Experimental calibration of flat jacks for in-situ testing of masonry
2018
Flat-jack testing method is one of the most commonly used techniques for the structural assessment of existing masonry structures. Single and double flat jacks are usually adopted to evaluate the acting normal stress, or the compressive behaviour of masonry material. Test procedures are codified by international standards (e.g., A.S.T.M D4729-87; C1196-04; C1197-04, R.I.L.E.M TC 177–MDT D.4; R.I.L.E.M. TC 177–MDT D.5), which provide the preliminary calibration of an experimental coefficient (km) and of the effective area (Aeff), which determination influences significantly the reliability of the test. This article presents the result of an experimental study on the calibration of flat jacks…
Total RNA-isolation of abdominal hernia of rats for quantitative real-time reverse transcription (RT) PCR assays.
2007
Abstract Increasing complications in incisional hernia surgery call for novel treatments. A gene expression analysis of injured tissues displays important parameters for tissue regeneration. Until today, no reliable method has been described for a quantitative gene expression analysis of hernia tissues. In this work, a protocol is described for the isolation of DNA‐free total RNA of incisional hernias for the first time. Moreover, real‐time RT PCR assays for collagen type I and III and TGF‐β1 are demonstrated for relative gene expression analyses. Both methods enable relative gene expression analyses of hernia tissues for the first time.
Energy efficiency analysis in Incremental Sheet Forming operations
2012
In literature, manufacturing processes have been studied with different approaches, but a lack in terms of their environmental impact and energy required is still present. The aim of this paper is to compute some energy efficiency analysis on a specific manufacturing process. More in detail, the attention will be focused on the Incremental Sheet Forming process in order to evaluate the required energy and how it changes for changing process conditions. Accordingly, a wide experimental investigation has been executed taking into account different process parameters (i.e. tool depth step, punch diameter, wall inclination angle and sheet thickness). The total energy consumption has been monito…
Improving formability in SPIF processes through high speed rotating tool: experimental and numerical analysis
2013
Single-point incremental forming (SPIF) is a quite new sheet-forming process which offers the possibility to deform complex parts without dedicated dies using a single-point tool and a standard three-axis CNC machine. Although the process mechanics enables higher strains with respect to traditional sheet-forming processes, research has been focused on further increasing the maximum forming angle. In the paper, a new approach is used to enhance the material formability through a localized sheet heating as a consequence of the friction work caused by high speed rotating tool. Numerical simulation was utilized to relate the effect of temperature with the main field variables distribution in th…

Self‐Directed Learning in Creative Activity : An Ethnographic Study in Technology‐Based Work
2020
Under conditions of rapid changes in working life, there is an urgent need to examine the nature of creativity and learning in organizations. The aim of this study was to investigate the nature of self‐directed learning (SDL) practices in creative activity in technology‐based work. We focus on both individual and collective practices but also on the importance of organizational culture. The data consist of 46 interviews and observational field notes collected from participating organizations. Thematic and ethnographic analyses were utilized as tools to reveal the nature of SDL in creative activity. We found three themes describing of the nature of SDL in creative activity: a combination of …
Geldanamycin and its derivatives as Hsp90 inhibitors
2012
The Hsp90 molecule, one of the most abundant heat shock proteins in mammalian cells, maintains homeostasis and prevents stress-induced cellular damage. Hsp90 is expressed under normal conditions at a level of about 1-2 Percent of total proteins, while its expression increases 2-10 fold in cancer cells. The two main constitutively expressed isoforms of Hsp90 are known as Hsp90-alpha and Hsp90-beta, and their upregulation is associated with tumor progression, invasion and formation of metastases, as well as development of drug resistance. The Hsp90 is a key target for many newly established, potent anticancer agents containing Hsp90 N-terminal ATP binding inhibitors, such as geldanamycin, and…
Cigarette smoke promotes inflammasome‐independent activation of caspase‐1 and ‐4 leading to gasdermin D cleavage in human macrophages
2022
Mechanisms and consequences of gasdermin D (GSDMD) activation in cigarette smoke (CS)-associated inflammation and lung disease are unknown. GSDMD is a downstream effector of caspase-1, -8, and -4. Upon cleavage, GSDMD generates pores into cell membranes. Different degrees of GSDMD activation are associated with a range of physiological outputs ranging from cell hyperactivation to pyroptosis. We have previously reported that in human monocyte-derived macrophages CS extract (CSE) inhibits the NLRP3 inflammasome and shifts the response to lipopolysaccharide (LPS) towards the TLR4-TRIF axis leading to activation of caspase-8, which, in turn, activates caspase-1. In the present work, we investig…
