Retinol and retinyl esters in pigment epithelium of rats with inherited retinal degeneration

1976

A comparative study of the retinol and retinyl ester concentrations was performed in the retinal pigment epithelium of the normal and affected rats. Our findings indicate that in dystrophic rat retinol content increases, whereas the amount of retinyl esters is always lower than normal. An hypothesis can be made on the deficiency of enzymic activities which regulate retinol retinol and retinyl esters levels in the pigment epithelium.

Efficient gene delivery to photoreceptors using AAV2/rh10 and rescue of the Rho–/– mouse

2015

As gene therapies for various forms of retinal degeneration progress toward human clinical trial, it will be essential to have a repertoire of safe and efficient vectors for gene delivery to the target cells. Recombinant adeno-associated virus (AAV) serotype 2/2 has been shown to be well tolerated in the human retina and has provided efficacy in human patients for some inherited retinal degenerations. In this study, the AAV2/8 and AAV2/rh10 serotypes have been compared as a means of gene delivery to mammalian photoreceptor cells using a photoreceptor specific promoter for transgene expression. Both AAV2/8 and AAV2/rh10 provided rescue of the retinal degeneration present in the rhodopsin kno…

Combination of Resveratrol with omega-3 fatty acids synergize to counteract VEGF-R pathway in sick retinal pigment epithelium cells mimicking AMD

2015

Purpose The aim of this work is to assess whether RSV (Resveratrol) can act synergistically with omega-3 fatty acids to modulate VEGF (Vascular Endothelial Growth Factor) signaling pathway in order to identify a new and more effective therapy for the treatment of AMD (Age-Related Macular Degeneration). Methods In this study, undifferentiated and differentiated human retinal pigment epithelial cells (ARPE-19) were used. The cells were treated with an omega-3/RSV preparation (Resvega®), or a RSV-free formulation or RSV alone for 24 h. The expression of key proteins in VEGF signaling pathway was evaluated by Western Blotting. Results We observed that the combination omega-3/RSV preparation (Re…

Status of Recombinant Factor VIII Concentrate Treatment for Hemophilia A in Italy: Characteristics and Clinical Benefits

2019

The current interest in recombinant factor VIII (rFVIII) products stems from the fact that they offer a technological solution to prolonging the half-life of and reducing the risk of formation of alloantibodies (inhibitors) against FVIII in treated patients with hemophilia A (HA). The Italian health care system has authorized the use of a wide range of rFVIII concentrates of the first, second, and third generation, as well as new innovative rFVIII preparates with an extended half-life (EHL) (Kogenate FS®-Bayer, belonging to the second generation and replaced since 2017 by a product consisting of the same modified molecule; because it is only available until the end of the current year, it w…

Rewilding Urban Space. The Regeneration of Budolfi Plads (Aalborg, DK)

2022

Tackling environmental degradation and halting the growing loss of biodiversity are among the most urgent imperatives at the turn of the third millennium. In response to the global environmental crisis, different approaches to nature conservation, biodiversity protection and ecosystem restoration are emerging.

Proteostasis Deregulation in Neurodegeneration and Its Link with Stress Granules: Focus on the Scaffold and Ribosomal Protein RACK1.

2022

The role of protein misfolding, deposition, and clearance has been the dominant topic in the last decades of investigation in the field of neurodegeneration. The impairment of protein synthesis, along with RNA metabolism and RNA granules, however, are significantly emerging as novel potential targets for the comprehension of the molecular events leading to neuronal deficits. Indeed, defects in ribosome activity, ribosome stalling, and PQC—all ribosome-related processes required for proteostasis regulation—can contribute to triggering stress conditions and promoting the formation of stress granules (SGs) that could evolve in the formation of pathological granules, usually occurring during ne…

Management of immature teeth by dentin-pulp regeneration : a recent approach

2010

Treatment of the young permanent tooth with a necrotic root canal system and an incompletely developed root is very difficult and challenging. Few acceptable results have been achieved through apexification but use of long-term calcium hydroxide might alter the mechanical properties of dentin. Thus, one alternative approach is to develop and restore a functional pulp-dentin complex. Procedures attempting to preserve the potentially remaining dental pulp stem cells and mesenchymal stem cells of the apical papilla can result in canal revascularization and the completion of root maturation. There are several advantages of promoting apexogenesis in immature teeth with open apices. It encourages…

Ultrafast optics in coherently spinning molecules

2020

This thesis reports on the study of unidirectional rotation of molecules and focuses on the rotational Doppler effect occuring in the course of third-harmonic generation in coherently spinning molecules. The rotational Doppler effect appears when a circularly polarized wave interacts with an anisotropic rotating object. During the interaction, an exchange of angular momentum and energy between the field and the object occurs, leading to a frequency shift of the wave. In this work, the frequency shift of the third-harmonic generated in unidirectionaly rotating molecules has been observed. An anlytical model developped in the mansucript forsees four kinds of frequency shifts depending on the m…

A reconfigurable platform for evaluating the performance of QoS networks

2010

Nowadays, high performance System and Local Area Networks (SAN/LAN) have to serve heterogeneous traffic consisting of information flows with different bandwidth and latency requirements. This makes it necessary to provide Quality of Service (QoS) and optimize the design of network components. In this paper we present a hardware tool designed to analyze the performance of QoS networks, under given traffic conditions and server models. In particular, a reprogrammable multimedia traffic Generator/Monitor platform has been built. This permits prototyping the communication system of a high speed LAN/SAN on a single FPGA device. Hence, it can be used at design to produce more efficient devices. T…
