A novel putative insect chitinase with multiple catalytic domains: hormonal regulation during metamorphosis

0264-6021 (Print) Journal Article Research Support, Non-U.S. Gov't; We have used differential display to identify genes that are regulated by juvenile hormone in the epidermis of the beetle Tenebrio molitor. One of the genes encodes T. molitor chitinase 5 (TmChit5), a chitinase possessing an unusual structure. Sequence analysis of TmChit5 identified five 'chitinase units' of approx. 480 amino acids with similarity to chitinase family 18. These units are separated by less conserved regions containing putative PEST (rich in proline, glutamic acid, serine and threonine) sequences, putative chitin-binding domains and mucin domains. Fluorescent 4-ethylthio-1,8-naphthalimides containing rhodium(I) N-heterocyclic carbene (NHC) and ruthenium (II) NHC fragments were synthesised and evaluated for their antiproliferative effects, cellular uptake and DNA-binding activity. Both types of organometallics triggered ligand dependent efficient cytotoxic effects against tumor cells with the rhodium(I) NHC derivatives causing stronger effects than the ruthenium (II) NHC analogues. Antiproliferative effects could also be observed against several pathogenic Gram-positive bacterial strains, whereas the growth of Gram-negative bacteria was not substantially affected. Carbon nanotubes embedded in a polyimide foil for proton acceleration with a sub-ns laser

A series of thin films made of aligned carbon nanotubes (CNTs) embedded in a polyimide substrate was designed, fabricated and used for the first time to accelerate protons and C ions by interaction with a sub-nanosecond, high power laser beam (600 J energy and 300 ps pulse width) with peak intensity of about 3 × 1016 W/cm2 on target. Each target was 5 μm thick, and the composite material contained CNTs aligned in different directions in the substrate. UV photostability of metal phthalocyanines in organic solvents

Kinetic studies of photochemical reactions induced by UV radiation in solutions of metal phthalocyanines have been carried out to determine the factors which might have influenced the stability of photosensitized phthalocyanines. Complexes of the molecular type Mpc, M'(2)pc, and Lnpc(2) (where M = Li, Mg, Fe, Co, Zn, Pb; M'= Tl; Ln = rare earth; pc = phthalocyanine ligand, C(32)H(16)N(8)(2-)) were investigated in DMF, DMSO, and pyridine. Progressive decay of the phthalocyanine macrocycle due to absorption of UV light was observed. In situ synthesis of lipopeptides as versatile receptors for the specific binding of nanoparticles and liposomes to solid-supported membranes.

A detailed study of the in situ coupling of small peptides such as CGGH6 (H6) and CGWK8 (K8) to maleimide functionalized phospholipid bilayers is presented. Individually addressable microstructured membranes are employed to unequivocally probe the conjugation. The in situ coupling of peptides via a terminal cysteine moiety to maleimide functionalized phospholipids is shown to be a convenient and versatile way to selectively fabricate peptide-modified phospholipid bilayers serving as specific receptor platforms for functionalized vesicles and nanoparticles. Effects of Escherichia coli hemolysin on human monocytes. Cytocidal action and stimulation of interleukin 1 release.

Abstract This study reports on the potent cytocidal and interleukin-1 releasing properties of Escherichia coli hemolysin (ECH) on human monocytes. Nanomolar concentrations of purified ECH (250-2,000 ng/ml) caused rapid and irreversible depletion of cellular ATP to levels below 20% of controls within 60 min. Subcytocidal doses (10-200 ng/ml) of ECH induced rapid release within 60-120 min of large amounts of interleukin 1 beta (IL-1 beta) from cultured monocytes. IL-1 beta release occurred in the presence of actinomycin D and cycloheximide, and was thus probably due to processing and export of intracellular IL-1 beta precursor. In vitro induction of cecropin genes — an immune response in a Drosophila blood cell line

The Drosophila melanogaster cell line mbn-2 was explored as a model system to study insect immune responses in vitro. This cell line is of blood cell origin, derived from larval hemocytes of the mutant lethal (2) malignant blood neoplasm (1(2)mbn). The mbn-2 cells respond to microbial substances by the activation of cecropin genes, coding for bactericidal peptides. The response is stronger than that previously described for SL2 cells, and four other tested Drosophila cell lines were totally unresponsive. Strong contact sensitizers are able to induce distinct signal transduction mechanisms in antigen-presenting cells by coupling to cell proteins. The predominant target structures of haptens are thought to be thiol and amino groups in cysteine and lysine residues. Anti-inflammatory activity of saikosaponins from Heteromorpha trifoliata.

Increased Level of Intracellular MHC Class II Molecules in Murine Langerhans Cells Following In Vivo and In Vitro Administration of Contact Allergens

Treatment of murine Langerhans cells (LC) with contact allergens results in increased internalization of cell membrane constituents and therefore in depressed cell-surface expression of major histocompatibility complex (MHC) class II molecules during the first hours after haptenization. In this presentation we show that this downregulation of cell-surface-expressed Ia-antigens is accompanied by an augmentation of the intracellular pool of MHC class II molecules.
