Thus far, the synthesis of C1q by liver cells has not been demonstrated. To investigate this possibility, viable hepatocytes were isolated from the liver of guinea pigs and primary cultures were established. The cells (10(6) cells/ml) were cultured under serum-free conditions for 8 days and the culture medium was changed every 24 h. The few contaminating Kupffer cells were lysed by preincubating the cell cultures with a monoclonal (22C4-8) antibody directed against a nonpolymorphic Ia determinant and preabsorbed rabbit serum. In most cells, cationic amino acids such as l-arginine, l-lysine, and l-ornithine are transported by cationic (CAT) and y(+)L (y(+)LAT) amino acid transporters. In human erythrocytes, the cysteine-modifying agent N-ethylmaleimide (NEM) has been shown to inhibit system y(+) (most likely CAT-1), but not system y(+)L (Devés, R., Angelo, S., and Chávez, P. (1993) J. Physiol. 468, 753-766). We thus wondered if sensitivity to NEM distinguishes generally all CAT and y(+)LAT isoforms. Abstract We studied how changing the anion affects the self-assembly of three diimidazolium salts bearing a perylene bisimide spacer. In particular we used [BF 4 − ] and [NTf 2 − ] based salts together with a mixed salt bearing both [I − ] and [BF 4 − ] anions. Thermal as well as concentration and temperature dependent spectroscopic investigations revealed that changing the anion impacts on solid state and solution aggregation, leading to articulate thermotropic behaviors and contrasting solvent effects, respectively. This secretion required protein synthesis, but was independent of RNA formation (Mormeneo & Sentandreu 1982). In addition accumulation of invertasespecific messenger RNA occurred in the absence of protein synthesis but was expressed only after synthesis of protein. Factor B of the alternative pathway of complement activation was shown to be synthesized and secreted by unstimulated mouse peritoneal macrophages. The activity of B in the culture supernatants from macrophage monolayers was detected by consumption of C3 in reaction mixtures containing supernatant and guinea pig factors C3, D and insoluble C3b. Using a monospecific antiserum, factor B in concentrated culture supernatants was shown by immunodiffusion and immunoelectrophoresis to be identical to factor B in mouse plasma and to form a characteristic complex with cobra venom factor in the presence of D. AbstractHuman papillomavirus virus-like particles (VLPs) have recently been used to deliver genes into mammalian cells in vitro and in vivo. Here, we investigated whether VLPs may serve as an efficient carrier of low molecular weight compounds (e.g. hormones, vitamins, peptides etc.) into cells. COS7 cells were incubated with recombinant HPV-16L1/L2 VLPs labelled with the fluorescence dye carboxyfluorescein diacetate succinimidyl ester. Using flow cytometry, we demonstrate that labelled VLPs can specifically bind to the cell surface followed by their complete internalisation. A new Ni(II) layered hybrid organic-inorganic compound of formula Ni{sub 2}[(NDI-BP)(H{sub 2}O){sub 2}].2H{sub 2}O has been prepared in very mild conditions from N,N'-bis(2-phosphonoethyl)napthalene-1,4:5,8-tetracarboximide (NDI-BP ligand) and NiCl{sub 2}. The X-ray powder structure characterization of the title compound suggests a pillared layered organic-inorganic hybrid structure. The distance between the organic and inorganic layers has been found to be 17.8 A. The inorganic layers consist of corner sharing [NiO{sub 5}(H{sub 2}O)] octahedra and they are pillared by the diphosphonate groups.
