Conserved TCR β chain usage in reactive arthritis; evidence for selection by a putative HLA-B27-associated autoantigen
2002
Previous work suggested that expanded CD8+ T-cell clones in the synovial fluid (SF) of HLA-B27+ patients with reactive arthritis (ReA) preferentially use the T-cell receptor variable region (TCRBV) 1, similar CDR3 sequences, and joining region (BJ) 2S3. To determine the range of conservation and disease-specificity of CDR3-sequences, we analyzed the TCRBV1-J2S3 repertoire from 33 healthy HLA-B27+ individuals, patients with various types of spondyloarthropathies (SpA), and with rheumatoid arthritis (RA) by CDR3-spectratyping. After collection and database submission of all available TCRB-CDR3 from HLA-B27-restricted or SpA-derived T cells, we systematically screened the entire human sequence…
Enterobacterial Antigens with Tropism for Joint Structures and HLA-B27=Restricted Cytotoxic T-Cells in Reactive Arthritis
1995
In reactive arthritis (ReA), sterile synovitis is an immunological sequela following gastrointestinal or urogenital infection with facultatively intracellular bacteria (Yersinia, Salmonella, Shigella, Chlamydia). It is widely accepted now that the development of arthritis is closely related to the persistance of bacteria or bacterial antigens in extraarticular mucosal or lymphoid tissues (i.e. gut mucosa, gut associated lymphoid tissue, genitourinary mucosa); however, it is still unclear which host mechanisms are responsible for the poorer elimination of arthritis-causing microorganisms in those ReA patients. Bacterial components are also camed to the joints where they can be demonstrated i…
HLA class II haplotypes differentiate between the adult autoimmune polyglandular syndrome types II and III.
2013
Background: Genetics of the adult autoimmune polyglandular syndrome (APS) is poorly understood. Aim: The aim of this study was to gain further insight into the genetics of the adult APS types. Site: The study was conducted at a university referral center. Methods: The human leukocyte antigen (HLA) class II alleles, haplotypes, and genotypes were determined in a large cohort of patients with APS, autoimmune thyroid disease (AITD), and type 1 diabetes and in healthy controls by the consistent application of high-resolution typing at a four-digit level. Results: Comparison of the allele and haplotype frequencies significantly discriminated patients with APS vs AITD and controls. The HLA class…
Identification of the Yersinia enterocolitica urease beta subunit as a target antigen for human synovial T lymphocytes in reactive arthritis.
1993
The local T-cell response to bacterial antigens is involved in the pathogenesis of reactive arthritis (ReA). Here, we have identified a 19-kDa antigen of Yersinia enterocolitica O:9 recognized by Yersinia-specific synovial fluid CD4+ T cells in two patients with Yersinia-induced ReA. N-terminal amino acid sequencing of this protein revealed that it was identical to the 19-kDa urease beta subunit of Y. enterocolitica O:9. This protein has previously been shown to be arthritogenic in preimmunized rats after intra-articular injection. Analysis of the T-cell response to this protein showed that it contains several T-cell epitopes, one of which cross-reacts with other enterobacteria not able to …
Sequence analysis of the DRB1 promoter reveals limited polymorphism with no influence on gene expression.
2001
HLA-class II promoters contain a set of conserved regulatory regions necessary for constitutive and induced gene expression. For the HLA-DQB as well as for the DRB1 promoter sequence, polymorphisms with influence on gene expression have been reported. In contrast to these data we could show that there is very limited allele-specific polymorphism among the HLA-DRB1 promoter alleles. In a long range PCR we amplified a DNA sequence containing the promoter and the second exon of the DRB1 gene in one fragment. Nested PCR products of this PCR fragment for the promoter and for the second exon were analysed by DNA sequencing to allow the linkage of a promoter to its DR allele. Most investigated DRB…
Non-conventional forms of HLA-B27 are expressed in spondyloarthritis joints and gut tissue
2016
Objectives Human leukocyte antigen (HLA)-B27 (B27) is the strongest genetic factor associated with development of Ankylosing Spondylitis and other spondyloarthropathies (SpA), yet the role it plays in disease pathogenesis remains unclear. We investigated the expression of potentially pathogenic non-conventional heavy chain forms (NC) of B27 in synovial and intestinal tissues obtained from SpA patients. We also determined the presence of NC-B27 in joints, lymphoid and gastrointestinal tissue from B27 transgenic (TG1) rats with M.tuberculosis-induced SpA. Methods Expression of NC-B27 in human SpA joints and gut and in (21-3 × 283-2)F1 HLA-B27/Huβ2m rat tissue was determined by immunohistochem…
Arthritis Associated with Ulcerative Colitis: Our Experience in Palermo
2009
To The Editor: Ulcerative colitis (UC) is one of the inflammatory bowel diseases, which affects the small intestine and colon [1]. The pathophysiology of UC is not known [2, 3]. The association between arthritis and UC is well known. The authors report the frequency of arthritis in 53 Sicilian patients with UC. The diagnosis of UC was made according to criteria described by Schachter and Kirsner [4]. The involvement of sacroilitic joints was assessed radiologically according to Bennett and Burch grade (0=normal joint, 1=suspicious sacroiliitis 2=abnormal joint with sclerosis and/or erosions, 3=unequivocally abnormal with erosions, sclerosis, widening or narrowing or partly ankylosed, 4=tota…

"–jokainen taulu on omakuva!" : Tove Janssonin Juhlat maalla ja Juhlat kaupungissa -freskot taiteilijan omakuvina
2017
Tutkimuksen aiheena on Tove Janssonin Juhlat maalla ja Juhlat kaupungissa -freskot, joita analysoitiin käyttämällä ikonologista ja narratiivista analyysimenetelmää. Menetelmien avulla freskoista löydettiin Tove Janssonin kokemusmaailmaan viittaavia omakuvallisia piirteitä. Tulosten perusteella teoksia ei voida mieltää täysin omakuvallisiksi teoksiksi, vaan niissä on nähtävissä myös taiteilijan rakkaudenosoitus rakastetulleen.
Cavalleria Rusticana & Pajatso Savonlinnan oopperajuhlilla ja niiden lehdistökritiikki
2010
Musiikkikritiikkejä on tutkittu erityisesti konserttien osalta jo kauan.
