An isolierten Herzohren von Meerschweinchen wurden intracellulare Aktionspotentiale unter gleichzeitiger Registrierung der Kontraktionen abgeleitet. Untersucht wurde der Einflus des Digitoxigenins nach Barbituratschadigung und unter Calciummangel bei euthyreoten und hyperthyreoten Tieren.

An isolierten, elektrisch gereizten Meerschweinchenvorhofen wurde das Verhalten der Kontraktionskraft und des cellularen Ca-Haushaltesunter dem Einflus von Digitoxigenin, Dihydrodigitoxigenin und 3α-Digitoxigenin uber einen weiten Dosierungsbereich vergleichend untersucht. Diese Substanzen zeigten betrachtliche Unterschiede in der Intensitat ihrer positiv inotropen und kontrakturerzeugenden Wirkung. Im positiv inotropen Dosierungsbereich verhielten sich die aquieffektiven Konzentrationen von Digitoxigenin, Dihydrodigitoxigenin und 3α-Digitoxigenin etwa wie 1:20:70. Es wurde uber einen weiten Bereich die Calciumkonzentration der Tyrodelosung verandert (0,9–9,0 mAq/l) und unter diesen Bedingungen die Konzentrationswirkungsbeziehungen fur verschiedene Digitoxigenindosen (1 × 10−7, 3 × 10−7, 5 × 10−7, 7 × 10−7 und 1 × 10−6 g/ml) am Meerschweinchenvorhof untersucht. Aus den Befunden geht hervor, das uber den gesamten untersuchten Bereich unter therapeutischen und toxischen Bedingungen ein eindeutiger Wirkungszusammenhang zwischen Digitoxigenin und Calcium besteht.

Secondary metabolites from the roots of Paronychia chionaea

Two novel secondary metabolites, compounds (1–2) were isolated from the roots of Paronychia chionaea. On the basis of spectroscopic data including 1D and 2D NMR experiments (COSY, TOCSY, HSQC, and HMBC), and mass spectroscopy, their structures were established as 6- C-[α-L-arabinopyranosyl-(1→2)-β-D-glucopyranosyl]-7- O-[β-D-glucopyranosyl]-luteolin 3′-methyl ether (1), and 2-(methoxy)-2-(3,5-dimethoxy 4-hydroxyphenyl)-ethane-1,2-diol 1- O-β-D-glucopyranoside (2).

Secondary metabolites from polar fractions of Piper umbellatum.

Seven known secondary metabolites were isolated from the methanol extract of the branches of Piper umbellatum. The identification of these compounds was mainly achieved by 2D NMR spectroscopic techniques and FAB-MS. Among them, the known cepharadiones A and B can be considered as chemotaxonomic markers of the genus Piper.

Spirostane-Type Saponins from Dracaena fragrans Yellow Coast

Three steroidal glycosides were isolated from the bark of Dracaena fragrans (L.) Ker Gawl. « Yellow Coast », and a fourth from the roots and the leaves. Their structures were characterized on the basis of extensive 1D and 2D NMR experiments and mass spectrometry, and by comparison with NMR data of the literature. These saponins have the spirostane-type skeleton and are reported in this species for the first time.

Malignant melanoma is a highly aggressive tumor that frequently resists chemotherapy, so the search for new agents for its treatment is of great importance. In the present study, the antiproliferative propensity against human melanoma cell lines of lauroside B (1), a megastigmane glycoside isolated from Laurus nobilis (bay laurel) leaves, was investigated.
