Stability and stoichiometry of some binary fluorophore-cyclodextrin complexes

The stability and stoichiometric ratio of binary complexes among five fluorophores and β-cyclodextrin (β-CD) or heptakis-(6-amino-6-deoxy)-β-cyclodextrin (am-β-CD) were determined by means of fluorescence measurements in borate buffer at pH=8.0 and 9.0. Structure of both host and guest affected the characteristics of the binary complexes. Pyrene and anthraquinone formed a 1:2 (fluorophore: cyclodextrin) complex with both cyclodextrins. Xanthone formed 1:1 complex with β-CD and 1:2 complex with am-β-CD. A more defined behaviour was observed for crysene. In fact, both stoichiometric different complexes were detected with both hosts. In the current study were used excitation-emission matrices (EEMs) and synchronous fluorescence spectroscopy (SFS) steady-state techniques in a broad spectral regions (excitation at 220-500 nm and emission at 280-850 nm) to achieve the whole set of endogenous fluorophores, existed in normal and neoplastic cutaneous tissues. Several types of benign, dysplastic and malignant types of skin lesions were investigated ex vivo using both EEM and SFS modalities, namely the basal cell papilloma and carcinoma, pigmented nevi, dysplastic nevi, squamous cell carcinoma and malignant melanoma. Specific labelling via bioorthogonal tetrazine chemistry confers minimal label size and rapid labelling kinetics. A new fluorescent terpyridyl-diphenylacetylene hybrid fluorophore 4'-[4-{(4-methoxyphenyl)ethynyl}phenyl]-2,2':6',2''-terpyridine, L, was synthesized via Sonogashira cross-coupling of 4'-(4-bromophenyl)-2,2':6',2''-terpyridine and 4-ethynylanisole in the presence of Pd(PPh3)4/CuI as a catalyst. The solid state structure of L shows a trans arrangement of pyridine nitrogen atoms along the interannular bond in the terpyridine domain. The emission behaviour of the 4,4′-bis(dimethylamino)biphenyl subunit covalently attached to aza-crown ethers is studied. Some new ligands have been synthesised in order to test the properties of this new fluorophore. The fluorescence of these new ligands and some other compounds previously described has been studied in acetonitrile in the presence of Ni2+, Cu2+, Zn2+, Hg2+, Pb2+, Cd2+ and also in the presence of some alkali and alkaline-earth cations.
An Off-On-Off Fluorescent Sensor for pH Windows Based on the 13aneN4-Zn 2+ System

International audience; The new ligand L was prepared and features a 13-membered tetraaza macrocyclic ring with a 1,8-naphthalimide fluorophore appended to a C atom of its backbone. The protonation constants of L as well as its complexation constants with Zn2+ ions were determined in 1:1 water/methanol solutions by potentiometric titrations. Fluorimetric pH titrations were performed with L alone and L in the presence of Zn2+ ions (1:1), and the species distributions (%) versus pH were compared. A window-shaped fluorescence trend was observed with pH for the L/Zn2+ system, which behaves as an off-on-off pH sensor. Poikkitieteelliset sovellukset ovat viime aikoina yleistyneet tieteellisessä tutkimuksessa. Tämä näkyy hyvin esimerkiksi elävän kudoksen kuvantamisen kehittymisessä, jota varten etsitään jatkuvasti parempia biologisia leimoja fysiikan ja kemian keinoin. Tässä tutkielmassa on kehitetty ensimmäinen lähi-infrapuna-alueen fluoresenssileima, joka hyödyntää hopeananopartikkeleja ja fytokromeja. Kompleksi on syntetisoitu itse ja sen rakenne on todennettu optisen spektroskopian, elektroforeesin ja elektronimikroskopian keinoin.
