Targeting NUPR1 with the Small Compound ZZW-115 Is an Efficient Strategy to Treat Hepatocellular Carcinoma

International audience; HCC is a highly lethal malignancy with Sorafenib as the only molecularly targeted drug. The multifunctional stress-associated protein, NUPR1, plays an essential role in controlling cell growth, migration, invasion and Sorafenib resistance in HCC. We report here that NUPR1 expression is absent in healthy liver and it is progressively upregulated in HCC premalignant lesions such as hepatitis and cirrhosis with a maximum expression in HCC samples, highlighting that NUPR1 is a potential drug target for HCC. Hepatocellular Carcinoma (HCC) is worldwide the 5th most common diagnosed cancer in men and the 7th in women and the 4th cause of cancer death with a median survival between 10 and 11 months. The incidence rate in France is relatively high (7.8/100 000 with 10 624 estimated new cases in 2018). HCC prognosis is very poor due to the difficulty to diagnose it at an early stage. Around 80% of HCC occur in the setting of cirrhosis, mainly caused by alcoholism, chronic infections with HBV and HCV or metabolic syndrome. Immune pathogenesis of hepatitis A

In an effort to elucidate the mechanism of liver damage resulting from Hepatitis A virus (HAV) infection, we have studied infected skin fibroblasts and autologous lymphocytes from HAV patients. We report here that HLA-restricted virus-specific T cells play an essential role in HAV-related hepatocellular injury.
