Accumulation of senescent cells may drive age-associated alterations and pathologies. Senolytics are promising therapeutics that can preferentially eliminate senescent cells. Here, we performed a high-throughput automatized screening (HTS) of the commercial LOPAC®Pfizer library on aphidicolin-induced senescent human fibroblasts, to identify novel senolytics. The purpose of this study was to show that velocity-specific training may be implicated in modifications in the level of coactivation of agonist and antagonist muscles. Healthy males (n = 20) were randomly placed in to two groups: one group trained using concentric contractions (n = 12), the other was an untrained control group (n = 8). The training group underwent unilateral resistance training at a level of 35 (5)% of a one-repetition maximal contraction of the elbow flexors, executed at maximal angular velocity. Analgēzijas Nocicepcijas Indekss (ANI) ir jauna metode, kas ir paredzēta akūtu sāpju diagnostikai, gadījumos, kad pacients nav pie samaņas. Mērķis: Noteikt ANI efektivitāti akūtu sāpju detekcijā vispārējās anestēzijas (VA) iespaidā esošiem pacientiem. Hipotēze: ANI ir noderīgs palīgrīks anesteziologiem savlaicīgai intraoperatīvai akūtu sāpju detekcijai un personalizētai pacienta atsāpināšanas terapijai. I n their Focus article, Chen, Arendt-Nielsen, and Plaghki [8] provide a comprehensive review of the . literature o~ human evoked potentials that ~re elicited by noxious heat, usually generated by Infrared laser stimulators. From this review, we identified four issues that may deserve a comment: (1) Standardize the experiment or its scientific report? (2) Nociception and pain. (3) What do laser-evoked potentials (LEPs) really measure? (4) Rational clinical use. For the sake of brevity, we do not attempt to quote all relevant literature; a complete literature survey can be found in a recent review paper by Bromm and Lorenz [4].

Vanilloid receptors (VRs) play a fundamental role in the transduction of peripheral tissue injury and/or inflammation responses. Molecules that antagonize VR channel activity may act as selective and potent analgesics. We report that synthetic arginine-rich hexapeptides block heterologously expressed VR-1 channels with submicromolar efficacy in a weak voltage-dependent manner, consistent with a binding site located near/at the entryway of the aqueous pore. Dynorphins, natural arginine-rich peptides, also blocked VR-1 activity with micromolar affinity. Many neurodegenerative disorders present with sensory loss. In the group of hereditary sensory and autonomic neuropathies loss of nociception is one of the disease hallmarks. To determine underlying factors of sensory neurodegeneration we performed whole-exome sequencing in affected individuals with the disorder. In a family with sensory neuropathy with loss of pain perception and destruction of the pedal skeleton we report a missense mutation in a highly conserved amino acid residue of atlastin GTPase 3 (ATL3), an endoplasmic reticulum-shaping GTPase. Most patients with chronic lower back pain (CLBP) exhibit degenerative disc disease. Disc specimens obtained during initial therapeutic discectomies are often infected/colonized with Propionibacterium acnes, a Gram-positive commensal of the human skin. Although pain associated with infection is typically ascribed to the body's inflammatory response, the Gram-positive bacterium Staphylococcus aureus was recently observed to directly activate nociceptors by secreting pore-forming α-hemolysins that disrupt neuronal cell membranes.
