous system, by assessing its influence on different behavioural parameters in the rat. Preliminarily, we tested the in vitro stability in plasmatic environment following the disappearance of DA-Phen from human plasma and the concurrent appearance of dopamine. Using rat brain homogenate, we also evaluated the level of DA-Phen cleavage by cerebral enzymes an…
Development of stimulus-sensitive electrospun membranes based on novel biodegradable segmented polyurethane as triggered delivery system for doxorubi…
2022
In this work, redox-sensitive polyurethane urea (PUU) based electrospun membranes have been exploited to chemically tether a pH-sensitive doxorubicin derivative achieved by linking a lipoyl hydrazide to the drug via a hydrazone linkage. First, the lipoyl-hydrazone-doxorubicin derivative labelled as LA-Hy-Doxo has been syn- thesized and characterized. Then, the molecule has been tethered, via a thiol-disulfide exchange reaction, to the redox-sensitive PUU (PolyCEGS) electrospun membrane. The redox-sensitive PolyCEGS PUU has been produced by using PCL-PEG-PCL polyol and glutathione-tetramethyl ester (GSSG-OMe)4 as a chain extender. The LA-Hy- Doxo tethered electrospun membrane has showed a du…
