Differences of stabilized organic carbon fractions and microbiological activity along Mediterranean Vertisols and Alfisols profiles

2010

This study examined the chemical and structural properties of humic substances and microbiological activity in order to verify differences in carbon dynamics along soil profiles in two Vertisols (Typic Haploxerert and Xeric Epiaquert) and two Alfisols (Mollic Haploxeralf and Ultic Haploxeralf) developed under Mediterranean climate in Italy. Humification parameters, thermal methods, including differential scanning calorimetry (DSC) and thermogravimetry (TG), together with Fourier transform infrared (FT-IR) and fluorescence spectroscopies were used to characterize humic acids (HA). Microbiological activity of soils was assessed by basal respiration, metabolic quotient (qCO2) and Cmic:Corg rat…

Energy use in the A and B horizons of the soil under a pine and a cedar stand

2002

Publisher Summary This chapter discusses a study that compares organic carbon content, availability of substrates, and microbial metabolism in two afforested sites in Sicily (Italy) developed under the same climatic and physical conditions but underlying different tree species. Given the key role of the soil microbiota in promoting energy exchanges and transformations in the soil profile, an analysis of the microbial activity and of organic carbon pools was performed on twenty profiles. The parameters considered in this study were based on microbial activity, organic carbon fractions, and kinetics of organic C mineralization. In the two systems studied, the competition for available energy …

Plasmid conjugation from Proteobacteria as evidence for the origin of xenologous genes in Cyanobacteria

2014

Comparative genomics have shown that 5% of Synechococcus elongatus PCC 7942 genes are of probable proteobacterial origin. To investigate the role of interphylum conjugation in cyanobacterial gene acquisition, we tested the ability of a set of prototype proteobacterial conjugative plasmids (RP4, pKM101, R388, R64, and F) to transfer DNA from Escherichia coli to S. elongatus. A series of BioBrick-compatible, mobilizable shuttle vectors was developed. These vectors were based on the putative origin of replication of the Synechococcus resident plasmid pANL. Not only broad-host-range plasmids, such as RP4 and R388, but also narrower-host-range plasmids, such as pKM101, all encoding MPFT-type IV …

The evolutionary genetics of the hobo transposable element in the Drosophila melanogaster complex.

1994

Hobo elements are a family of transposable elements found in Drosophila melanogaster and its three sibling species: D. simulans, D. mauritiana and D. sechellia. Studies in D. melanogaster have shown that hobo may be mobilized, and that the genetic effects of such mobilizations included the general features of hybrid dysgenesis: mutations, chromosomal rearrangements and gonadal dysgenis in F1 individuals. At the evolutionary level some hobo-hybridizing sequences have also been found in the other members of the melanogaster subgroup and in many members of the related montium subgroup. Surveys of older collected strains of D. melanogaster suggest that complete hobo elements were absent prior t…

Presence of multiple group I introns closely related to bacteria and fungi in plastid 23S rRNAs of lichen-forming Trebouxia

2009

The chloroplast-encoded large subunit ribosomal RNA gene of several free-living green algae contains group I introns at Escherichia coli genic positions 1917, 1931, 1951, and 2449. Herein we report the presence of group I introns at these positions within the chloroplast-encoded large subunit ribosomal RNA gene of several lichen-forming green algae belonging to the Trebouxia genus. In contrast to the introns inserted at position 2449, all introns inserted at positions 1917, 1931, and 1951 contained LAGLIDADG homing endonuclease genes. Phylogenetic analyses show that: (i) introns inserted at positions 1917, 1931, and 1951 are closely related to introns located at homologous insertion sites i…

What is in a lichen? A metagenomic approach to reconstruct the holo-genome of Umbilicaria pustulata

2019

AbstractLichens are valuable models in symbiosis research and promising sources of biosynthetic genes for biotechnological applications. Most lichenized fungi grow slowly, resist aposymbiotic cultivation, and are generally poor candidates for experimentation. Obtaining contiguous, high quality genomes for such symbiotic communities is technically challenging. Here we present the first assembly of a lichen holo-genome from metagenomic whole genome shotgun data comprising both PacBio long reads and Illumina short reads. The nuclear genomes of the two primary components of the lichen symbiosis – the fungus Umbilicaria pustulata (33 Mbp) and the green alga Trebouxia sp. (53 Mbp) – were assemble…

Branch-and-Price-and-Cut for the Truck-and-Trailer Routing Problem with Time Windows

2018

In this paper, we present a new branch-and-price-and-cut algorithm to solve the truck-and-trailer routing problem with time windows (TTRPTW) and two real-world extensions. In all TTRPTW variants, the fleet consists of one or more trucks that may attach a trailer. Some customers are not accessible with a truck-and-trailer combination, but can however be serviced by one if the trailer is previously detached and parked at a suitable location. In the first extension, the planning horizon comprises two days and customers may be visited either on both days or only once, in which case twice the daily supply must be collected. The second extension incorporates load transfer times depending on the …

Simultaneous Vehicle and Crew Routing and Scheduling for Partial- and Full-Load Long-Distance Road Transport

2013

AbstractThis paper studies a simultaneous vehicle and crew routing and scheduling problem arising in long-distance road transport in Europe: Pickup-and-delivery requests have to be fulfilled over a multi-period planning horizon by a heterogeneous fleet of trucks and drivers. Typically, in the vehicle routing literature, a fixed assignment of a driver to a truck is assumed. In our approach, we abandon this assumption and allow truck/driver changes at geographically dispersed relay stations. This offers greater planning flexibility and allows a better utilization of trucks, but also creates intricate interdependencies between trucks and drivers and requires the synchronization of their routes…

The Multi-period Multi-trip Container Drayage Problem with Release and Due Dates

2021

Abstract The Container Drayage Problem (CDP) aims at routing a fleet of trucks, based at a common terminal, to serve customers while minimizing the total travel distance. Each trip starts from and ends at the terminal, and handles a subset of customers. Each customer requires either that a container is picked up or delivered. We introduce a more realistic variant, i.e., the Multi-trip Multi-period CDP with Release and Due Dates (MM-CDP-RDD), in which the planning horizon is composed of several periods (days). On each day, each truck may perform more than one trip respecting the Release and Due Dates (RDD) associated with customer services, corresponding to the first and the last day on whic…

Mujer y FP: una fotografía en negativo

2018
