Plant mitochondrial genomes are generally not used to study evolution within and between species. They are larger than animal mitochondrial genomes and evolve by accumulating rearrangements rather than mutations [1]. These characteristics make it difficult to obtain complete assemblies using short-reads like in mammals. To address this problem, we developed and tested a plant-based assembly workflow using Nanopore and Pacbio long-reads from Pisum sativum and Phaseolus vulgaris, respectively. Shifts in frequency and magnitude of rain events may affect plant physiology and soil microbial activity. We aimed to determine how precipitation history shapes the response dynamics of soil microbial communities to rewetting, as well as plant-microbial competition for N, and how the N status of the system may modulate the effects of precipitation regime. Vicia faba (faba bean) is a nitrogen-fixing protein crop that, in contrast to soya bean, yields well in temperate climates. It is diploid but has a very large genome of ~13 Gb, and genomic resources are currently limited. To provide a high-quality reference gene set and expression atlas, we have carried out RNA-seq of multiple tissues from five faba bean accessions. For each tissue, an assembly was produced using the Trinity software package using both Illumina and PacBio reads. For each accession, the assemblies were then merged to create a more complete reference gene set. Nitrogen is the major nutrient limiting plant growth in terrestrial ecosystems. The transformation of inert nitrogen to forms that can be assimilated by plants is mediated by soil microorganisms. However, global cycling of nitrogen has transgressed a critical threshold: the amount of atmospheric N2 industrially converted into ammonia to produce fertilizers now exceeds that produced from all of the Earth’s terrestrial biological processes. This excess reactive N from fertilizer use poses a threat to soil, water, and air quality due to nitrate leaching and emission of N2O, a potent greenhouse gas.
