Mixed alkyl-methyl- and aryl-methylorganoaluminum chlorides 6 were formed by reaction of methylaluminum dichloride with organolithium or Grignard compounds and used for chemoselective 1,4 addition of higher alkyl, aryl, alkenyl and alkinyl groups to alkylidine malonic esters 1 and 2. As an alternative, mixed trimethylorganoaluminates 7 can also be applied for these Michael addition reactions. For conjugate addition of alkenyl groups to alkylidene malonates 1 and 2, alkenyl diisopropylalanes 10 obtained from alkynes and diisopropylaluminum hydride proved the most efficient reagents. Both enantiomers of sabina ketone were efficiently prepared via an easy synthesis of 1-diazo-5-methylene-6-methylheptan-2-one, starting from succinic anhydride, followed by its highly enantioselective cyclization catalyzed by chiral dirhodium(II) compounds having ortho-metalated phosphines as ligands.

Abstract A series of carboxylic acid derivatives 1–6 containing a second interacting function have been converted into the corresponding O -protonated species or acylium ions upon treatment with 96% sulfuric acid or oleum, respectively, at room temperature. The resulting stable cations have been spectroscopically characterized and submitted to irradiation. Cis-trans photoisomerization was observed in the ionic speciesderived from trans -crotonic, fumaric and maleic acids, and maleic anhydride. Hydride transfer is widespread in nature and has an essential role in applied research. However, the mechanisms of how this transformation occurs in living organisms remain a matter of vigorous debate. Here, we examined dihydrofolate reductase (DHFR), an enzyme that catalyzes hydride from C4′ of NADPH to C6 of 7,8-dihydrofolate (H2F). Despite many investigations of the mechanism of this reaction, the contribution of polarization of the π-bond of H2F in driving hydride transfer remains unclear. Abstract In the search for a new method of synthesis of hybrid peptides with aminopyrazole carboxylic acid, we tried to force selective acylation at the aromatic amino group instead of at the ring nitrogen atom with fairly gentle acylating agents. The acylating agents used were acid anhydrides: acetic anhydride, tert-butyl pyrocarbonate, and 2-(2-methoxyethoxy)ethoxyacetic acid/dicyclohexylcarbodiimide. We succceded in acylation at this amino group with almost none at the ring nitrogen atom. Sometimes, however, acylation in small quantities at the ring nitrogen atom was observed as a by-product. α-Hydro-ω-hydroxypoly(5-chloro-2-hydroxy-1,3-phenylenemethylene)s (1a, b), the oligomeric condensation products from 4-chloro-2,6-bis(hydroxymethyl)phenol and 4-chlorophenol with hydrochloric acid in 1,4-dioxane, could be completely dehalogenated at room temperature and atmospheric pressure with hydrogene/Raney-Ni. The resulting oligomers 2a, b were esterified with acetic anhydride, and the esters 3a, b were subjected to a oligomer analogous Fries rearrangement to form the linear α-hydro-ω-hydroxypoly(5-acetyl-2-hydroxy-1,3-phenylenemethylene)s 4a and 4b. The characterization of the products was mainly done by spectroscopic methods.

A new synthesis of pyrrolo[3,2-b] pyridine starting with pyrrole ring is described. The procedure allows the synthesis of 4-azaindoles bearing a sensitive group at C-7. The nitration of 4b with nitric acid and acetic anhydride at −15° gave 5. The hydrogenation of 5 led to simultaneous reduction of N-hydroxy and nitro groups and to hydrogenolysis of the isoxazole nucleus, affording an appropriate chain of atoms to building up the pyrrolo[3,2-b] pyridine ring.
Preparation of N,O-Aminals as Synthetic Equivalents of H2C=NAr and (H2C=NHAr)+ Ions: Neutral and Acid-promoted Transformations

A general method for the synthesis of N,O-aminals derived from primary aromatic amines is described. The reactivity of these compounds under neutral and acidic conditions has been studied and the title compounds can be envisaged as general purpose H2C=NAr or (H2C=NHAr)+ equivalents. N,O-Aminals have been converted into perhydrotriazines by moderate heating and into bis(4-aminoaryl) methane derivatives or N-benzylarylamines, respectively when heated in acidic media with pH control. Reduction of N, O-acetals with sodium cyanoborohydride has revealed that the C-O bond is broken exclusively in acidic media.

For the synthesis of symmetrical and unsymmetrical ethylidenediphenols the corresponding hydroxyphenylethanols (1a–c) were prepared by hydrogenation of hydroxyphenyl methyl ketones with hydrogen/Raney nickle or with lithium aluminium hydride. Condensations of the hydroxyphenylethanols with different phenols under the usual acidic conditions gave products formally resulting form transalkylations. Only the reaction of 1-(3-bromo-2-hydroxy-5-methylphenyl)ethanol (1c) with p-cresol gave the expected product 2d with 82% yield. Propanephosphonic Acid Anhydride–Mediated Cyclodehydration of Maleic Acid Monoamides

Abstract Propanephosphonic acid anhydride (T3P) has been proposed as a novel reagent for the preparation of maleic acid isoimides from the corresponding monoamides. A series of substituted aromatic and aliphatic isoimides have been prepared in good yields. The main advantage of this synthetic method is the use of environmentally benign, cost-efficient reagents and solvents, which are also safer to handle than the ones employed previously.
