Intraflagellar transport protein 172 is essential for primary cilia formation and plays a vital role in patterning the mammalian brain

AbstractIFT172, also known as Selective Lim-domain Binding protein (SLB), is a component of the intraflagellar transport (IFT) complex. In order to evaluate the biological role of the Ift172 gene, we generated a loss-of-function mutation in the mouse. The resulting Slb mutant embryos die between E12.5 and 13.0, and exhibit severe cranio-facial malformations, failure to close the cranial neural tube, holoprosencephaly, heart edema and extensive hemorrhages. Cilia outgrowth in cells of the neuroepithelium is initiated but the axonemes are severely truncated and do not contain visible microtubules. Electron microscopy of a double helical tubular filament in keyhole limpet (Megathura crenulata) hemolymph.

A approximately 25 nm hollow double helical filament has been detected ultrastructurally in the cell-free supernatant from hemolymph of the keyhole limpet Megathura crenulata (Gastropoda: Prosobranchia: Fissurellidae). Subsequently, much higher concentrations of this material were found in the cell pellet from hemolymph. Both negative staining and thin sectioning have been performed in an attempt to obtain a preliminary structural characterization of this new filament. It is proposed that the filaments are released or secreted from blood hemocytes in response to bleeding, but it has not been possible to define absolutely an intracellular organelle containing this material. Johdanto. Uimari on uidessaan käytännössä lähes koko vartalollaan kosketuksissa veteen. Vesi on väliaine, joka vastustaa uimarin etenemistä ja josta voiman ja vastavoiman periaatteen mukaisesti uimaria eteenpäin vievien voimien tukipinnat tulisi löytää. Veden viskoelastinen luonne luo omat erityispiirteensä eteenpäin suuntautuvien ja vastustavien voimien välille. Bakalaura darba tēma ir “Pacientu ar ventrikulāro stomu aprūpe intensīvās terapijas un reanimācijas nodaļā”. Smaga galvas smadzeņu trauma visbiežāk ir 15-35 gadu veciem cilvēkiem, un tā ir 1/3 ar traumatismu saistītu nāves gadījumu. Pieejamā medicīnas statistika liecina, ka gadā no katriem 100.000 iedzīvotājiem 150-200 tiek ievietoti stacionārā intensīvās terapijas nodaļā, galvas traumas dēļ. No hospitalizētiem pacientiem 8-11% ir galvas trauma ir ar 30-60% letalitāti. Glutathione (L-γ-glutamyl-L-cysteinyl-glycine; GSH) in cancer cells is particularly relevant in the regulation of carcinogenic mechanisms; sensitivity against cytotoxic drugs, ionizing radiations, and some cytokines; DNA synthesis; and cell proliferation and death. The intracellular thiol redox state (controlled by GSH) is one of the endogenous effectors involved in regulating the mitochondrial permeability transition pore complex and, in consequence, thiol oxidation can be a causal factor in the mitochondrion-based mechanism that leads to cell death. This study presents the effects of citric acid and extracellular pH (pHe) on the intracellular pH (pHi) of wild-type and citrate negative variants (cit−) Leuconostoc mesenteroides subsp. mesenteroides (Ln. mesenteroides M) and Lactococcus lactis subsp. lactis bv. diacetylactis (L. lactis LD). A recent method using a pH-sensitive fluorescent indicator carboxyfluorescein succinimidyl ester (cFSE) was adapted to measure the pHi of these two lactic acid bacteria in resting cells. DNA synthesis was studied autoradiographically in unfertilized ascidian eggs (Ascidia malaca and Ciona intestinalis) that had been artificially activated by a K+-free external medium or a Ca-ionophore. Naked eggs of A. malaca were incubated in K+-free seawater that contained [3H]-thymidine for 30 min, and naked eggs of C. intestinalis were incubated in seawater supplemented with a Ca-ionophore and [3H]-thymidine for 15 min.
