Determination of a conformational change in filamin A with Förster resonance energy transfer

2011

Filamins are large rod-like proteins that cross-link actin filaments into three-dimensional networks. They also bind to a plethora of proteins with distinct functions showing that they have a versatile role in cells. Functional filamins are dimers consisting of an N-terminal actin binding domain followed by 24 immunoglobulin-like domains. The most C-terminal domains mediate the dimerization. Two hinge regions are located between the domains 15 and 16 and 23 and 24, respectively, and produce structural flexibility that is essential for the protein function. The domains 18-19 and 20-21 are folded in a pairwise manner in which the first β strands of the even numbered domains are folded along w…

Innate immunity but not NLRP3 inflammasome activation correlates with severity of stable COPD.

2014

Background In models of COPD, environmental stressors induce innate immune responses, inflammasome activation and inflammation. However, the interaction between these responses and their role in driving pulmonary inflammation in stable COPD is unknown. Objectives To investigate the activation of innate immunity and inflammasome pathways in the bronchial mucosa and bronchoalveolar lavage (BAL) of patients with stable COPD of different severity and control healthy smokers and non-smokers. Methods Innate immune mediators (interleukin (IL)-6, IL-7, IL-10, IL-27, IL-37, thymic stromal lymphopoietin (TSLP), interferon γ and their receptors, STAT1 and pSTAT1) and inflammasome components (NLRP3, NA…

Discovery of varlaxins, new aeruginosin-type inhibitors of human trypsins

2022

Low-molecular weight natural products display vast structural diversity and have played a key role in the development of novel therapeutics. Here we report the discovery of novel members of the aeruginosin family of natural products, which we named varlaxins. The chemical structures of varlaxins 1046A and 1022A were determined using a combination of mass spectrometry, analysis of one- and two-dimensional NMR spectra, and HPLC analysis of Marfey's derivatives. These analyses revealed that varlaxins 1046A and 1022A are composed of the following moieties: 2-O-methylglyceric acid 3-O-sulfate, isoleucine, 2-carboxy-6-hydroxyoctahydroindole (Choi), and a terminal arginine derivative. Varlaxins 10…

Coscinolactams A and B: new nitrogen-containing sesterterpenoids from the marine sponge Coscinoderma mathewsi exerting anti-inflammatory properties

2009

Abstract Investigation of the marine sponge Coscinoderma mathewsi led to the isolation of two novel nitrogen-containing cheilanthane sesterterpenoids, coscinolactams A and B, together with known suvanine. The structures were elucidated by extensive spectroscopic measurements including NOE experiments to deduce the stereochemistry. The natural compounds, as well as a semisynthetic derivative, showed moderate anti-inflammatory activity measured as their capability to inhibit PGE2 and NO production. The suvanine aldehyde derivative 4 inhibited inducible nitric oxide protein expression with an IC50 value of 7.3 μM.

Global phylogeography and geographical variation in warning coloration of the wood tiger moth (Parasemia plantaginis)

2015

Aim To investigate the phylogeography of the aposematic wood tiger moth (Parasemia plantaginis) across its Holarctic distribution and to explore how its genetic structure relates to geographical differences in hindwing warning coloration of males and females. Males have polymorphic hindwing coloration, while female hindwing coloration varies continuously, but no geographical analyses of coloration or genetic structure exist. Location The Holarctic. Methods We sequenced a fragment of the mitochondrial cytochrome c oxidase subunit I gene (COI) from 587 specimens. We also examined more current population structure by genotyping 569 specimens at 10 nuclear microsatellite loci. Species distribut…

Lifting the Vendace, Coregonus albula, on the Life Table : Survival, Growth and Reproduction in Different Life-Stages during Very High and Low Abunda…

2021

We analyzed the variability in vendace population parameters in the context of a life table. Parameters related to growth, fecundity and survival were estimated for post-recruitment (age > 1 growing season) life stages of Lake Southern Konnevesi vendace in both very high and low population abundance regimes. Pre-recruitment survival producing population stability was then determined. We found a very strong compensatory density dependence in growth, fecundity and survival: during low abundance, a lifetime reproductive output of a female was almost 20 times that of the abundant regime. To maintain the low abundance regime, pre-recruitment survival must counterbalance it by decreasing to a ver…

Progresīvā iedzīvotāju ienākuma nodokļa ieviešanas iespējas Latvijā

2015

Impaired glucose regulation, depressive symptoms, and health-related quality of life

2020

IntroductionThis study aims to investigate whether the associations between impaired glucose regulation and health-related quality of life are modified by severity or type of depressive symptoms.Research design and methodsFor this cross-sectional study, we included 1939 individuals (mean age 61.5 years) from the Helsinki Birth Cohort Study. Between 2001 and 2004, a standard 2-hour 75 g oral glucose tolerance test was applied to define normoglycemia, pre-diabetes, and newly diagnosed diabetes. Information on previously diagnosed diabetes was collected from national registers and questionnaires. Pre-diabetes was defined as having either impaired fasting glucose or impaired glucose tolerance. …

Take-off aerodynamics in ski jumping

2001

The effect of aerodynamic forces on the force-time characteristics of the simulated ski jumping take-off was examined in a wind tunnel. Vertical and horizontal ground reaction forces were recorded with a force plate installed under the wind tunnel floor. The jumpers performed take-offs in non-wind conditions and in various wind conditions (21-33 m s(-1)). EMGs of the important take-off muscles were recorded from one jumper. The dramatic decrease in take-off time found in all jumpers can be considered as the result of the influence of aerodynamic lift. The loss in impulse due to the shorter force production time with the same take-off force is compensated with the increase in lift force, res…

Validation of portable 2D force binding systems for cross-country skiing

2013

The aim of the present study was to design, construct and scientifically validate a two-dimensional force measurement binding system for cross-country skiing. The system consists of two force measurement bindings. One binding was designed for analysing classic skiing (vertical and anterior–posterior [along the ski] force components) and the other one for skate (freestyle) skiing (vertical and medio-lateral [transverse to the ski] force components). Validation was accomplished using a three-step process: (1) accuracy tests for the sensors in two temperatures, (2) sport-specific imitation jump test on standard force plates in a laboratory and (3) comparing the system against force measurement…
