Candida yeasts are saprophytes naturally present in the environment and forming colonies on human mucous membranes and skin. They are opportunistic fungi that cause severe and even fatal infections in immunocompromised individuals. Several essential oils, including eucalyptus, pine, cinnamon and lemon, have been shown to be effective against Candida strains. mRNA homoeostasis is favoured by crosstalk between transcription and degradation machineries. Both the Ccr4-Not and the Xrn1-decaysome complexes have been described to influence transcription. While Ccr4-Not has been shown to directly stimulate transcription elongation, the information available on how Xrn1 influences transcription is scarce and contradictory. In this study we have addressed this issue by mapping RNA polymerase II (RNA pol II) at high resolution, using CRAC and BioGRO-seq techniques in Saccharomyces cerevisiae. The transition of blastospores to the mycelial phase in Candida albicans was induced after the blastospores were kept at 4°C for several hours and then transferred to a fresh medium prewarmed at 37°C. Glucan was the most abundant polymer in the wall in the two morphogenetic forms but the amount of chitin was higher in the mycelial form than in blastospores. Efficient protoplasting required reducing agents and proteases together with β-glucanases (zymolyase). Protein synthesis in regenerating protoplasts was initiated after about 30 min. Monoclonal antibody 3H8: A useful tool in the diagnosis of candidiasis

In a previous series of experiments six mAbs were obtained against cell wall extracts of Candida albicans ATCC 26555. After several studies only one of them, designated 3H8, has been used to produce a commercial kit for the rapid diagnosis of candidiasis, Bichro-latex albicans (Fomouze Diagnostics). The present study involved the generation and characterization of this mAb as an immunoglobulin G1 which recognizes mannoproteins of high molecular mass present in the C. albicans cell wall. The 58-kDa surface mannoprotein of Candida albicans (mp58) elicits strong antibody responses during infection. Epitope mapping with sera from patients with candidiasis and control individuals indicated the presence of multiple IgG-reactive continuous epitopes on the protein, expanding both the amino- and carboxy-terminal domains and several internal regions. These immunoreactive regions were similar to the ones previously identified using sera from immunized animals. Non-wild-type cryptococcosis in a child with multivisceral organ transplant who owned bird pets.

Cryptococcosis is the third most common invasive fungal infection in solid-organ transplant recipients, although it is rarely reported in transplanted children. It typically appears as a late-onset infection with central nervous system or pulmonary involvement. We present a case of cryptococcosis caused by a non-wild strain in a 10-year-old child who owned two pet birds, and review the existent literature.
