Stability of tocopherols in adapted milk-based infant formulas during storage

Abstract An evaluation is made of the effects of added α -tocopherol ( α -T) or α -tocopherol acetate ( α -TAc) combined with iron lactate or sulfate, storage time (up to 17 months) and storage temperature (22°C and 37°C) on the α -TAc, α -T, γ -tocopherol ( γ -T) and δ -tocopherol ( δ -T) content of infant formulas. Tocopherols were measured by normal phase HPLC and fluorescence detection after direct extraction, which allows the determination of both α -TAc and α -T. The α -TAc content was not affected by temperature or storage time, although the α -, γ - and δ -T contents, which were lower at 37°C than at 22°C, were significantly affected. Pharmacologically active macromolecules, such as peptides, are still a major challenge in terms of designing a delivery system for their transport across absorption barriers and at the same time provide sufficiently high long-term stability. Spray freeze dried (SFD) lyospheres® are proposed here as an alternative for the preparation of fast dissolving porous particles for nasal administration of insulin. Abstract Nitrite is prepared in situ for the determination of sulfadiazine. The method is based on solid-phase reduction of copperized cadmium of nitrate; the nitrite reagent merges with the sample stream in hydrochloric acid medium and the resulting mixture is injected into the water carrier, pure distilled water, and then merges with the N-(1-naphthyl)ethylendiamine reagent and is measured spectrophotometrically at 542.0 nm. The calibration graph is linear over the range 0.5–50 μg ml−1 sulfadiazine, mid-range R.S.D. = 0.3% (n = 5) and sample throughput 72 h−1. Casein phosphopeptides released by simulated gastrointestinal digestion of infant formulas and their potential role in mineral binding

Abstract Adapted and follow-up milk-based infant formulas were subjected to gastrointestinal digestion simulating physiological conditions. The naturally occurring casein phosphopeptides (CPPs) generated were fractionated by anion exchange high-performance liquid chromatography and sequenced by tandem mass spectrometry. In both infant formula digests, a total of 19 CCPs from bovine casein were identified, of which 7 corresponded to α s1 -casein and 12 to α s2 -casein. Most CPPs had the cluster sequence SpSpSpEE, representing the binding sites for minerals. A flow injection procedure for the indirect chemiluminescent determination of isoniazid is proposed. The method is performed in a flow-injection manifold provided with a solid-phase reactor. The reactor was made from manganese dioxide physically entrapped by polymerization; the redox reaction isoniazid-manganese dioxide released Mn(II) which was monitored through its inhibitory effect on the reaction between luminol and hydrogen peroxide in presence of potassium hexacyanoferrate (III). Abstract This article deals with the simultaneous determination of dissolution profiles of two drugs with overlapped spectra, present in the same pharmaceutical formulation. The official procedure for the dissolution profile is adapted to the continuous-flow methodology; the dissolution vessel is connected to an FIA manifold, in which the sample aliquots from the dissolution vessel are treated in order to adjust to the suitable pH and dilution degree to be monitored. Comparison of spectrophotometric and HPLC methods for determining sialic acid in infant formulas

Abstract Two methods for determining sialic acid in infant formulas – spectrophotometry and HPLC with fluorescence detection – have been optimised and validated, the first one allows to determine total sialic acid while the second allows to differentiate the two main forms of sialic acid (N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (Neu5Gc)). A common sample preparation procedure (hydrolysis and purification) for both methods has been proposed.
