RT-PCR combined with immunoblotting showed the expression of group-I (mGlu1 and 5) and group-II (mGlu2 and 3) metabotropic glutamate receptors in whole mouse thymus, isolated thymocytes and TC-1S thymic stromal cell line. Tetanus toxin (TeTX) is a dichain protein that blocks neuroexocytosis, an action attributed previously to Zn(2+)-dependent proteolysis of synaptobrevin (Sbr) by its light chain (LC). Herein, its cleavage of Sbr in rat cerebrocortical synaptosomes was shown to be minimized by captopril, an inhibitor of certain metalloendoproteases, whereas this agent only marginally antagonized the inhibition of noradrenaline release, implicating a second action of the toxin. Monoclonal antibodies (mAb) against the native form of rat kidney gamma-glutamyl transpeptidase (GGT) were isolated by screening hybridomas with rat kidney brush-border membrane vesicles. They were directed against protein rather than sugar epitopes in that each recognized all GGT isoforms. All of them inhibited partially the enzyme activity of GGT. They were specific in that they inhibited the rat enzyme, but not the mouse or human enzyme. Many biomolecules are esterified with carbonic acid, phosphoric acid or sulphuric acid. The enzymes responsible for the hydrolytic cleavage of the esters are the carboxylester hydrolases, phosphoesterases and sulphatases. In addition to the phosphomonoesterases, we will also consider the phosphodiesterases and nucleases that are specific for cAMP and cGMP, as well as the ATPases that cleave phosphoric acid anhydride bonds. The ion-transporting ATPases will be discussed together with comparative biochemical data on ion channels. The chapter ends with a discussion of carboanhydrases.

Signal transduction in response to interleukin-6 (IL-6) results from homodimerization of gp130. This dimerization occurs after binding of IL-6 to its surface receptor (IL-6R) and can also be triggered by the complex of soluble IL-6R and IL-6. We fused IL-6 to the constant region of a human IgG1 heavy chain (Fc). IL-6Fc was expressed in COS-7 cells and purified via Protein A Sepharose. 2,3,6,7,10,11-Hexahydroxytriphenylene of good quality and purity can be obtained via anodic treatment of catechol ketals and subsequent acidic hydrolysis. The electrolysis is conducted in propylene carbonate circumventing toxic and expensive acetonitrile. The protocol is simple to perform and superior to other chemical or electrochemical methods. The key of the method is based on the low solubility of the anodically trimerized product. The shift of potentials is supported by cyclic voltammetry studies.
