Two-photon light-sheet microscopy for high-speed whole-brain functional imaging of zebrafish neuronal physiology and pathology

We present the development of a custom-made two-photon light-sheet microscope optimized for high-speed (5 Hz) volumetric imaging of zebrafish larval brain for the analysis of neuronal physiological and pathological activity. High-speed volumetric two-photon light-sheet microscopy is challenging to achieve, due to constrains on the signal-to-noise ratio. To maximize this parameter, we optimized our setup for high peak power of excitation light, while finely controlling its polarization, and we implemented remote scanning of the focal plane to record without disturbing the sample. Investigations of the emission properties of single CdS-nanocrystallites

Semiconductor nanocrystallites (NCs) with a radius in the range of the bulk exciton radius exhibit a strong quantum size effect. We investigated CdS-NCs, that were spin coated on a glass cover slip, with a low temperature confocal microscope. At low coverage we could image the fluorescence light from single NCs. The typical emission spectra of single NCs show four peaks. Besides the main peak related to near bandgap recombination, up to two LO-phonon satellites and one blue shifted peak were observed. A device for pH mapping derived from optical sensors similar to total internal reflection fluorescence microscopy (TIRFM) has been developed for future possible applications in the field of localized corrosion. The sensing principle is the increase of the fluorescent yield of the fluorescein with the pH of the medium. Single Molecule Studies of a Ladder Type Conjugated Polymer: Vibronic Spectra, Line Widths, and Energy Transfer

Confocal fluorescence microscopy and spectroscopy are employed to investigate single poly(ladder-type pentaphenylene) (LPPentP) molecules dispersed in thin poly(methyl methacrylate) (PMMA) films at 1.2 K. Emission spectra of single chains show single as well as multi-chromophore emission indicating variegated communication along the chains. The vibronic structure in the emission spectra resembles the one found for other ladder-type polymers. Purely electronic zero-phonon lines in emission are substantially broadened, most probably due to fast spectral diffusion. Confocal technology in fluorescence in situ hybridization evaluation for cancer: a diagnostic improvement.

During the cancer routine diagnosis course, we commonly use fluorescence in situ hybridization (FISH) technique. FISH studies are conducted for genes amplification analyses (ErBb2/Neu) and also for genes translocation studies such as CMYC, BCL6, or BCL2. All-organic fast intersystem crossing assisted exciplexes exhibiting sub-microsecond thermally activated delayed fluorescence

A novel strategy is presented towards acquisition of exciplex systems exhibiting thermally activated delayed fluorescence (TADF) with a high reverse intersystem crossing (RISC) rate (exceeding 107 s−1). This approach involves constructing exciplex donor–acceptor molecular pairs, where the acceptor molecule possesses the ability to undergo fast and efficient intersystem crossing (ISC). We demonstrate the construction of wavelength λ-ratiometric images that allow visualizing the distribution of microscopic dynamics within living cells and tissues by using the newly developed principle of fluorescence response. The bent-to-planar motion in the excited state of incorporated fluorescence probes leads to elongation of the π-delocalization, resulting in microviscosity-dependent but polarity-insensitive interplay between well-separated blue and red bands in emission spectra. Isoindolo[2,1-a]quinoxaline derivatives, novel potent antitumor agents with dual inhibition of tubulin polymerization and topoisomerase I.

Isoindoloquinoxalines 4 and 5 were obtained by refluxing 2-(2'-aminoaryl)-1-cyanoisoindoles 3a- e in acetic or formic acid. All derivatives were screened by the National Cancer Institute (Bethesda, MD) for the in vitro one dose primary anticancer assay against a 3-cell line panel. Compounds 4a- e, screened against a panel of about 60 human tumor cell lines, showed remarkable antineoplastic activity; they had GI 50 values in the low micromolar or submicromolar range and reached, in the case of 4c, nanomolar concentrations on 88% of the 59 tested cell lines.
