T Cell Polarization toward T
2016
IgG4-related disease (IgG4-RD) is a fibro-inflammatory disorder involving virtually every organ with a risk of organ dysfunction. Despite recent studies regarding B cell and T cell compartments, the disease’s pathophysiology remains poorly understood. We examined and characterized subsets of circulating lymphocytes in untreated patients with active IgG4-RD. Twenty-eight consecutive patients with biopsy-proven IgG4-RD were included in a prospective, multicentric study. Lymphocytes’ subsets were analyzed by flow cytometry, with analysis of TH1/TH2/TH17, TFH cells, and cytokine release by peripheral blood mononuclear cells. Results were compared to healthy controls and to patients with primary…
Identifying and validating the presence of guanine-quadruplexes (G4) within the blood fluke parasite schistosoma mansoni
2021
Schistosomiasis is a neglected tropical disease that currently affects over 250 million individuals worldwide. In the absence of an immunoprophylactic vaccine and the recognition that mono-chemotherapeutic control of schistosomiasis by praziquantel has limitations, new strategies for managing disease burden are urgently needed. A better understanding of schistosome biology could identify previously undocumented areas suitable for the development of novel interventions. Here, for the first time, we detail the presence of G-quadruplexes (G4) and putative quadruplex forming sequences (PQS) within the Schistosoma mansoni genome. We find that G4 are present in both intragenic and intergenic regi…
OPTIMIZATIONS FOR TENSORIAL BERNSTEIN–BASED SOLVERS BY USING POLYHEDRAL BOUNDS
2010
The tensorial Bernstein basis for multivariate polynomials in n variables has a number 3n of functions for degree 2. Consequently, computing the representation of a multivariate polynomial in the tensorial Bernstein basis is an exponential time algorithm, which makes tensorial Bernstein-based solvers impractical for systems with more than n = 6 or 7 variables. This article describes a polytope (Bernstein polytope) with a number of faces, which allows to bound a sparse, multivariate polynomial expressed in the canonical basis by solving several linear programming problems. We compare the performance of a subdivision solver using domain reductions by linear programming with a solver using a c…
Exact simulation of diffusion first exit times: algorithm acceleration
2020
In order to describe or estimate different quantities related to a specific random variable, it is of prime interest to numerically generate such a variate. In specific situations, the exact generation of random variables might be either momentarily unavailable or too expensive in terms of computation time. It therefore needs to be replaced by an approximation procedure. As was previously the case, the ambitious exact simulation of exit times for diffusion processes was unreachable though it concerns many applications in different fields like mathematical finance, neuroscience or reliability. The usual way to describe exit times was to use discretization schemes, that are of course approxim…
Case Report: Association of a Variant of Unknown Significance in the FIG4 Gene With Frontotemporal Dementia and Slowly Progressing Motoneuron Disease…
2020
BackgroundModern genetics have in many ways revolutionized clinical routine and have, for instance, shown that formerly distinct disease entities relate to common pathogenic mutations. One such example is the connection between dementia and amyotrophic lateral sclerosis (ALS) in a continuous disease spectrum affirmed by the discovery of shared mutations.Case ReportWe describe a new variant in the FIG4 gene in a patient with slowly progressing frontotemporal dementia (FTD) and probable primary lateral sclerosis (PLS). The patient initially showed depressive symptoms and global cognitive deficits. Severe difficulties with language and hallucinations became clearer as the disease progressed. N…
Pārtikas alergēnu kvantitatīva izpēte atopiskā dermatīta pacientiem pieaugušo vecumā
2015
Evolution of melanoma cross-resistance to CD8⁺ T cells and MAPK inhibition in the course of BRAFi treatment
2018
The profound but frequently transient clinical responses to BRAFV600 inhibitor (BRAFi) treatment in melanoma emphasize the need for combinatorial therapies. Multiple clinical trials combining BRAFi and immunotherapy are under way to further enhance therapeutic responses. However, to which extent BRAFV600 inhibition may affect melanoma immunogenicity over time remains largely unknown. To support the development of an optimal treatment protocol, we studied the impact of prolonged BRAFi exposure on the recognition of melanoma cells by T cells in different patient models. We demonstrate that autologous CD8+ tumor-infiltrating lymphocytes (TILs) efficiently recognized short-term (3, 7 days) BRAF…
On Apache Log4j2 Exploitation in Aeronautical, Maritime, and Aerospace Communication
2022
Apache Log4j2 is a prevalent logging library for Java-based applications. In December 2021, several critical and high-impact software vulnerabilities, including CVE-2021-44228, were publicly disclosed, enabling remote code execution (RCE) and denial of service (DoS) attacks. To date, these vulnerabilities are considered critical and the consequences of their disclosure far-reaching. The vulnerabilities potentially affect a wide range of internet of things (IoT) devices, embedded devices, critical infrastructure (CI), and cyber-physical systems (CPSs). In this paper, we study the effects and feasibility of exploiting these vulnerabilities in mission-critical aviation and maritime environment…
Progressive multi-organ expression of immunoglobulin G4-related disease: A case report.
2012
A 63-year-old Caucasian man presented with a cholestatic syndrome, renal failure and arthralgias. A laboratory examination revealed high immunoglobulin G (IgG) and IgG4 levels (5.95 g/L; normal range: 0.08-1.4 g/L), pointing to a diagnosis of systemic IgG4-related disease, with definite radiological evidence of biliary and pancreatic expression, and plausible renal, articular, salivary and lacrimal glands involvement. Due to the rarity of the condition, there are currently no random control trials to point to the optimal therapeutic approach. The patient has been on steroid therapy with the subsequent introduction of azathioprine, with a complete resolution of all symptoms, a rapid reductio…
The Right to Education and ICT during COVID-19: An International Perspective
2020
There is a lack of concluding evidence among epidemiologists and public health specialists about how school closures reduce the spread of COVID-19. Herein, we attend to the generalization of this action throughout the world, specifically in its quest to reduce mortality and avoid infections. Considering the impact on the right to education from a global perspective, this article discusses how COVID-19 has exacerbated inequalities and pre-existing problems in education systems around the world. Therefore, the institutional responses to guaranteeing remote continuity of the teaching&ndash
