A comparative study of carboxy myoglobin in saccharide-water systems by molecular dynamics simulation.

Results from room-temperature molecular dynamics simulation on a system containing carboxy-myoglobin, water, and maltose molecules are reported. Protein atomic fluctuations, protein−solvent and solvent−solvent hydrogen bonding have been analyzed and compared to the ones in trehalose−water and sucrose−water systems (Proteins 2005, 59, 291−302). Results help in rationalizing, at a molecular level, the effects of homologues disaccharides on protein structure/dynamics experimentally observed. Furthermore, the effectiveness of disaccharides in bioprotection in terms of peculiar protein−matrix coupling is also discussed.

Abstract The reversible redox couples (Fe(III)/Fe(II); Ce(IV)/Ce(III); Fe(CN) 6 3– /Fe(CN) 6 4− ; I 2 /I − ; Br 2 /Br − and VO 3 − /VO 2+ ) were studied as indicating redox systems for biamperometric determination of paracetamol in a flow injection assembly. Considering the selectivity of assayed systems against excipients and antioxidants that typically are presented in pharmaceutical formulations (e.g. sucrose, glucose, ascorbic acid and hydrogen sulphite) the systems Fe(III)/Fe(II) and VO 3 − /VO 2+ were selected for the determination of the drug. A novel and simple method for the purification of extracellular levansucrase from Zymomonas mobilis.

A new and simple method for the purification of extracellular levansucrase from Zymomonas mobilis from highly viscous fermentation broth was developed. After incubation of the fermentation broth with a fructose-polymer cleaving enzyme preparation (Fructozyme, Novozymes, DK) for 48 h, levansucrase precipitated as aggregates and was redissolved in a 3 M urea solution. By ongoing size-exclusion chromatography on Sephacryl S-300 the final levansucrase preparation was purified 100-fold and exhibited a specific activity of 25-35 U/mg(protein). Activities of NAD(+)-dependent sorbitol dehydrogenase (SDH), sorbitol oxidase (SOX), sucrose synthase (SS), acid invertase (AI), and neutral invertase (NI) in 'Encore' peach (Prunus persica L.) fruits and developing shoot tips were assayed during the growing season to determine whether carbohydrate metabolizing enzymes could serve as indicators of sink strength. In fruit flesh, SS activity was detected during Stage I of growth, when cells were actively dividing, and SDH activity was detected during Stage III, when cells were actively enlarging. In peach (Prunus persica [L.] Batsch.), sorbitol and sucrose are the two main forms of photosynthetic and translocated carbon and may have different functions depending on the organ of utilization and its developmental stage. The role and interaction of sorbitol and sucrose metabolism was studied in mature leaves (source) and shoot tips (sinks) of'Nemaguard' peach under drought stress. Plants were irrigated daily at rates of 100, 67, and 33% of evapotranspiration (ET). The relative elongation rate (RER) of growing shoots was measured daily. Terminal portions of `Flordaguard' peach roots [Prunus persica (L.) Batsch] were divided into six segments and the activities of NAD+-dependent sorbitol dehydrogenase (SDH), sorbitol oxidase (SOX), sucrose synthase (SS), soluble acid invertase (AI), and soluble neutral invertase (NI) were measured in each segment 10, 15, and 20 days after seed germination. The regioselective condensation of resorcarene tetratosylates 3/4 with formaldehyde and various primary amines readily gives bis-benzoxazines 5/6 in 65−86% yield. The chiral, C2-symmetrical structure of these compounds has been proved by NMR-spectroscopy and single-crystal X-ray analysis. Bis-benzoxazines 5f and 5i assume in the crystalline state a boat conformation in which the two resorcinol rings bearing the oxazine fragments are nearly parallel. The tosylated resorcinol units are horizontally arranged in a propeller-like fashion, and two intramolecular hydrogen bonds are formed between the phenolic hydroxy groups and the oxygens of the neighboring sulfonyl fragments. Abstract A water-soluble iron(III) complex [Fe(H2O)3(L)]·5H2O (1) was prepared by reaction of iron(III) chloride with 5-chloro-3-(2-(4,4-dimethyl-2,6-dioxocyclohexylidene)hydrazinyl)-2-hydroxy-benzenesulfonic acid (H3L). The complex was characterized by IR, 1H NMR and ESI-MS spectroscopies, elemental and X-ray crystal structural analyses. The coordination environment of the central iron(III) is a distorted octahedron, three sites being occupied by L3− ligand, which chelates in O,N,O fashion, while three other sites are filled with the water molecules.
