Angular Dependency of Hyperspectral Measurements over Wheat Characterized by a Novel UAV Based Goniometer

2015

In this study we present a hyperspectral flying goniometer system, based on a rotary-wing unmanned aerial vehicle (UAV) equipped with a spectrometer mounted on an active gimbal. We show that this approach may be used to collect multiangular hyperspectral data over vegetated environments. The pointing and positioning accuracy are assessed using structure from motion and vary from σ = 1° to 8° in pointing and σ = 0.7 to 0.8 m in positioning. We use a wheat dataset to investigate the influence of angular effects on the NDVI, TCARI and REIP vegetation indices. Angular effects caused significant variations on the indices: NDVI = 0.83–0.95; TCARI = 0.04–0.116; REIP = 729–735 nm. Our analysis high…

The Prairie Diagnosis: a tool to assess and characterize the grassland base of long-term temporary and permanent grasslands

2022

Permanent and temporary grasslands represent nearly 44 % of the French UAA. This vast area is rich in a very wide range of flora, from hillside meadows with subterranean clover to flooded meadows and marshes with Baldingere, not forgetting sown meadows which, over the years, can host numerous spontaneous species. The method of prairie diagnosis is based on a botanical approach to the prairie with the objective of qualifying the forage aptitudes of the cover. The diagnosis of a meadow is only of interest and relevance if it is integrated into a forage system consistent with the strategies of a farmer. It is based on the premise that the flora of a meadow is the result of the interaction of "…

Micro-scale post-fire surface cover changes monitored using high spatial resolution photography in a semiarid environment: A useful tool in the study…

2012

[ES] Although post-fire soil erosion has been studied, little attention has been paid to changes in soil surface cover following fires, despite this being a key factor in understanding the water and sediment yield. This study, at Peñaflor (Spain), investigated the effect of fire on soil erosion using data from soil erosion plots and high spatial resolution photography (HSRP). Measurements were made from October 2003 to October 2005 in a control plot and a plot experimentally exposed to a fire in October 2004. Ground cover components were identified, including vegetation, bare soil, stones, charcoal and ash. Runoff and sediment concentrations were low because of the absence of intense rainfa…

RPB1 as a marker gene to analyze communities of arbuscular mycorrhizal fungi (Glomeromycota)

2014

The influence of agricultural practice on the diversity of Arbuscular mycorrhizal fungi (AMF, Glomeromycota) continues to be an important research question as these mutualistic symbionts are known to improve plant growth and soil quality. To this day, studies of community diversity in AMF have exclusively been based on nuclear ribosomal gene regions, such as the small and large subunits and the ITS region. In the Glomeromycota, these regions show high intra-organism polymorphism, which can seriously complicate interpretation of diversity data. We chose the rpb1 gene as an alternative marker as this putative single-copy gene has been shown to be monomorphic within fungal isolates but shows a…

Validation of a set of reference genes to study response to herbicide stress in grasses

2012

Abstract Background Non-target-site based resistance to herbicides is a major threat to the chemical control of agronomically noxious weeds. This adaptive trait is endowed by differences in the expression of a number of genes in plants that are resistant or sensitive to herbicides. Quantification of the expression of such genes requires normalising qPCR data using reference genes with stable expression in the system studied as internal standards. The aim of this study was to validate reference genes in Alopecurus myosuroides, a grass (Poaceae) weed of economic and agronomic importance with no genomic resources. Results The stability of 11 candidate reference genes was assessed in plants res…

Variation in the TonB-dependent outer-membrane proteins in plant-associated strains of Pseudomonas fluorescens

2011

International audience; Genomic sequences of ten strains of plant-associated Pseudomonas spp. were surveyed for the presence of TonB-dependent outer-membrane proteins (TBDPs), which function in the uptake of substrates from the environment by many Gram-negative bacteria. The ten strains represent P. fluorescens, P. chlororaphis, and P. synxantha isolated from the phyllosphere, rhizosphere or soil. 14 to 45 TBDPs were identified in each strain, and phylogenetic analysis of the TBDPs identified five that are conserved across all ten genomes. Comparisons to proteins with known functions allowed the assignment of putative roles in uptake of heme, vitamin B12, copper, and the siderophore ferrich…

In-Field LAMP Detection of Flavescence Dorée Phytoplasma in Crude Extracts of the Scaphoideus titanus Vector

2022

One of the most destructive diseases affecting grapevine in Europe is caused by Flavescence Dorée phytoplasma (FDp), which belongs to the 16Sr-V group and is a European Union quarantine pathogen. Although many molecular techniques such as loop-mediated isothermal amplification (LAMP) are widely used for the rapid detection of FDp in infected grapevine plants, there is no developed isothermal amplification assay for FDp detection in the insect vectors that are fundamental for the spread of the disease. For this reason, a simple in-field real-time LAMP protocol was optimized and developed for the specific detection of FDp in the insect vector Scaphoideus titanus. The LAMP assay was optimized …
