Involvement of neuronal processes and nitric oxide in the inhibition by endotoxin of pentagastrin-stimulated sastric acid secretion

Administration of E. coli endotoxin (1 mg kg-1, i.v.) abolished the acid response induced by the i.v. infusion of pentagastrin (8 micrograms kg-1 h-1) in the continuously perfused stomach of the anaesthetized rat. Local serosal application of tetrodotoxin (36 ng per rat) completely restored acid responses to pentagastrin in endotoxin-treated rats. However, pretreatment with atropine (0.5 mg kg-1, s.c.), capsaicin (20, 30, and 50 mg kg-1, s.c. 2 weeks before the study) or guanethidine (16 mg kg-1, s.c. 3 and 16h before) did not influence the inhibitory effects of endotoxin. Tetrodotoxin-dependent effects of menthol on mouse gastric motor function.

Menthol, the main active constituent of peppermint oil, exerts gut spasmolytic effects, although its mechanism of action remains unclear. We investigated the effects of menthol on gastric emptying and spontaneous- or evoked- mechanical activity of whole murine stomach. Gastric emptying was calculated after i.p. administration of menthol (50mg/Kg). Responses induced by menthol on gastric intraluminal pressure and evoked-cholinergic contractions were analyzed in vitro. Menthol decreased the gastric emptying rate. Modulation by nitric oxide of spontaneous mechanical activity in rat proximal colon.

Summary 1 In order to examine the role of nitric oxide (NO) in the tonic neural inhibition in rat proximal colon, the effects of Nω-nitro- l-arginine methyl ester (L-NAME) were studied on the spontaneous contractions of circular muscle (monitored as intraluminal pressure changes) and of longitudinal muscle (detected as isometric tension changes). 2 L-NAME (3 × 10−−6–3 × 10−−4 m) caused a concentration-dependent increase in the amplitude of circular contractions, without affecting those of longitudinal muscle. Influence of nitric oxide on neurogenic contraction and relaxation of the human gastroepiploic artery.

Abstract Background The objective of this study was to characterize the neurogenic contraction and relaxation of the human gastroepiploic artery and to determine whether the responses are mediated by nitric oxide (NO) from neural or endothelial origin. Methods Rings of human gastroepiploic artery were obtained from 18 patients (12 men, 6 women) undergoing gastrectomy. The rings were suspended in organ baths for isometric recording of tension. Spontaneous electromechanical activity in the rat duodenumin vitro

Isolated rat duodenum shows spontaneous mechanical and electrical activities. Mechanical activity consists in changes both in endoluminal pressure and in isometric tension. Electrical activity is characterized by slow waves with superimposed bursts. This spontaneous activity is tetrodotoxin (TTX) resistant and therefore it is myogenic in origin. Indeed, TTX pretreatment, even in the presence of atropine and guanethidine, caused an increase in amplitude and in frequency of the electrical and mechanical activities. Modulation of adrenergic contraction of dog pulmonary arteries by nitric oxide and prostacyclin.

Abstract The aim of this work was to investigate the influence of endothelium-derived nitric oxide and prostaglandins on the contractile responses of isolated dog pulmonary arteries to electrical field stimulation and noradrenaline. Electrical field stimulation (1–8 Hz, 20 v, 0.25 ms duration, for 30 s) produced frequency-dependent contractions that were abolished by tetrodotoxin, guanethidine and, prazosin (all at 10−6 M). Noradrenaline induced concentration-dependent contractions with an EC50 of 1.85 × 10−6 M. Inhibition of mechanical activity by neurotensin in rat proximal colon: involvement of nitric oxide.

The aim of the present study was to define the nature of inhibitory action of neurotensin in rat proximal colon. Mechanical activity was detected as changes of intraluminal pressure. Neurotensin (10(-10) to 10(-7) M), in the presence of atropine (10(-6) M), guanethidine (10(-6) M), and nifedipine (10(-8) M), induced a tetrodotoxin-insensitive inhibitory effect characterized by the complete disappearance of the spontaneous phasic contractions. Guanethidine was intravenously injected into rats and was subsequently determined in the hearts by extracting the tissue with trichloroacetic acid, purifying the extracts on cation exchange columns and by paper chromatography, and coupling the guanethidine with ninhydrin in alkaline solution before fluorimetric estimation. The concentration in the heart reached a maximum of 9 μg/g within 15 min of injecting 15 mg/kg, and was still 6.2 and 5.7 μg/g after 1 and 3 hours, respectively.
