Simultaneous enzymatic determination of l(−) malic acid and l(+) lactic acid in wine by flow injection analysis
1991
Abstract A simultaneous determination of two organic acid species in wine by a flow injection analysis method using enzyme-immobilized open tubular reactors is described. The design of the optimal geometry of the FIA system is especially important to avoid the matrix fluorescence. Reproducible results were obtained in the determination of l (−) malic acid and l (+) lactic acid using l -MDH and l -LDH, respectively. The assay reactions utilize the fluorimetric determination of a common product, NADH. The sample was dialyzed on line and injected into the FIA-system. The system responds linearly (log-log calibration graphs) to the injected sample (80 μl) in concentrations ranging from 0·1 m m …
Evaluation of the interelemental effect in X-ray fluorescence analysis by the total addition method
1992
An algorithm for quantifying interelemental effects in X-ray fluorescence techniques is developed. By applying an addition process, the ratio between the mass absorption coefficients of the analyte and the unknown sample (μi*/μs*) is calculated to correct the fluorescence intensity of the element to be determined and linearize the I-c calibration plot. This coefficient can be calculated graphically and numerically. The method is applied to the determination of tin in lead alloys with good results over wide concentration ranges.
Chemical composition and roughness of enamel and composite after bleaching, acidic beverages and toothbrushing
2019
Background In this study was assessed the surface roughness and chemical composition of tooth enamel and composite resin after bleaching treatment, immersion in acidic beverages, and simulated toothbrushing. Material and methods One hundred and twenty dental blocks (10 x 10 x 3 mm) were randomly assigned (n = 10) according to surface treatment [none (N), bleaching (B), toothbrushing (T), and B+T] and storage medium [saliva (S), whiskey (W), and orange juice (O)]: experimental groups - N+S, N+W, N+O, B+S, B+W, B+O, S+T, W+T, O+T, B+S+T, B+W+T, and B+O+T. Two bleaching sessions were conducted using 38% hydrogen peroxide (3 applications). Surface roughness was measured using a roughness tester…
Determination of cobalt, copper, iron, nickel and zinc in cemented tungsten carbides with cobalt as a binder by FAAS: Matrix effect control by multiv…
1997
A new approach for the determination of cobalt, copper, iron, nickel and zinc in cemented tungsten carbides with cobalt as a binder by flame atomic absorption spectrophotometry (FAAS) is reported. Real samples were dissolved in phosphoric, hydrochloric and nitric acid. PTFE bomb or alternatively small amounts of HF were used for the enhancement of the recovery of the elements investigated. Synthetic samples were used for interference studies. Multiple linear regression was applied for the control of matrix effects and it proved to be very effective in the search for interfering elements. Using simple acid based standards, all investigated elements could be determined sequentially in a compl…
Novel leaf-level measurements of chlorophyll fluorescence for photosynthetic efficiency
2015
Solar induced chlorophyll fluorescence (SIF) from vegetation can now be obtained from satellites as well as ground-based field studies, at select wavelengths associated with atmospheric features. At the leaf level, full spectrum (650–800 nm) chlorophyll emissions (ChlF) can be measured using specialized instrumentation to support interpretation of these SIF observations. We found that ChlF spectra differ for leaf bottoms versus upper leaf surfaces, potentially affecting within-canopy radiative scattering. Our ChlF measurements for leaves of eight tree species (n≥125) obtained during fall 2013 senescence at the Duke Forest in North Carolina, USA and the 2014 growing season (n=72) at the USDA…
Design of molecular imaging and theranostic agents for the detection and targeted therapy of cancers
2020
Despite significant advances in cancer detection and treatment, cancer is now the second leading cause of death worldwide. The aim of this thesis project was to design and optimize imaging agents for the diagnosis and/or therapy of various cancers. The first axis of this thesis focused on the development of a bimodal PET/fluorescence imaging agent capable of targeting neurotensin NTS1 receptors overexpressed in certain cancers. PET imaging would allow efficient detection of tumors and their metastases, while fluorescence imaging would facilitate the evaluation of tumor margins during surgery. Several compounds were synthesized, labelled with gallium-68 and studied in preclinical studies in a…
Probing ensemble polymorphism and single aggregate structural heterogeneity in insulin amyloid self-assembly.
2020
Ensembles of protein aggregates are characterized by a nano- and micro-scale heterogeneity of the species. This diversity translates into a variety of effects that protein aggregates may have in biological systems, both in connection to neurodegenerative diseases and immunogenic risk of protein drug products. Moreover, this naturally occurring variety offers unique opportunities in the field of protein-based biomaterials. In the above-mentioned fields, the isolation and structural analysis of the different amyloid types within the same ensemble remain a priority, still representing a significant experimental challenge. Here we address such complexity in the case of insulin for its relevance…
Phasor-FLIM analysis of Thioflavin T self-quenching in Concanavalin amyloid fibrils
2020
The formation of amyloid structures has traditionally been related to human neurodegenerative pathologies and, in recent years, the interest in these highly stable nanostructures was extended to biomaterial sciences. A common method to monitor amyloid growth is the analysis of Thioflavin T fluorescence. The use of this highly selective dye, diffused worldwide, allows mechanistic studies of supramolecular assemblies also giving back important insight on the structure of these aggregates. Here we present experimental evidence of self-quenching effect of Thioflavin T in presence of amyloid fibrils. A significant reduction of fluorescence lifetime of this dye which is not related to the propert…
Evaluation of near-Infrared fluorescence-conjugated peptides for visualization of human epidermal receptor 2-overexpressed gastric cancer.
2021
338 Background: HER2 is highly overexpressed in many kinds of cancers with a poor prognosis. Recently, near-infrared (NIR) fluorescence-based imaging is a growing field for both pre-clinical and clinical application. In this study, we aimed to synthesize Human Epidermal Receptor2 (HER2)-specific near-infrared (NIR) fluorescence probes and evaluate their applicability in cancer-specific image-guided surgeries using an animal model. Methods: An NIR dye emitting light of 800 nm (IRDye800CW, Li-COR, USA) was conjugated to trastuzumab and HER2-specific affibody using click mechanism. HER2 affinity was assessed by the surface plasmon resonance technique. HER2 positive/negative gastric cancer cel…
