Translation of HTT mRNA with expanded CAG repeats is regulated by the MID1-PP2A protein complex.

Expansion of CAG repeats is a common feature of various neurodegenerative disorders, including Huntington's disease. Here we show that expanded CAG repeats bind to a translation regulatory protein complex containing MID1, protein phosphatase 2A and 40S ribosomal S6 kinase. Binding of the MID1-protein phosphatase 2A protein complex increases with CAG repeat size and stimulates translation of the CAG repeat expansion containing messenger RNA in a MID1-, protein phosphatase 2A- and mammalian target of rapamycin-dependent manner. Primary cultures of brain-derived rodent cells are widely used to study molecular and cellular mechanisms in neurobiology. In this chapter, we describe methods of purifying and culturing oligodendroglial cells from mouse perinatal brains. In addition, we describe methods of coculturing the purified oligodendrocytes with neurons. When prepared and cultured according to these protocols, many essential aspects of the biology of oligodendrocytes, such as their proliferation, differentiation, and myelination, can be studied in culture.

Concentrations of metallothioneins (MT) were analysed in bivalve molluscs Anodonta spp. and Unio spp. from 6 lakes and watercourses of Latvia with different anthropogenic pressure. The aim of the study was to find the relationship between MT in molluscs and concentrations of heavy metals in sediments. Unio spp. and Anodonta spp. showed no significant differences in the concentrations of MT. High concentrations of heavy metals (Cd, Pb, Hg, Cu, Zn) were detected in the deepest parts of polluted water bodies, however their concentrations significantly decreased in near shore sediments inhabited by molluscs. L’ernia diaframmatica congenita (EDC) è una malformazione caratterizzata da un difetto di formazione del diaframma che rimane incompleto, determinando un diverso grado di risalita degli organi addominali nel torace durante la vita fetale. La sopravvivenza è condizionata principalmente dalla compromissione del parenchima polmonare che causa un mancato sviluppo alveolare e vascolare con conseguenti ipoplasia e ipertensione polmonare. L’aumentata sopravvivenza alla nascita dei neonati con EDC ha comportato un parallelo aumento della morbilità respiratoria a lungo termine.
