Molecular dynamics of electrosprayed water nanodroplets containing sodium bis(2-ethylhexyl)sulfosuccinate

2013

The behavior of aqueous solutions of sodium bis(2-ethylhexyl)sulfosuccinate (AOTNa) subject to electrospray ionization (ESI) has been investigated by molecular dynamics (MD) simulations at three temperatures (350, 500 and 800 K). We consider several types of water nanodroplets containing AOTNa molecules and composed of a fixed number of water molecules (1000), N(AOT)(0) AOT(-) anions (N(AOT)(0) =  0, 5, 10) and N(Na)(0) sodium ions (N(Na)(0) =  0, 5, 10, 15, 20): in a short time scale (less than 1 ns), the AOTNa molecules, initially forming direct micelles in the interior of the water nanodroplets, are observed in all cases to diffuse nearby the nanodroplet surface, so that the hydrophilic …

Sigma-1 Receptor Activation Induces Autophagy and Increases Proteostasis Capacity In Vitro and In Vivo

2019

Dysfunction of autophagy and disturbed protein homeostasis are linked to the pathogenesis of human neurodegenerative diseases and the modulation of autophagy as the protein clearance process has become one key pharmacological target. Due to the role of sigma-1 receptors (Sig-1R) in learning and memory, and the described pleiotropic neuroprotective effects in various experimental paradigms, Sig-1R activation is recognized as one potential approach for prevention and therapy of neurodegeneration and, interestingly, in amyotrophic lateral sclerosis associated with mutated Sig-1R, autophagy is disturbed. Here we analyzed the effects of tetrahydro-N,N-dimethyl-2,2-diphenyl-3-furanmethanamine hyd…

Chemical Strategies for the Synthesis of Protein–Polymer Conjugates

2012

Protein-polymer conjugates have achieved tremendous attention in the last few years. The synergistic combination of properties has led to certain advantages in bio-applications. Over the past few years, numerous chemical strategies have been developed to conjugate different synthetic polymers onto proteins, most of which can be summarized within the scope of click-chemistry. Here we highlight conjugation strategies based on available functional groups present on the synthetic polymer and existing groups of proteins from the natural pool. In particular, the chapter organizes the various possible reactions by classes of functional groups present on protein surfaces, deriving from selected ami…

Long-term cropping systems and tillage management effects on soil organic carbon stocks and steady state level of C sequestration rates in a semiarid…

2010

A calcareous and clayey xeric Chromic Haploxerept of a long-term experimental site in Sicily (Italy) was sampled (0–15 cm depth) under different land use management and cropping systems (CSs) to study their effect on soil aggregate stability and organic carbon (SOC). The experimental site had three tillage managements (no till [NT], dual-layer [DL] and conventional tillage [CT]) and two CSs (durum wheat monocropping [W] and durum wheat/faba bean rotation [WB]). The annually sequestered SOC with W was 2·75-times higher than with WB. SOC concentrations were also higher. Both NT and CT management systems were the most effective in SOC sequestration whereas with DL system no C was sequestered. …

Chemical conjugation of dexamethasone to a polyaspartamide and in vitro evaluation studies

2004

Two macromolecular conjugates of dexamethasone containing different drug amounts were synthesized using PHEA as the polymeric carrier and a succinic group as spacer. The content of linked drug was equal to 25.3% w/w (conjugate A) and 12.7% w/w (conjugate B). Both polymeric conjugates, unlike the free drug, were water-soluble and the amount of unlinked drug was evaluated to be approximately about 0.01% w/w. Both conjugates were relatively stable in vitro at pH 7.4 whereas in the presence of esterase only the conjugate B was able to release drug under the used experimental conditions. This dissimilar behavior has been attributed to the distinct macromolecular conformations assumed in aqueous …

Glycoconjugate expression and cartilage development of the cranial skeleton.

1998

Only few detailed investigations have focused on the glycobiology of cranial development. The functional elements in most inductive and morphogenetic processes are not individual cells, but rather collectives of interacting populations and extracellular matrix components that give rise to specific tissues and organs. Experimental evidence strongly suggests that sugar chains not only confer morphological characteristics. Complex carbohydrate molecules and their corresponding receptors are involved in recognition processes decoding biological information during cranial morphogenesis. The distribution patterns of glycoconjugates are highly dynamic and show a clear correlation with characterist…
