Selective depletion of alloreactive T cells from stem-cell allografts should abrogate graft-versus-host disease while preserving beneficial T cell specificities to facilitate engraftment and immune reconstitution. We therefore explored a refined immunomagnetic separation strategy to effectively deplete alloreactive donor lymphocytes expressing the activation antigen CD69 upon stimulation, and examined the retainment of antiviral, antileukemic, and immunoregulatory T cells. In addition to the CD69high T cell fraction, our studies retrieved two T cell subsets based on residual CD69 expression. Abstract The gas chromatography of mixtures of n -alkyl acetates (CH 3 COOR, R = C 1 —C 8 ) and methyl esters of aliphatic n -carboxylic acids (R′COOCH 3 , R′ = C 1 —C 8 ) and certain of their monochlorinated derivatives has been studied on Carbowax 20M and SE-30 glass capillary columns under the same operating conditions. The separation of the isomeric monochlorinated esters was complete on Carbowax 20M, whereas on SE-30 the peaks of 6- and 7-chlorooctyl acetates and methyl (ω − 1)- and (ω − 2)-chlorooctanoates and -nonanoates partly overlapped. Some haematic parameters were investigated in female COBS mice treated with 7 alpha, 17 beta Dimethyltestcsterone (DMT). The drug causes an increase of circulating platelets in normal mice. Bone marrow graft from DMT-treated donors facilitates in irradiated mice repopulation of white blood cells and platelets but lowers % survival. These data are interpreted on the basis of a commitment and a loss of self-maintenance induced by DMT on CFUs compartment. © 1979 The Italian Pharmacological Society.

AbstractAllogeneic hematopoietic stem cell transplantation can induce considerable tumor remissions in metastatic renal-cell carcinoma (RCC) patients. The precise effector mechanisms mediating these graft-versus-tumor reactions are unknown. We studied RCC-directed CD8+ T-cell responses in blood lymphocytes of healthy individuals matched with established RCC cell lines for HLA-class I. In 21 of 22 allogeneic mixed lymphocyte/tumor-cell cultures (MLTCs), RCC-reactive cytotoxic T-lymphocytes (CTLs) were readily obtained.
