Context-dependent Pax-5 repression of a PU.1/NF-κB regulated reporter gene in B lineage cells

Enhancers located in the 3' end of the locus in part regulate immunoglobulin heavy chain (IgH) gene expression. One of these enhancers, HS 1,2, is developmentally regulated by DNA binding proteins like NF-kappaB, Pax-5 and the protein complex NF-alphaP in B lineage cells. Here we report that NF-alphaP is the ets protein PU.1. A glutathione-S-transferase (GST)-pulldown assay demonstrated that PU.1 can physically interact with NF-kappaB in solution. Experiments in COS cells showed that PU.1 and NF-kappaB (p50/c-Rel) can activate transcription of an enhancer linked reporter gene. Xanthones and sesquiterpene derivatives from a marine-derived fungus Scopulariopsis sp.

Abstract Two new xanthone derivatives, 12-dimethoxypinselin ( 1 ) and 12- O -acetyl-AGI-B4 ( 2 ), as well as two new phenolic bisabolane-type sesquiterpenes, 11,12-dihydroxysydonic acid ( 15 ) and 1-hydroxyboivinianic acid ( 16 ), together with one new alkaloid, scopulamide ( 21 ) and one new α-pyrone derivative, scopupyrone ( 26 ), in addition to twenty-three known compounds ( 3 – 14 , 17 – 20 , 22 – 25 , 27 – 29 ) were isolated from solid rice cultures of the marine-derived fungus Scopulariopsis sp. obtained from the Red Sea hard coral Stylophora sp. Cytotoxic eudesmane sesquiterpenes from Crepis sancta

Abstract Chemical exploration of Crepis sancta (L.) Bornm. (Asteraceae) aerial parts afforded two new eudesmane sesquiterpenes (1 and 2) together with three known congeners (3-5) and two known methylated flavonoids (6 and 7). Structure elucidation of the new compounds was unambiguously performed based on HRESIMS, 1D and 2D NMR spectroscopic analyses. All isolated compounds were subjected to an in vitro cytotoxicity assay against mouse lymphoma (L5178Y) cells, revealing moderate activities with IC50 ranging from 7.9 to 21.0 μM.

A STUDY OF POPULATION GENETICS IN THE EUROPEAN LOBSTER, HOMARUS GAMMARUS (DECAPODA, NEPHROPIDAE)

By means of an RAPD analysis, the degree of genetic variation in the European lobster, Homarus gammarus, was assessed. We focused our attention on the Helgoland population, which is isolated by its geographical location. By comparison with seven other European lobster populations we demonstrated that the Helgoland lobster is genetically widely separated from other European lobster stocks. However, all populations investigated showed high values of genetic distance, indicating that genetic exchange among European populations of Homarus gammarus may be restricted. Ophiobolin Sesterterpenoids and Pyrrolidine Alkaloids from the Sponge-Derived Fungus Aspergillus ustus

Chemical examination of the fungus Aspergillus ustus isolated from the Mediterranean sponge Suberites domuncula yielded the five new ophiobolin-type sesterterpenoids 1–5 and the two new pyrrolidine alkaloids 6 and 7, together with the known compound aurantiamine and cerebroside D. The structures of the new compounds were unambiguously elucidated on the basis of extensive spectroscopic-data analysis (1D- and 2D-NMR, MS, and UV) and comparison with literature data. All compounds were evaluated for their cytotoxicity against murine lymphoma cell line L5178Y.

Eurhomalea exalbida (Bivalvia): A reliable recorder of climate in southern South America?

Abstract Due to the lack of suitable high-resolution archives, regional and continental-scale climate dynamics of southern South America are not well understood. Shells of the long-lived, shallow-marine bivalve mollusk, Eurhomalea exalbida (Dillwyn), are likely to contain information on the past water temperatures. As yet, however, no rigorous calibration study has been presented so that growth history traits and the reliability of shell oxygen isotope-based temperature estimates remain unknown.
