AbstractOligomeric species formed during α-synuclein fibrillation are suggested to be membrane-disrupting agents, and have been associated with cytotoxicity in Parkinson’s disease. The majority of studies, however, have revealed that the effect of α-synuclein oligomers is only noticeable on systems composed of anionic lipids, while the more physiologically relevant zwitterionic lipids remain intact. We present experimental evidence for significant morphological changes in zwitterionic membranes containing cholesterol, induced by α-synuclein oligomers. Aim: To date, assessment of the anaerobic components to determine performance in rowers is not part of standard evaluation or monitoring set for training. The aim of this study was to evaluate the relation between anaerobic power output and ergometer rowing performance over 2000 meters, in a group of 14 -17 years old rowers (n = 11). Methods: Each subject performed, in three separate days, two “all-out” tests, over 20 and 60 seconds, and a 2000 meters trial, using a Concept II rowing ergometer. Staphylococcal alpha-toxin forms homo-oligomeric channels in lipid bilayers and cell membranes. Here, we report that electrophysiological monitoring of single-channel function using a derivatized cysteine substitution mutant allows accurate determination of the subunit stoichiometry of the oligomer in situ. The electrophysiological phenotype of channels formed in planar lipid bilayers with the cysteine replacement mutant I7C is equal to that of the wild type. When pores were formed with I7C, alterations of several channel properties were observed upon modification with SH reagents. Seven physically fit (well-trained, maximal oxygen uptake 69.6 +/- 4.4 ml x kg-1 min-1) and eight less fit (moderately trained, maximal oxygen uptake 56.1 +/- 5.7 ml x kg-1 x min-1) healthy male subjects were exercised for 4 h by bicycle ergometry against a pedalling resistance calculated to cause oxygen consumption corresponding to approximately 30% of each individual's maximal oxygen uptake value. Respiratory exchange ratio was estimated at 1 h and blood glucose and lactate concentrations and muscle glycogen content at 2 h intervals.
