Contributions of ACE and mast cell chymase to endogenous angiotensin II generation and leucocyte recruitment in vivo
2011
Aims In vitro studies suggest that mast cell chymase (MCP) is more important than angiotensin-converting enzyme (ACE) for generating angiotensin II (Ang II) within the cardiovascular system. We investigated in vivo the relative contributions of ACE and MCP to leucocyte recruitment induced by endogenously generated Ang II. Methods and results Exposure of the murine cremasteric microcirculation of C57BL/6 mice to Ang I (100 nM for 4 h) induced leucocyte–endothelium interactions. Either losartan (an Ang II receptor-1 antagonist, AT1) or enalapril (an ACE inhibitor), but not chymostatin (a chymase inhibitor), inhibited Ang I-induced responses. Mast cell degranulation with compound 48/80 (CMP48/…

Epistaxis and systemic disease
2008
Abstract We report the case of a 77-year-old man who presented nasal obstruction sensation and epistaxis. Otorhinolaryngologic examination revealed occupation of the left nasal passage and the left maxillary sinus by an inflammatory tumour, the biopsy results of which were inconclusive. While diagnostic tests were being carried out, the patient presented a severe systemic condition consisting mainly of anemia, acute renal failure, and cavitated diffuse bilateral lung infiltrates. In the light of the results of anti-neutrophilic cytoplasmic antibodies and renal biopsy, Wegener's granulomatosis was diagnosed and treatment for the disease was instituted, with a favourable response. Finally, cl…
Mechanism of the cardiovascular activity of laudanosine: comparison with papaverine and other benzylisoquinolines.
1994
1. The activity of (+/-)-laudanosine, a benzyltetrahydroisoquinoline alkaloid, was investigated in pithed rats and rat isolated aorta. Its effects on [3H]-(+)-cis-diltiazem and [3H]-nitrendipine binding to rat cerebral cortical membranes, and on the different molecular forms of cyclic nucleotide phosphodiesterases (PDE) isolated from bovine aorta were investigated. 2. The dose-response curve to methoxamine (3-300 micrograms kg-1, i.v.) in normotensive pithed rats was shifted to the right by (+/-)-laudanosine, 3 and 6 mg kg-1. 3. (+/-)-Laudanosine inhibited in a concentration-dependent manner the contractile responses evoked by noradrenaline (NA 1 microM), depolarizing solution (KCl 80 mM) o…
Anatomical differences in responsiveness to vasoconstrictors in the mesenteric veins from normal and portal hypertensive rats
1996
The present study evaluates the effects of pre-hepatic portal hypertension, induced in rats by partial portal vein ligation, on the responsiveness of rostral (proximal) and caudal (distal) rings from the mesenteric vein. The anatomical origin of the sample influenced the response to vasoconstrictors in sham-operated animals, and this pattern of reactivity was specifically modified in portal-ligated rats. In veins from sham-operated rats, contraction induced by a submaximal concentration of KCl (60 mM) was greater in proximal than in distal rings. Vasopressin and 5-hydroxytryptamine contracted mainly distal rings, methoxamine showed a greater effect on proximal rings, and endothelin-1 and an…
Nebivolol inhibits superoxide formation by NADPH oxidase and endothelial dysfunction in angiotensin II-treated rats.
2006
Nebivolol is a β 1 -receptor antagonist with vasodilator and antioxidant properties. Because the vascular NADPH oxidase is an important superoxide source, we studied the effect of nebivolol on endothelial function and NADPH oxidase activity and expression in the well-characterized model of angiotensin II–induced hypertension. Angiotensin II infusion (1 mg/kg per day for 7 days) caused endothelial dysfunction in male Wistar rats and increased vascular superoxide as detected by lucigenin-derived chemiluminescence, as well as dihydroethidine staining. Vascular NADPH oxidase activity, as well as expression at the mRNA and protein level, were markedly upregulated, as well as NOS III uncoupled, …
Water-soluble carbon nanotubes through sugar azide functionalization
2011
In this work we report a covalent functionalization of pristine single-walled carbon nanotubes (SWCNTs) directly with three sugar azides, 2,3,4,6-tetra-O-acetyl-β-d-glucopyranosyl, 2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl or 2,3,4,6-tetra-O-acetyl-β-d-mannopyranosyl azide. Microwave-assisted functionalization was carried out for SWCNTs prepared with the HiPCO method. The as-prepared, new type of sugar-functionalized SWCNTs were analyzed by Raman and IR spectroscopy. Deacetylation of the functionalized tubes by sodium methoxide yielded nitrogen-linked, sugar-functionalized carbon nanotubes (CNTs) that formed stable dispersions in water. Reactivity of the sugar azides towards SWCNTs was es…

Synthesis of a Hydrothermally Stable, Periodic Mesoporous Material Containing Magnetite Nanoparticles, and the Preparation of Oriented Films
2006
Magnetite nanoparticles modified covalently with triethoxysilane having a quaternary dicetyl ammonium ion are used together with tetraethylorthosilicate as building blocks to prepare a mesoporous material. Cetyltrimethylammonium bromide is used as a structure-directing agent under conditions typically used for mesoporous MCM-41 silicas. The resulting mesoporous material (MAG-MCM-41), containing up to 15 wt % of magnetite is characterized by transmission electron microscopy (TEM), isothermal gas adsorption, and X-ray diffraction. In contrast to siliceous MCM-41, mesoporous MAG-MCM-41 exhibits a remarkable hydrothermal stability. The magnetic properties of MAG-MCM-41 are characterized by DC a…
