Physico-chemical state influences in vitro release profile of curcumin from pectin beads
2014
International audience; Curcumin is a polyphenolic compound with diverse effects interesting to develop health benefit products but its formulation in functional foods or in food supplement is hampered by its poor water solubility and susceptibility to alkaline conditions, light, oxidation and heat. Encapsulation of curcumin could be a mean to overcome these difficulties. In this paper, curcumin was encapsulated by ionotropic gelation method in low methoxyl pectin beads associated with different surfactants: Solutol®, Transcutol® and sodium caseinate. After encapsulation, physico-chemical properties of encapsulated curcumin such as its solubility, physical state, tautomeric forms and encaps…
Mesomorphic and electrooptical properties of viologens based on non-symmetric alkyl/polyfluoroalkyl functionalization and on an oxadiazolyl-extended …
2019
Two different sets of ionic liquid crystals based on bistriflimide salts of non-symmetrically substituted polyfluorinated bipyridinium (viologens) and bent symmetrically substituted dialkyl-oxadiazolyl-bipyridinium have been synthesized, in order to study the effect on the mesomorphic and electrooptical properties of the non-symmetric functionalization (alkyl chain and fluoroalkyl chains of different lengths) on the two pyridinium rings and additionally the effect of a bent conjugated spacer among the two pyridinium units of the viologen. POM and DSC characterization show that the synthesized salts have a mesomorphic and, in some cases, polymesomorphic behaviour in a wide thermal range, als…
Wild-type Cu/Zn superoxide dismutase (SOD1) does not facilitate, but impedes the formation of protein aggregates of amyotrophic lateral sclerosis cau…
2009
Aggregation of Cu/Zn superoxide dismutase (SOD1) is a hallmark of a subset of familial amyotrophic lateral sclerosis (ALS) cases. The expression of wild-type SOD1 [SOD(hWT)] surprisingly exacerbates the phenotype of mutant SOD1 in vivo. Here we studied whether SOD1(hWT) may affect mutant SOD1 aggregation by employing fluorescence microscopy techniques combined with lifetime-based Förster resonance energy transfer (FRET). Only a very minor fraction of SOD1(hWT) was observed in aggregates induced by mutant SOD1(G37R), SOD1(G85R) or SOD1(G93C). Quite in contrast, co-expression of SOD(hWT) reduced the amount of mutant SOD1 in the aggregate fraction. Furthermore, we did not detect endogenous mou…
Synthesis and mesomorphism of related series of triphilic ionic liquid crystals based on 1,2,4-triazolium cations
2021
Abstract The synthesis, liquid crystal and conductivity properties of a series of 27 salts based on the 5-(4-(alkyloxy)phenyl)-1,2,4-triazol-4-ium cation bearing a perfluoroalkyl chain with triflate, tetrafluoroborate and bistriflimide anion are reported. The cations are regarded as triphilic on account of their three distinct regions – hydrocarbon, fluorocarbon and ionic. The mesophases were characterised by a combination of polarised optical microscopy, calorimetry and small-angle scattering experiments using both X-rays and neutrons, while thermal stability was probed using thermogravimetric analysis. The liquid crystal properties are found to be dependent on the anion and the length of …
Thermodiffusion anisotropy under a magnetic field in ionic liquid-based ferrofluids
2021
International audience; Ferrofluids based on maghemite nanoparticles (NPs), typically 10 nm in diameter, are dispersed in an ionic liquid (1-ethyl 3-methylimidazolium bistriflimide - EMIM-TFSI). The average interparticle interaction is found to be repulsive by small angle scattering of X-rays and of neutrons, with a second virial coefficient A2 = 7.3. A moderately concentrated sample at Φ = 5.95 vol% is probed by forced Rayleigh scattering under an applied magnetic field (up to H = 100 kA m-1) from room temperature up to T = 460 K. Irrespective of the values of H and T, the NPs in this study are always found to migrate towards the cold region. The in-field anisotropy of the mass diffusion c…
Highly tunable protein microspheres for drug delivery
2019
It is well-known that protein amyloid aggregation has profound implications in several neurodegenerative diseases. In contrast, a natural role for amyloid structures as protection, adhesion and storage materials in living system is also reported, promoting protein aggregates as an interesting platform for the design of multifunctional biomaterials. Among the broad range of different amyloid structures protein particulates deserve special attention; they are spherical protein aggregates with radius ranging from hundreds of nm to few um which are readily formed in solution at pHs values near the isoelectric point of the protein they are made of. Interestingly, particulate appears to be a gene…
TRANSPORTAN 10 INTERACTION WITH GIANT VESICLES: INSERTION EFFECTS AND PORE FORMATION
Transportan 10 (TP10) is a 21 residues peptide that belongs to the family of the antimicrobial and cytolytic or cytotoxic amphipathic peptides. It contains a high proportion of positively charged amino acids (four lysines), no negative charges and the N-terminus that impart it a formal +5 charge at neutral pH.1 This large number of positive charges is an essential feature for the electrostatic interaction of TP10 with microbial and tumoral membranes, which are characherized by a net negative charge and also by a higher fluidity if compared with mammalin ones.2 Here, combining spectroscopic and fluorescence lifetime imaging techniques, we analyse the fate of the multifunctional3-4 TP10 and i…
Peptide–Membrane Interactions Monitored by Fluorescence Lifetime Imaging: A Study Case of Transportan 10
2021
The interest on detailed analysis of peptide-membrane interactions is of great interest in both fundamental and applied sciences as these may relate to both functional and pathogenic events. Such interactions are highly dynamic and spatially heterogeneous, making the investigation of the associated phenomena highly complex. The specific properties of membranes and peptide structural details, together with environmental conditions, may determine different events at the membrane interface, which will drive the fate of the peptide-membrane system. Here, we use an experimental approach based on the combination of spectroscopy and fluorescence microscopy methods to characterize the interactions …
Phasor-FLIM for a direct investigation of Transportan 10 interactions with model membranes
2023
Transportan 10 (TP10), a short and positive charged peptide, belonging to the family of the cell penetrating peptides has gained increasing attention for its antimicrobial and anticancer activity but also for its applications in drug delivery as it is able to translocate therapeutic molecules in cellular environment. Due to the complexity of the phenomena involved in cellular uptake and following processes, which strongly depend on the membrane lipid composition, structural details of the peptide (e.g., charge, hydrophobicity, steric hindrance) and environmental conditions, it is not easy to understand the general rules governing them. Here, we combine spectroscopic techniques and fluoresce…
