The Translesion Polymerase Rev3L in the Tolerance of Alkylating Anticancer Drugs

Temozolomide and fotemustine, representing methylating and chloroethylating agents, respectively, are used in the treatment of glioma and malignant melanoma. Because chemoresistance of these tumors is a common phenomenon, identification of the underlying mechanisms is needed. Here we show that Rev3L, the catalytic subunit of the translesion DNA polymerase zeta, mediates resistance to both temozolomide and fotemustine. Rev3L knockout cells are hypersensitive to both agents. It is remarkable that cells heterozygous for Rev3L showed an intermediate sensitivity. ABSTRACT CTX-M β-lactamases, which show a high cefotaxime hydrolytic activity, constitute the most prevalent extended-spectrum β-lactamase (ESBL) type found among clinical isolates. The recent explosive diversification of CTX-M enzymes seems to have taken place due to the appearance of more efficient enzymes which are capable of hydrolyzing both cefotaxime and ceftazidime, especially among the CTX-M-1 cluster. Dehydroepiandrosterone sulphate (DHEAS) is a major adrenal secretory product, particularly in the fetus where it serves as a substrate for oestrogen biosynthesis by the placenta. The enzyme in the adrenal responsible for synthesising DHEAS, hydroxysteroid sulphotransferase (HST), is therefore essential for human development. We have isolated a full-length cDNA clone, encoding human fetal adrenal HST, and constructed a stable cell line expressing it by transfection into V79 Chinese hamster lung fibroblast cells. L’articolo riporta i primi risultati di uno studio finalizzato al monitoraggio delle deformazioni della cupola del Teatro Massimo di Palermo provocate dalle dilatazioni termiche, con l’impiego di tecniche topografiche e fotogrammetriche integrate. In particolare, sono state utilizzate due stazioni totali robotizzate e sistemi di fotogrammetria digitale di elevata precisione. Per correlare gli spostamenti dei carrelli sui quali poggia la struttura in acciaio della cupola con i gradienti termici, sono state acquisite immagini termiche diversificate. Progettare senza un obiettivo concreto, al di là dei costi di produzione, dei processi, dei materiali, e cioè al di là di una strategia aziendale o quanto meno di un’intenzione auto-produttiva (programmazione economica), non è davvero progettare; è al massimo un concept, tutto da verificare. Più interessante è passare dall’idea all’azione, per realizzare risultati concreti. Tetragonal phase of CuMnAs progressively appears as one of the key materials for antiferromagnetic spintronics due to efficient current-induced spin-orbit torques whose existence can be directly inferred from crystal symmetry. Theoretical understanding of spintronic phenomena in this material, however, relies on the detailed knowledge of electronic structure (band structure and corresponding wave functions) which has so far been tested only to a limited extent.
