T Cell Polarization toward T
2016
IgG4-related disease (IgG4-RD) is a fibro-inflammatory disorder involving virtually every organ with a risk of organ dysfunction. Despite recent studies regarding B cell and T cell compartments, the disease’s pathophysiology remains poorly understood. We examined and characterized subsets of circulating lymphocytes in untreated patients with active IgG4-RD. Twenty-eight consecutive patients with biopsy-proven IgG4-RD were included in a prospective, multicentric study. Lymphocytes’ subsets were analyzed by flow cytometry, with analysis of TH1/TH2/TH17, TFH cells, and cytokine release by peripheral blood mononuclear cells. Results were compared to healthy controls and to patients with primary…

Mutation analysis of the SPG4 gene in Italian patients with pure and complicated forms of spastic paraplegia
2010
Mutations in the SPG4 gene are the most common causes of hereditary spastic paraplegia (HSP) accounting for up to 40% of autosomal dominant (AD) forms and 12-18% of sporadic cases. The phenotype associated with HSP due to mutations in the SPG4 gene tends to be pure. There is increasing evidence, however, of patients with complicated forms of spastic paraplegia in which SPG4 mutations were identified. A cohort of 38 unrelated Italian patients with spastic paraplegia, of which 24 had a clear dominant inheritance and 14 were apparently sporadic, were screened for mutations in the SPG4 gene.We identified 11 different mutations, six of which were novel (p.Glu143GlyfsX8, p.Tyr415X, p.Asp548Asn, c…
Immunoglobulin G4-Related Aortitis and Severe Aortic Valve Stenosis Treated With Transcatheter Aortic Valve Replacement and Immunosuppression.
2021
Abstract IgG4-related disease is an immune-mediated fibro-inflammatory disorder with multisystemic involvement. Aortitis and peri-aortitis are the most common cardiovascular manifestations of the disease. We present the case of a 65-year-old man with symptomatic severe aortic stenosis and concomitant IgG4 aortitis. The diagnosis was confirmed by IgG4 serum levels, positive positron emission computed tomography (PET-CT) scans, and pathology from mediastinal dissection. Surgical aortic valve replacement (SAVR) was unfeasible owing to extensive mediastinal fibrosis, and transcatheter aortic valve replacement (TAVR) was successfully performed. As ascending aorta access for SAVR in IgG4 aortitis…
Evolution of melanoma cross-resistance to CD8⁺ T cells and MAPK inhibition in the course of BRAFi treatment
2018
The profound but frequently transient clinical responses to BRAFV600 inhibitor (BRAFi) treatment in melanoma emphasize the need for combinatorial therapies. Multiple clinical trials combining BRAFi and immunotherapy are under way to further enhance therapeutic responses. However, to which extent BRAFV600 inhibition may affect melanoma immunogenicity over time remains largely unknown. To support the development of an optimal treatment protocol, we studied the impact of prolonged BRAFi exposure on the recognition of melanoma cells by T cells in different patient models. We demonstrate that autologous CD8+ tumor-infiltrating lymphocytes (TILs) efficiently recognized short-term (3, 7 days) BRAF…
Fundamental isomorphism theorems for quantum groups
2017
The lattice of subgroups of a group is the subject of numerous results revolving around the central theme of decomposing the group into "chunks" (subquotients) that can then be compared to one another in various ways. Examples of results in this class would be the Noether isomorphism theorems, Zassenhaus' butterfly lemma, the Schreier refinement theorem for subnormal series of subgroups, the Dedekind modularity law, and last but not least the Jordan-H\"older theorem. We discuss analogues of the above-mentioned results in the context of locally compact quantum groups and linearly reductive quantum groups. The nature of the two cases is different: the former is operator algebraic and the latt…

Unveiling the spatial structure of the overionized plasma in the supernova remnant W49B
2011
W49B is a mixed-morphology supernova remnant with thermal X-ray emission dominated by the ejecta. In this remnant, the presence of overionized plasma has been directly established, with information about its spatial structure. However, the physical origin of the overionized plasma in W49B has not yet been understood. We investigate this intriguing issue through a 2D hydrodynamic model that takes into account, for the first time, the mixing of ejecta with the inhomogeneous circumstellar and interstellar medium, the thermal conduction, the radiative losses from optically thin plasma, and the deviations from equilibrium of ionization induced by plasma dynamics. The model was set up on the basi…
On the regularity of critical and minimal sets of a free interface problem
2015
We study a free interface problem of finding the optimal energy configuration for mixtures of two conducting materials with an additional perimeter penalization of the interface. We employ the regularity theory of linear elliptic equations to study the possible opening angles of Taylor cones and to give a different proof of a partial regularity result by Fan Hua Lin [Calc Var. Partial Differential Equations, 1993].
