''Deferoxamine blocks death induced by glutathione depletion in PC 12 cells''
2013
Chouraqui, E. | Leon, A. | Repesse, Y. | Prigent-Tessier, A. | Bouhallab, S. | Bougle, D. | Marie, C. | Duval, D.; International audience; ''The purpose of the present work was to investigate the mechanisms by which glutathione depletion induced by treatment with buthionine sulfoximine (BSO) led within 24-30 h to PC 12 cells apoptosis. Our results showed that treatment by relatively low concentrations (10-30 mu M) of deferoxamine (DFx), a natural iron-specific chelator, almost completely shielded the cells from BSO-induced toxicity and that DFx still remained protective when added up to 9-12 h after BSO treatment. On the other hand, phosphopeptides derived from milk casein and known to carr…
Dihydroquercetin (DHQ) induced HO-1 and NQO1 expression against oxidative stress through the Nrf2-dependent antioxidant pathway.
2013
Dihydroquercetin (DHQ) is a well-known antioxidant agent. In the present investigation, we reported for the first time that DHQ stimulates the expression of phase II detoxifying enzymes through the Nrf2-dependent signaling pathway. The IC50 values of DHQ for reduction of 2,2-diphenyl-1-picrylhydrazol (DPPH), reducing power assay, lipid peroxidation assay, and xanthine oxidase inhibition were 5.96, 4.31, 2.03, and 13.24 μM, respectively. DHQ possessed considerable protective activity from oxidative DNA damage. A luciferase reporter assay also demonstrated that DHQ-activated signaling resulted in the increased transcriptional activity of Nrf2 through binding to the ARE (antioxidant response e…
Cadmium as a transcriptional modulator in human cells
2010
Cadmium (Cd) is an underground mineral widely used in the steel industry, in plastics, and as a component of batteries. It is an industrial and environmental pollutant released as an air contaminant from fertilizers and, more prominently, in the form of wastewater. Food, drinking water, and, mainly, inhalation of smoke from cigarettes are sources of daily exposure of humans to the heavy metal. Although Cd has no known useful function for humans as well as other organisms, it appears to evoke in cells a number of responses that involve not only death signaling but also protective reactions against the toxicity. This finding prompted a number of experimental studies aimed to elucidate the cel…
Heme oxygenase-1 induction modulates microsomal prostaglandin E synthase-1 expression and prostaglandin E2 production in osteoarthritic chondrocytes
2009
Pro-inflammatory cytokines such as interleukin-1beta (IL-1beta) may participate in the pathogenesis of cartilage damage in osteoarthritis (OA) through the production of catabolic enzymes and inflammatory mediators. Induction of heme oxygenase-1 (HO-1) has previously been shown to exert anti-inflammatory effects in different cell types. We have investigated whether HO-1 induction may modify chondrocyte viability and the production of relevant mediators such as oxidative stress and prostaglandin E(2) (PGE(2)) elicited by IL-1beta in OA chondrocytes. Chondrocytes were isolated from OA cartilage and used in primary culture. Cells were stimulated with IL-1beta in the absence or presence of the H…
Influence of oxygen tension on the anti-inflammatory and chondroprotective effects of heme oxygenase-1 in healthy and osteoarthritic human chondrocyt…
2012
s / Osteoarthritis and Cartilage 20 (2012) S54–S296 S136 their isolation. The absence of cross-reaction with the IIA isoform was established by ELISA andWB. In addition, the Saos-2 cell linewas chosen to test a possible labelling of other fibrillar procollagens, mainly type I, V and XI. In fact, this cell line is described to synthesize the (a1)I, (a2)I, (a1)V, (a2)V, (a1)XI and (a2)XI, but no (a3)XI chains. No signal was detected on WB of cellular extracts or conditioned media with anti-pNIIB52, whereas antibodies to the collagen I, V and XI triple-helical parts revealed indeed the presence of proforms of these collagens. Conclusions: Anti-pNIIB52 antibodies allow a very sensitive and spec…
Characterization of the Antioxidant Properties of Pentaerithrityl Tetranitrate (PETN)-Induction of the Intrinsic Antioxidative System Heme Oxygenase-…
2009
Organic nitrates are among the oldest and yet most commonly employed drugs in the chronic therapy of coronary artery disease and congestive heart failure. While they have long been used in clinical practise, our understanding of their mechanism of action and of their side effects remains incomplete. To date, the most commonly employed nitrates are isosorbide mononitrate (ISMN), isosorbide dinitrate (ISDN), and nitroglycerin (GTN). Another nitrate, pentaerithrityl tetranitrate (PETN), has long been employed in eastern European countries and is currently being reintroduced also in western countries. So far, PETN is the only organic nitrate in clinical use, which is devoid of induction of oxid…
Antioxidant and Antiinflammatory Properties of Heme Oxygenase-1 in Osteoarthritic Articular Cells
2012
Heme oxygenase-1 (HO-1) is induced in cells by various stimuli as a defense system against oxidative stress. It is known that reactive oxygen species (ROS) participates in the initiation and progression of osteoarthritis (OA) and several antioxidant systems may protect cartilage components. HO-1 induction or CO release from CORM-2 counteracts oxidative stress and protects against proinflammatory and catabolic effects of interleukin-1β in OA chondrocytes, osteoblasts, and synoviocytes as well as in OA osteochondral explants. Both approaches have been able to downregulate the production of mediators such as reactive oxygen species, nitric oxide, matrix metalloproteinases, prostaglandin E2, cy…
Tema starosti v romane Gajto Gazdanova "Nočnye dorogi"
2016
Do tematu starości niejednokrotnie zwracał się w swojej twórczości przedstawiciel młodszego pokolenia pierwszej fali emigracji rosyjskiej, Gajto Gazdanow, najczęściej rozpatrując starość w kontekście sensu życia i śmierci. Temat starości dla Gazdanowa to przede wszystkim możliwość rozważania o sensie życia i śmierci, pokazania kontrastu pomiędzy ograniczonością, bezsensem istnienia i przeżywaniem…
BIFOCAL PAIRS OF IMAGES FOR LOW-COST SURVEY IN CLOSE RANGE PHOTOGRAMMETRY
2019
Abstract. This paper present a method for close range photogrammetry based on an camera positioning scheme in which two cameras capture an equal portion of an object at the same scale, but have different focal lengths and camera-to-object distances. This scheme is alternative to the stereoscopic scheme and is associated with a system of equations which permits one to calculate first the relief displacement of points on a photograph and then their relief relative to a reference plane. The obtained relief and relief displacement values can be used to produce low-cost orthophotographs by using software for image processing, which doesn’t need to be dedicated, but has to provide measurement and…
ROLE OF HEME OXYGENASE-1 (HSP32) AND HSP90 IN GLIOBLASTOMA
2017
Glioblastoma (GBM) is the most common and malignant primary brain tumor in adults. The current treatment regimes for glioblastoma demonstrated a low efficiency and offer a poor prognosis. Advancements in conventional treatment strategies have only yielded modest improvements in overall survival. The heat shockproteins, heme oxygenase-1 (HO-1) and Hsp90, serve these pivotal roles in tumor cells and have been identified as effective targets for developing therapeutics. This topic review summarizes the current preclinical and clinical evidences and rationale to define the potential of HO-1 and Hsp90 in GBM progression and chemoresistance.
