Graft-versus-host disease (GvHD) is a key contributor to the morbidity and mortality after allogeneic hematopoetic stem cell transplantation (HSCT). Regulatory Foxp3(+) CD4(+) T cells (Treg) suppress conventional T cell activation and can control GvHD. In our previous work, we demonstrate that a basic mechanism of Treg mediated suppression occurs by the transfer of cyclic adenosine monophosphate (cAMP) to responder cells. Whether this mechanism is relevant for Treg mediated suppression of GvHD is currently unknown. To address this question, bone marrow and T cells from C57BL/6 mice were transferred into lethally irradiated BALB/c recipients, and the course of GvHD and survival were monitore…
Novel antiproliferative chimeric compounds with marked histone deacetylase inhibitory activity.
2014
Given our interest in finding potential antitumor agents and in view of the multifactorial mechanistic nature of cancer, in the present work, taking advantage of the multifunctional ligands approach, new chimeric molecules were designed and synthesized by combining in single chemical entities structural features of SAHA, targeting histone deacetylases (HDACs), with substituted stilbene or terphenyl derivatives previously obtained by us and endowed with antiproliferative and pro-apoptotic activity. The new chimeric derivatives were characterized with respect to their cytotoxic activity and their effects on cell cycle progression on different tumor cell lines, as well as their HDACs inhibitio…
Perceived Reciprocity and Well-Being at Work in Non-Professional Employees: Fairness or Self-Interest?
2012
This article assesses the links between non-professional employees' perceptions of reciprocity in their relationships with their supervisors and the positive and negative sides of employees' well-being at work: burnout and engagement. Two hypotheses were explored. First, the fairness hypothesis assumes a curvilinear relationship where balanced reciprocity (when the person perceives that there is equilibrium between his/her efforts and the benefits he/she receives) presents the highest level of well-being. Second, the self-interest hypothesis proposes a linear pattern where over-benefitted situations for employees (when the person perceives that he/she is receiving more than he/she deserves)…
Multiplicity of Radial Ground States for the Scalar Curvature Equation Without Reciprocal Symmetry
2022
AbstractWe study existence and multiplicity of positive ground states for the scalar curvature equation $$\begin{aligned} \varDelta u+ K(|x|)\, u^{\frac{n+2}{n-2}}=0, \quad x\in {{\mathbb {R}}}^n\,, \quad n>2, \end{aligned}$$ Δ u + K ( | x | ) u n + 2 n - 2 = 0 , x ∈ R n , n > 2 , when the function $$K:{{\mathbb {R}}}^+\rightarrow {{\mathbb {R}}}^+$$ K : R + → R + is bounded above and below by two positive constants, i.e. $$0<\underline{K} \le K(r) \le \overline{K}$$ 0 < K ̲ ≤ K ( r ) ≤ K ¯ for every $$r > 0$$ r > 0 , it is decreasing in $$(0,{{{\mathcal {R}}}})$$ ( 0 , R ) and increasing in $$({{{\mathcal {R}}}},+\infty )$$ ( R , + ∞ ) for a certain $${{{\mathcal {R}}}}&g…

The crystal structure of a 250-kDa heterotetrameric particle explains inhibition of sheddase meprin β by endogenous fetuin-B
2021
Meprin β (Mβ) is a multidomain type-I membrane metallopeptidase that sheds membrane-anchored substrates, releasing their soluble forms. Fetuin-B (FB) is its only known endogenous protein inhibitor. Herein, we analyzed the interaction between the ectodomain of Mβ (MβΔC) and FB, which stabilizes the enzyme and inhibits it with subnanomolar affinity. The MβΔC:FB crystal structure reveals a ∼250-kDa, ∼160-Å polyglycosylated heterotetrameric particle with a remarkable glycan structure. Two FB moieties insert like wedges through a “CPDCP trunk” and two hairpins into the respective peptidase catalytic domains, blocking the catalytic zinc ions through an “aspartate switch” mechanism. Uniquely, the …
Under my wing: lesser kestrels and jackdaws derive reciprocal benefits in mixed-species colonies
2011
In mixed-species assemblages, antipredator benefits for a timid species nesting close to a more pugnacious one are often reported. Advantages for the protected species are usually manifested in terms of higher reproductive success than conspecifics nesting remote to the protector species. Whether the protector species also accrues any benefit remains untested, and the species-specific behavioral traits underlying enhanced reproductive output in mixed-species associations remain poorly documented. We studied associations between lesser kestrels (Falco naumanni) and jackdaws (Corvus monedula) nesting in rural buildings in the Gela Plain (Italy). We tested for interspecific interactions of jac…
Myślenie dialogiczne. W polu gry o prawdę etyczną
2019
Najpierw przepro…
The NMDA receptor complex: a promising target for novel antiepileptic strategies.
2001
Antiepileptic drugs (AEDs) cover a broad spectrum of pathological conditions ranging from seizures following congenital or acquired brain disorders to behavioural and psychiatric disorders and recently neuropathic pain. The need for novel antiepileptics raises from the expanding field of indications as well as from the fact, that special seizure types are refractory to common AEDs. In addition, many of the conventional antiepileptic drugs exhibit an unfavourable side-effect profile. Since there is growing evidence, that NMDA receptor activation might play a crucial role in epilepsy, NMDA receptor antagonists have become compounds of interest in preventing and treating seizures. This review …
